Abstract
Description
The invention relates to the field of experimental medicine, mainly to methods for studying the function of organs.
An analogue of the claimed method is a method for producing blood from the tail vein, in which the rat tail is placed in a chamber with negative pressure and the tail vein is crossed. This method does not allow to obtain blood from the central veins, which differs in composition from peripheral. It is not suitable for private obtaining blood, because it leads to the development of anemia in the animal, since there is no way to make up for blood loss.
- прокалывание отверстия стальной иглой, через которую вводят в вену канюлю;
- проверка проходимости канюли и фиксации ее к поясничной мышце;
The prototype method is based on cannulation of the posterior vena cava (caudal) vein of the rat and the removal of the catheter on the skin behind the ears of the animal. The prototype method includes the following basic techniques:
- piercing the hole with a steel needle through which a cannula is inserted into the vein;
- checking the patency of the cannula and fixing it to the lumbar muscle;
- removing the end of the catheter to the skin surface 2.5 cm behind the ears and fixing.
The main disadvantage of the prototype is that during catheterization of the posterior vena cava, the vessel wall due to the small thickness of the muscle membrane does not tightly cover the needle, and therefore blood comes from the puncture site, which often leads to the death of the animal due to blood loss. In addition, an open frame at the exit site of the catheter on the skin promotes the penetration of infection into the wound canal and the development of sepsis, as well as the ingestion of the catheter by animals, which leads to the termination of the experiment.
The purpose of the invention is to eliminate the noted drawbacks and increase the reliability of angiostomy by eliminating damage to the catheter by animals and preventing bleeding during puncture of the posterior vena cava, which allows you to repeatedly receive blood samples from the central veins and intravenously inject drugs in a chronic experiment, as well as effectively replenish blood loss unavoidable when taking blood samples for analysis.
This goal is achieved by catheterizing one of the branches of the posterior vena cava - the testicular in males, the ovarian - in females, the vein through which the catheter is passed to the posterior vena cava. After that, the vein is ligated distally, and above the site of insertion of the catheter, it is fixed to the wall of the vein, its free end, conically expanding and tightened by an elastic membrane, is brought out under the skin 1.5-2 cm below the biacular line and fixed without leading to the surface.
Due to the catheterization of not the vena cava itself, but one of its branches, the testicular (ovarian) vein, less trauma is achieved, since the lumen of the main vessel, the posterior vena cava, is not obstructed by the catheter (as in the prototype method), moreover, more high reliability of catheter attachment. Due to the presence of an elastic membrane tightening the free end of the catheter, as well as the fact that the catheter is not brought to the surface, but left under the skin, it becomes possible to take multiple blood samples, piercing the skin and membrane. This eliminates the possibility of gnawing the catheter by the animal, since the latter is located under the skin of the back.
П р и м е р. PRI me R. An outbred white male rat weighing 210 g was anesthetized by intraperitoneal administration of 0.9 ml of a 1% sodium pentobarbital solution (40 mg / kg). After reaching the third stage of anesthesia, the abdominal cavity was opened. The right testicular vein was isolated from retroperitoneal fiber and ligatures were performed under it. The latter were pulled up and a puncture of the vein was performed between them with sharpened tweezers. Tweezers jaws were diluted 1 mm and between them a 0.6 mm diameter catheter was inserted into the lumen of the vein. The catheter was guided to the posterior vena cava, after which it was fixed in the testicular vein by ligature. The distal end of the testicular vein was bandaged. A catheter of 1-2 mm in silk sutures was fixed to the muscles of the back. The transverse and oblique muscles of the abdomen at the border with the lumbar muscle were bluntly bred. A tunnel was bluntly formed between the skin and muscles, into which the end of the catheter was placed, made in the form of an expanding cone and tightened by an elastic membrane. The muscles were sutured with separate catgut sutures, while fixing the distal end of the catheter with one silk suture. The wound of the anterior abdominal wall was sutured tightly. The catheter was subcutaneously conducted to a point located 1.5-2 cm below the biauricular line.
(56) J Appl Physiol 57 (5), 1594-1596, 1984. During the 1st week, the animal was kept on a standard diet in conditions of free access to food and water. Every day in the morning, the catheter was washed with a 1% solution of heparin on polyglucin in order to prevent catheter thrombosis, as well as to accustom the animal to the experimental conditions as follows: under the skin of the animal, the expanded end of the catheter was palpated, fixed between the fingers and percutaneous cavity puncture was performed catheter; blood sampling was performed; filled with 1% heparin solution on polyglucin; the needle was removed, the rubber membrane contracted and prevented bleeding from the site of puncture of the catheter. At the end of this period, the rat was simulated with pathology and blood samples were taken for 48 hours. (56) J Appl Physiol 57 (5), 1594-1596, 1984.
