Abstract
Способ оценки эффективности применения иммуномодулятора имунофана при экспериментальной холере, включающий заражение животных в изолированную петлю тонкого кишечника взрослых кроликов вирулентным штаммом холерного вибриона, отличающийся тем, что до заражения холерой животным весом 1,5-2,0 кг, вводят через день иммуномодулятор имунофан в дозе 0,2 мкг, разведенный в 0,3 мл новокаина в количестве 10 инъекций, затем животных выдерживают без пищи 24 ч, после этого, под эфирным наркозом извлекают петли тонкого кишечника, накладывают лигатуры, перевязывают одинаковые участки кишечника длинной 12 см с промежутками между ними 4-5 см, при этом в одну лигированную петлю вводят 1 мл 0,9%-ного раствора хлорида натрия для контроля, в другую - 10клеток 24-часовой культуры Vibrio cholerae 5879, после брюшные стенки зашивают и через 18 ч животных забивают для последующего изучения и оценки эффективности использования имунофана, которую проводят по наличию патоморфологических изменений в перевязанных петлях тонкого кишечника, выраженность которых оценивают в крестах, и наличию холерогенного эффекта, который определяют по коэффициенту растяжения петли, рассчитываемого по формуле:где К - коэффициент растяжения петли;Vж - объем жидкости;Lп - длинна петли,если К<1, то применение иммуномодулятора имунофана при холере является эффективным.A method of evaluating the effectiveness of the use of an immunomodulator of immunofan in experimental cholera, which involves infection of animals in an isolated loop of the small intestine of adult rabbits with a virulent strain of cholera vibrio, characterized in that before the infection with cholera of an animal weighing 1.5-2.0 kg, the immunomodulator is administered every other day in a dose 0.2 μg, diluted in 0.3 ml of novocaine in the amount of 10 injections, then the animals are kept without food for 24 hours, after which, under ether anesthesia, loops of the small intestine are removed, ligatures are applied, identical sections of the intestine are 12 cm long with 4-5 cm intervals between them, while 1 ml of a 0.9% sodium chloride solution is introduced into one ligated loop for control, and 10 cells of a 24-hour Vibrio cholerae 5879 culture are introduced into the other, after the abdominal walls are sutured and after 18 hours the animals are slaughtered for subsequent study and evaluation of the effectiveness of the use of immunofan, which is carried out by the presence of pathomorphological changes in the tied loops of the small intestine, the severity of which is evaluated in the crosses, and the presence of a cholerogenic effect, which limit by the loop stretching coefficient calculated by the formula: where K is the loop stretching coefficient; Vzh is the liquid volume; Lp is the loop length, if K <1, then the use of the immunofan modulator for cholera is effective.
