Abstract
1. Рекомбинантная плазмида pAd-SN, кодирующая белки SOX2 и NANOGчеловека, являющаяся основой для получения вирулентных аденовирусов, предназначенных для получения индуцированных плюрипотентных стволовых клеток человекапредставляющая собой генетическую конструкцию, построенную путем гомологичной рекомбинации вектора pAdEasy-1, содержащего основную часть генома аденовируса и плазмиды pAdTrack-CMV, включающей фрагмент SOX2-P2A-NANOG под контролем конститутивного промотора CMV, встроенный в сайт рестрикцииI, размер встройки 1964 пар нуклеотидов, где кДНК генов SOX2 и NANOG, кодирующие белки SOX2 и NANOG человека соединены последовательностью, кодирующей P2A-пептид, и ген, кодирующий флуоресцентный белок EGFP под контролем промотора CMV.2. Рекомбинантная плазмида pAd-SN по п.1, где конструктивная основа вектораpAdEasy-1 обеспечивает получение аденовирусных частиц с одновременной доставкой генов и экспрессию белков SOX2, NANOG и EGFP.3. Рекомбинантная плазмида pAd-SN по п.1, гдефрагмент ДНК, кодирующий белки SOX2 и NANOG человека запускает репрограммированиеклеток для получения индуцированных плюрипотентных стволовых клеток человека, а фрагмент ДНК кодирующий флуоресцентный белок EGFP является маркером трансдукции клеток и последующей элиминации вирусной ДНК.1. The recombinant plasmid pAd-SN according to claim 1, wherein the DNA fragment encoding the human SOX2 and NANOG proteins triggers cell reprogramming to produce induced human pluripotent stem cells, and the DNA fragment encoding the EGFP fluorescent protein is a marker for cell transduction and subsequent elimination of viral DNA.
