Abstract
A INVENÇÃO ATUAL REFERE-SE A UMA CONSTRUÇÃO GENÉTICA PARA O TRANSPORTE DE UMA PROTEÍNA DE INTERESSE EM VESÍCULAS EXTRACELULARES (EV) DE SYNECHOCYSTIS SP. PCC6803. A INVENÇÃO ATUAL RELACIONA-SE AINDA COM A ESTIRPE SYNECHOCYSTIS TRANSFORMADA DERIVADA QUE COMPREENDE O REFERIDO SISTEMA. AS CIANOBACTÉRIAS TRANSFORMADOS COM O REFERIDO SISTEMA SÃO, SURPREENDENTEMENTE, CARACTERIZADOS POR HIPERVESICULAÇÃO DOS EVS EM MAIORES QUANTIDADES DO QUE AS ESTIRPES SELVAGENS E MUTANTES CONHECIDAS NO ESTADO DA ARTE. AS CIANOBACTÉRIAS TRANSFORMADAS COM A REFERIDA CONSTRUÇÃO SEGREGAM A PROTEÍNA DE INTERESSE EM EVS E PARA O MEIO EXTRACELULAR SENDO QUE ASSIM, A INVENÇÃO PRESENTE TAMBÉM SE REFERE A COMPOSIÇÕES COMPOSTAS POR EV DERIVADAS DA REFERIDA ESTIRPE SYNECHOCYSTIS TRANSFORMADA, BEM COMO COMPOSIÇÕES QUE COMPREENDEM O MEIO EXTRACELULAR SEM EV DERIVADO DA DITA ESTIRPE TRANSFORMADA DE SYNECHOCYSTIS, PARA UTILIZAÇÃO NO TRANSPORTE DE PROTEÍNAS/ENZIMAS EM ANIMAIS, INCLUINDO MAMÍFEROS E PEIXES. A INVENÇÃO ATUAL REFERE-SE AINDA AOS MÉTODOS DE TRANSFORMAÇÃO DE CÉLULAS EPITELIAS DE MAMÍFEROS COM EVS ISOLADAS DA REFERIDA ESTIRPE SYNECHOCYSTIS, QUE INTERIORIZAM AS REFERIDAS EV E APRESENTAM A PROTEÍNA DE INTERESSE. ALÉM DISSO, A INVENÇÃO ATUAL REFERE-SE A MÉTODOS PELOS QUAIS AS EV CIANOBACTERIANAS PODEM FUNCIONAR COMO NANOTRANSPORTADORES DE PROTEÍNAS INDIVIDUALIZADAS EM PEIXES, COM APLICAÇÕES PROSPETIVAS NA MODULAÇÃO DO ESTADO NUTRICIONAL OU ESTIMULANDO RESPOSTAS IMUNES ADAPTATIVAS ESPECÍFICAS. A CONSTRUÇÃO, ESTIRPE, COMPOSIÇÕES E MÉTODOS NA PRESENTE INVENÇÃO PODEM SER VANTAJOSAMENTE UTILIZADOS COMO UMA NOVA FERRAMENTA 2 BIOTECNOLÓGICA PARA A ENTREGA ESPECÍFICA DE PROTEÍNAS E ENZIMAS BIOLOGICAMENTE ATIVAS NA BIOTECNOLOGIA EM GERAL E NA AQUACULTURA EM ESPECÍFICO.THE CURRENT INVENTION REFERS TO A GENETIC CONSTRUCTION FOR THE TRANSPORT OF A PROTEIN OF INTEREST IN EXTRACELLULAR VESCLES (EV) OF SYNECHOCYSTIS SP. PCC6803. THE CURRENT INVENTION FURTHER RELATES TO THE DERIVED TRANSFORMED SYNECHOCYSTIS STRAIN THAT COMPRISES SAID SYSTEM. THE CYANOBACTERIA TRANSFORMED WITH THE SAID SYSTEM ARE, SURPRISINGLY, CHARACTERIZED BY HYPERVESICULATION OF EVS IN GREATER AMOUNTS THAN THE WILD AND MUTANTS STRAINS KNOWN IN THE STATE OF THE ART. CYANOBACTERIA TRANSFORMED WITH SAID CONSTRUCTION SECRETATE THE PROTEIN OF INTEREST INTO EVS AND INTO THE EXTRACELLULAR ENVIRONMENT THEREFORE, THE PRESENT INVENTION ALSO REFERS TO COMPOSITIONS COMPOSED BY EV DERIVED FROM SAID TRANSFORMED SYNECHOCYSTIS STRAIN, AS WELL AS COMPOSITIONS COMPRISING THE EXTRACELLULAR MEDIUM WITHOUT EV DERIVED FROM SAID TRANSFORMED STRAIN OF SYNECHOCYSTIS, FOR USE IN THE TRANSPORT OF PROTEINS/ENZYMES IN ANIMALS, INCLUDING MAMMALS AND FISH. THE CURRENT INVENTION FURTHER REFERS TO METHODS OF TRANSFORMING MAMMALIAN EPITHELIAS CELLS WITH EVS ISOLATED FROM SAID SYNECHOCYSTIS STRAIN, WHICH INTERIORIZE SAID EV AND PRESENT THE PROTEIN OF INTEREST. IN ADDITION, THE CURRENT INVENTION REFERS TO METHODS BY WHICH EV CYANOBACTERIALS CAN FUNCTION AS NANOTRANSPORTERS OF INDIVIDUALIZED PROTEINS IN FISH, WITH PROSPECTIVE APPLICATIONS IN MODULATING NUTRITIONAL STATUS OR STIMULATING SPECIFIC ADAPTIVE IMMUNE RESPONSES. THE CONSTRUCTION, STRAIN, COMPOSITIONS AND METHODS IN THE PRESENT INVENTION CAN BE ADVANTAGEOUSLY USED AS A NEW BIOTECHNOLOGICAL TOOL FOR THE SPECIFIC DELIVERY OF BIOLOGICALLY ACTIVE PROTEINS AND ENZYMES IN BIOTECHNOLOGY IN GENERAL AND IN AQUACULTURE IN SPECIFIC.
