SUMMARY OF THE INVENTION; Dry yeast is divided into two parts, one part is subjected to alkaline hydrolysis with a 1 M alkali solution at 100-120 ° C for 1 hour, and the other is subjected to acid hydrolysis with a 1 M HCI solution in the same modes. The resulting hydrolysates were mixed, adjusted to a neutral pH and the target product was isolated as an insoluble fraction. The drug exhibits immunostimulatory activity. 1 tab.
Description
The invention relates to the field of biotechnology, in particular to a technology for producing an immunostimulating drug.
The essence of the invention is that a biologically active preparation is obtained by hydrolysis of yeast (baking GOST 20085 or feed GOST 171-81). The yeast is divided into two parts, one part is destroyed by acid hydrolysis, and the other, equal, alkaline. After carrying out both hydrolyses, the hydrolysates are mixed. The pH is neutral, the precipitate is separated, which is washed three to four times with distilled water. The washed precipitate contains up to 90% of the sugars represented by glucan. The resulting precipitate was resuspended in saline to a concentration of 1 mg / cell and used as a biologically active substance.
Example. 50 grams of dry baker's yeast pour 200 ml of distilled water. The resulting suspension was divided into two equal parts, HCI was added to one part, adjusting the final acid concentration to 0.5 M. To the second part
NaOH is added, adjusting its concentration to 0.5 M. The hydrolysis of both parts is carried out for one hour at 10C in reflux tanks. After hydrolysis is complete, the hydrolysates are mixed, cooled to room temperature and adjusted to pH-7.0. The mixture of hydrolysates is centrifuged, the precipitate is separated, which is washed 3 times with distilled water, purifying it of soluble proteins and polysaccharides. The resulting preparation is suspended in physiological saline and sterilized and used to normalize the immune status of animals.
П р и м е р 2. PRI me R 2. Make two samples of fodder yeast 25 grams. One weighed yeast is resuspended in 100 ml of 1 M HCI, the other in 1 M NaOH. Both parts are hydrolyzed in reflux flasks at 120 ° C for one hour. After hydrolysis, the hydrolysates are combined, cooled, the pH is neutral, the precipitate is separated by settling, draining the supernatant, then the precipitate is washed with distilled water.

П р и м е р 3. Биологическую активность препаратов исследуют in vivo и in vitro. PRI me R 3. The biological activity of the drugs investigated in vivo and in vitro. By conventional methods, the number of antibody-forming spleen cells (Erne test) and the stimulation index, the number of macrophages and the phagocytic index, complement fixing activity of the drug and lysozyme activity of blood serum are determined. The ability of the preparation to protect mice from salmonella infection was studied. The mice were injected with the preparation obtained in examples 1 and 2, and after 48 hours they were infected with a virulent strain of Salmonella.tlphimyrium 415 at a dose of 10 LDso. The animals were counted for 10 days and the percentage of surviving mice was calculated.
