Abstract
Spiro-amidines of formula (I) and their physiologically tolerable salts are new: (n = 1 or 2; R1 and R2 = H; alkyl (opt. forming a 5-7 membered ring with the N which can be substd. by 1-3C alkyl, 1-2C alkoxy, OH or 1-4C alkoxycarbonyl) and one C atom can be replaced by O, S or N (opt. substd. by H, HCO, phenyl (itself opt. substd. by >=1 1-4C alkyl or alkoxy, methylenedioxy, OH, halo, NO2 or NH2) or 1-4C alkyl); or R1 = H and R2 = 5-8C cycloalkyl opt. substd. by 1-4C dialkylamino; R3 = H or CH3; R4 = phenyl (opt. substd. by 1-4C alkyl or alkoxy, methylenedioxy, OH, halo, CN, NO2, CF3, NH2, 2-5C acylamino or mono- or di-(1-4C)alkylamino) or thiophene residue). (I) are antidepressants usually administered at 10-60 mg./day orally or by injection. Generally the oral LD50 in mice is 0.1-0.5 g./kg.
