Abstract

A FASCIOLOSE É UMA DOENÇA ANTROPOZOONÓTICA CAUSADA PELO TREMATODO FASCIOLA HEPATICA. ESTE VERME É UM PARASITA COMUM DOS RUMINANTES, SOBRETUDO OVINOS, CAPRINOS E BOVINOS, CUJO VERME ADULTO É ENCONTRADO NOS DUCTOS BILIARES DO HOSPEDEIRO DEFINITIVO. A FASCIOLOSE É UMA ZOONOSE PARASITÁRIA QUE ACARRETA GRAVES PERDAS ECONÓMICAS COM IMPACTO NA SAÚDE PÚBLICA. OS DIAGNÓSTICOS A NÍVEL DA FASCIOLOSE SÃO ESSENCIALMENTE SEROLÓGICO E PARASITOLÓGICO PELA IDENTIFICAÇÃO DE OVOS NAS FEZES OU VERMES NO FÍGADO. NÃO ESTÃO DISPONÍVEIS DIAGNÓSTICOS QUE PERMITAM EFECTUAR O DIAGNÓSTICO NO CAMPO DIFICULTANDO O CONTROLE EFECTIVO DA DOENÇA. EFECTUAMOS O ISOLAMENTO DE CLONES DE CDNA CODIFICANDO PARA UM POLIPÉPTÍDO DE 69 AMINOÁCIDOS COM UMA MASSA MOLECULAR CALCULADA DE 8 KDA, O QUAL DESIGNAMOS POR FH8 OU FASCIOLINA (GENBANK NUMBER AE2 13970). RESULTADOS OBTIDOS MOSTRARAM QUE O FH8 TEM CARACTERÍSTICAS DE ALERGÉNIO. ESTES RESULTADOS SERVIRAM DE BASE PARA O DESENVOLVIMENTO DA INVENÇÃO QUE TEM POR OBJECTIVO A DETECÇÃO DE HOSPEDEIROS DEFINITIVOS INFECTADOS PELO VERME F. HEPATICA ATRAVÉS DA APLICAÇÃO IN VIVO DO ANTIGÉNIO FH8 (FASCIOLINA), OU OUTRO ANTIGÉNIO COM SEQUÊNCIA SEMELHANTE, A NÍVEL CUTÂNEO. OUTRO ASPECTO DA INVENÇÃO DIZ RESPEITO Á FORMULAÇÃO DA APLICAÇÃO CUTÂNEA E LEITURAS DA REACÇÃO DECORRENTE NOS OVINOS E BOVINOS. EMBORA AS FORMULAÇÕES DESCRITAS DIGAM RESPEITO A ESSES DOIS TIPOS DE HOSPEDEIRO O MÉTODO IDEALIZADO TEM APLICAÇÃO EM TODOS OS HOSPEDEIROS QUE POSSAM SER POTENCIALMENTE INFECTADOS COM F. HEPATICA. ESTES TESTES TÊM COMO PRINCIPAIS VANTAGENS A POSSIBILIDADE DE PODEREM SER REALIZADOS NO TERRENO, SEREM DE FÁCIL EXECUÇÃO E RAPIDEZ NA OBTENÇÃO DOS RESULTADOS PODENDO APRESENTAR GRANDE UTILIDADE NO CONTROLE DA FASCIOLOSE.
