Способ определения летального фактора сибирской язвы на основе иммунодетекции, сопряженной с полимеразной цепной реакцией, включающий адсорбцию белка летального фактора на микропланшетах или микрострипах при помощи специфического моноклонального антитела, связывание иммобилизованного белка летального фактора при помощи биотинилированного моноклонального антитела, узнающего другой эпитоп белка летального фактора, детекцию комплекса белка летального фактора с биотинилированным антителом посредством нековалентного коньюгата фрагментов ДНК с нейтравидином, служащего матрицей для проведения ПЦР-амплификации с флюоресцентной детекцией сигнала в режиме реального времени и регистрацию присутствия протективного антигена в исследуемых образцах по изменению уровня флюоресценции в соответствующих лунках планшета или пробирках стрипа против контрольных.A method for determining lethal factor of anthrax based on immunodetection coupled with a polymerase chain reaction, including adsorption of a lethal factor protein on microplates or microstrips using a specific monoclonal antibody, binding of an immobilized lethal factor protein using a biotinylated monoclonal antibody that recognizes another epitope of a lethal factor protein, a complex of a lethal factor protein with a biotinylated antibody via a non-covalent conjugate fra DNA fragments with neutravidin, which serves as a matrix for PCR amplification with fluorescence detection of the signal in real time and registration of the presence of protective antigen in the studied samples by changing the fluorescence level in the corresponding wells of the tablet or strip tubes against the control.
