Description
"Medical ultrasound systems." Interface Focus 1.4 (2011): 477-489). (Wang, Lihong V., and Song Hu. "Photoacoustic tomography: in vivo imaging from organelles to organs." Science 335, no. 6075 (2012): 1458-1462).The utility model relates to equipment for radiation diagnostics, in particular for imaging biological tissue in vivo. Bioimaging of the studied biological tissue by the proposed device is carried out with the complementary and simultaneous use of three types of visualization - optical, optoacoustic and ultrasonic. Optical imaging consists in probing biological tissue with electromagnetic radiation of the visible and near-IR ranges (VV Tuchin, “Optical Biomedical Diagnostics.” News of Saratov University. New Series. Physics 5.1 (2005) series. Ultrasound imaging - in probing the biological tissue under investigation with acoustic pulses (Powers, Jeff, and Frederick Kremkau. "Medical ultrasound systems." Interface Focus 1.4 (2011): 477-489). Optoacoustic imaging consists in recording ultrasonic waves excited in the test medium upon absorption of pulsed laser radiation by optical inhomogeneities (Wang, Lihong V., and Song Hu. "Photoacoustic tomography: in vivo imaging from organelles to organs." Science 335, no. 6075 ( 2012): 1458-1462).
Devices that implement hybrid optical-acoustic methods combine the advantages of optical and ultrasonic devices. In particular, the weak scattering of ultrasound by biological tissues provides devices for optoacoustic bioimaging with a submillimeter resolution at depths from a few millimeters to several centimeters. At the same time, the absorption spectra of endogenous chromophores (hemoglobin, melanin, water, etc.) and exogenous markers (organic dyes, nanoparticles, fluorescent proteins, etc.) allow optical-acoustic imaging with a contrast unattainable by completely acoustic installations.
A combination of optical, optoacoustic and ultrasonic (trimodal) imaging in one device opens up interesting possibilities. In this case, complementary information on various heterogeneities in the biological tissue can be obtained.

US 20130190595 Laser Optoacoustic Ultrasonic Imaging System (LOUIS) and Methods of Use From the patent application US 20130190595 Laser Optoacoustic Ultrasonic Imaging System (LOUIS) and Methods of Use, a technical solution is known that allows simultaneous optical and ultrasonic imaging, in which a laser is used for optical and acoustic sounding of the medium, the conversion of the probing optical radiation into sounding acoustic radiation is carried out using a light-absorbing element. An ultrasonic antenna is used to record optoacoustic and backscattered acoustic pulses.
The disadvantages of the analogue is that, firstly, it does not allow optical imaging at the same time as optoacoustic and ultrasonic and, secondly, the conversion of the probe optical radiation into probe acoustic radiation is carried out using a separately located light-absorbing element, which complicates the design of the device analogue.
The closest in design to the proposed utility model is the technical solution taken as a prototype, known from patent RU137459 for the utility model Optoacoustic Microscope for Functional Bioimaging (IPC: А61В 6/00; application: 2013139983/14, priority date 08.29.2013; published 02.20.2014, Bull. No. 5). In this technical solution, an optoacoustic microcop contains a focused acoustic sensor, a system for generating optical radiation, a system for fiber-optic delivery of optical radiation from a system for generating optical radiation to a test biological tissue, a scanning system for a test biological tissue, an analog-to-digital converter connected to the output of the focused acoustic sensor and an external a personal computer brought into contact with the receiving surface of the focused acoustic sensor and investigated biological tissue immersion chamber.
The disadvantage is that this device is adapted exclusively for optoacoustic imaging of biological tissue.
The problem that the proposed utility model solves is the creation of a trimodal device that implements bioimaging through simultaneous optical, optoacoustic and ultrasonic imaging of biological tissue in vivo.
The technical effect of increasing the amount of useful information obtained by visualizing biotissue in vivo is ensured by the fact that the optoacoustic microcope contains a focusable acoustic sensor, an optical radiation generating system, a fiber optic optical radiation delivery system from the optical radiation generating system to the biological tissue under study, a system scanning the biological tissue under study, an analog-to-digital converter connected to the output of the focused acoustic sensor and external a personal computer, brought into contact with the receiving surface of the focused acoustic sensor and the biological tissue under study, an immersion chamber.
New is that the receiving surface of a spherically focused acoustic sensor is made light-absorbing.
In the proposed utility model, all signals recorded by the receiving light-absorbing surface of a spherically focused acoustic sensor are acoustic, but they are formally divided into optical, optoacoustic and ultrasonic depending on the mechanism of their occurrence (optical signals characterize the amount of diffuse optical scattering of the probe laser radiation by biological tissue, optical-acoustic characterize the value of the local optical absorption of probing laser radiation biotk Anyuh, ultrasonic signals characterize the magnitude of local acoustic scattering of probing ultrasonic radiation by biological tissue).
Использование той же самой принимающей светопоглощающей поверхности сферически фокусируемого акустического датчика для генерации зондирующих ультразвуковых импульсов и регистрации оптоакустических и ультразвуковых сигналов приводит к эффективному перемножению диаграммы направленности датчика, улучшая пространственное разрешение ультразвуковой модальности устройства (важное отличие от второго аналога).The probe ultrasonic pulses are excited in the proposed utility model by receiving a light-absorbing surface of a spherically focused acoustic sensor upon absorption of pulsed optical radiation backscattered by the studied biological tissue (a pulsed laser with a tunable wavelength is used as a system for generating optical radiation in the proposed utility model). In this case, back-scattered optical radiation absorbed by a spherically focused acoustic sensor simultaneously performs two functions: it generates a measured optical signal and a probe ultrasonic pulse. The absence in the design of the proposed utility model of a separately performed optical detector significantly facilitates the weight and dimensions of the device (compared to the first analog). Instead, the levels of acoustic signals recorded by the receiving light-absorbing surface of a spherically focused acoustic sensor at the times of laser illumination are used for optical visualization, which allows the utility model to characterize the magnitude of diffuse optical scattering at given points of the studied biological tissue. The use of the same receiving light-absorbing surface of a spherically focused acoustic sensor to generate probing ultrasonic pulses and register optical-acoustic and ultrasonic signals leads to an efficient multiplication of the radiation pattern of the sensor, improving the spatial resolution of the ultrasonic modality of the device (an important difference from the second analogue).
The utility model is illustrated by the following drawings:

8.The proposed optoacoustic microscope for bioimaging includes (see Fig. 1) a system for generating
3.The proposed trimodal device operates as follows. The one-time laser pulse generated by the optical
The amplitude of the optical signal generated by back-scattered optical radiation on the receiving light-absorbing
The optoacoustic signal (generated inside the studied
The probe ultrasonic pulse generated by the receiving light-absorbing
9).The possibility of joint registration of optical, optoacoustic and ultrasonic signals is provided by the presence of a matching liquid layer filling the
2.The optical, optoacoustic and ultrasonic A-scans registered by the receiving light-absorbing

In FIG. 2, the intravital photograph of the tumor is shown on the upper left (case a), on which the contours of the node, hemorrhages, and large vessels are marked. In Fig. 2, an optical image with a hemorrhage of a tumor highlighted by a dotted line is shown in the upper right (case b). In FIG. 2, the lower left (case c) presents an optoacoustic image with the blood vessels of the tumor, the largest are indicated by arrows. In FIG. 2, the bottom right (case d) shows an ultrasound image with the contours of the tumor node indicated by a dotted line.
To obtain a 3B image, a spherically focused
As a result of the application of the utility model, it is possible to carry out simultaneous functional optical and optoacoustic and structural ultrasonic bioimaging of the studied areas of biological tissues. As a result of optical and optoacoustic sensing, it is possible to visualize the spatial distributions of endogenous (organic dye, hemoglobin, lipids, water) and exogenous (organic dyes, nanoparticles, fluorescent proteins) fluorophores with a spatial resolution of up to 50 μm at a depth of at least 2 mm. Structural ultrasonic bioimaging is carried out simultaneously with optical and optoacoustic as a result of probing tissue with high-frequency (up to 50 MHz) ultrasound. FIG. 2 shows the capabilities of the developed trimodal device that allows for intravital imaging of the internal biological structure of the experimental tumor. While structural ultrasonic bioimaging methods have long been widely used in clinical practice, the complementary use of functional optical and optoacoustic bioimaging methods implemented in the utility model allows us to provide unique information about the state and functioning of the studied organelles, organs and organ systems. Methods of optical and optoacoustic diagnostics include, but are not limited to, the possibility of measuring local oxygenation of biological tissues, visualizing the bloodstream, diagnosing atherosclerotic changes in the walls of blood vessels, and high-contrast visualization of malignant tumors and signal lymph nodes. The utility model can be used for simultaneous functional and structural diagnostics in biomedical research related to the use of laboratory animals. It is promising to use the proposed utility model for trimodal intravital diagnosis of human soft tissues.
Thus, the proposed utility model successfully implements trimodal optical, optoacoustic and ultrasonic bioimaging and significantly increases the amount of information obtained on the visualized biological tissue compared to analogues and prototype.
