Изобретение относится к биотехнологии, сельскому хозяйству, в частности к генетическим исследованиям зерновых культур, и может быть использовано в селекции и семеноводстве для отбора форм кукурузы с улучшенным качеством зерна. The invention relates to biotechnology, agriculture, in particular to genetic research of grain crops, and can be used in selection and seed production for the selection of forms of corn with improved grain quality.
Селекционные программы по улучшению качества зерна кукурузы включают использование эндоспермовых мутаций, изменяющих сбалансированность белкового комплекса. Для биологической ценности зерна является важным также оптимальное накопление каротиноидов, содержание которых изменяется под действием эндоспермовых мутаций. Для решения этой важной для селекции проблемы необходимы способы экспресс-диагностики, определяющие специфичность накопления каротиноидов в зерне эндоспермовых мутантов кукурузы. For the biological value of grain, the optimal accumulation of carotenoids, the content of which changes under the influence of endosperm mutations, is also important. To solve this important problem for breeding, rapid diagnostic methods are needed that determine the specificity of the accumulation of carotenoids in the grain of corn endosperm mutants.
Известные способы оценки воздействия эндоспермовых мутаций на каротиноидный комплекс зерна кукурузы включают экстракцию пигментов органическим растворителем, хроматографическое разделение экстракта на основные каротиноидные фракции, их идентификацию и количественное определение. К основным недостаткам указанных способов следует отнести их длительность и трудоемкость, необходимость использования набора химических реагентов и хроматографических сорбентов. Согласно способа навеску размолотого зерна кукурузы экстрагируют водонасыщенным бутанолом при постоянном встряхивании в течение 25 мин. Экстракт каротиноидов отделяют фильтрованием и измеряют оптическую плотность окрашенного раствора на спектрофотометре при 435,8 нм. С использованием известного коэффициента экстинктции по величине оптической плотности рассчитывают содержание каротиноидов, которое используют для оценки селекционного материала. Отбор высококаротиноидных мутантных форм проводят по максимальному значению спектрального показателя среди анализируемых селекционных образцов. На основании установленного спектрального параметра возможен вариант неразрушающего позернового экспресс-отбора, что дает возможность сохранить высококаротиноидные зерновки перспективных форм для селекционно-генетических исследований. Процесс селекционной диагностики с применением маркерного признака идентифицирует генотипы с повышенным содержанием каротиноидов среди однотипных мутаций, определяет генотипическую вариабельность данного признака для эндоспермовых мутаций различной природы, а также их сочетаний, выявляет перспективные селекционные формы среди синтетиков и высоколизиновых гибридов кукурузы.
