Description

/ dnia 14 kwietnia 1962 POLSKIEJ RZECZYPOSPOLITEJ LUDOWEJ OPIS PATENTOWY Nr 45792 Instytut Farmaceutyczny*) Warszawa, Polska KI. 30 h, 9/02 Sposób powlekania tabletek powloczkami nierozpuszczalnymi w soku zoladkowym Patent dodatkowy do patentu nr 45717 Patent trwa od dnia 17 lutego 1961 r.Przedmiotem patentu nr 45717 jest sposób powlekania tabletek powloczkami nierozpusz¬ czalnymi w soku zoladkowym, które otrzymuje sie przez spryskiwanie mieszanina roztworów ftalanu acetylocelulozy i zeiny. Sposób ten, któ¬ ry daje doskonale wyniki w przypadkach tab¬ letek zawierajacych najrozmaitsze leki, zawiódl w sosunku do tabletek z p-aminosalicylanu so¬ dowego, p-aminosalicylanu hydrazydu kwasu izonikotynowego oraz w stosunku do tabletek zawierajacych mieszanine tych leków. Przyklad. Tabletki p^aminosalicylanu so¬ dowego (PAS-Naitrium) obustronnie wypukle o srednicy 12 mm zwilzono w drazownicy 5%-wym roztworem metakrylanu metylu w oc¬ tanie etylu i chloroformie, a nastepnie wilgotne tabletki zasypano mieszanina talku i stearynia¬ nu magnezu. Po wysuszeniu w temperaturze pokojowej, w nastepnym dniu tabletki spryska¬ no roztworem ftalanu acetylocelulozy z zeina w acetonie i etanolu. PL/ on April 14, 1962 OF THE POLISH PEOPLE'S REPUBLIC PATENT DESCRIPTION No. 45792 Pharmaceutical Institute *) Warsaw, Poland KI. 30 h, 9/02 The method of coating tablets with coatings insoluble in gastric juice Additional patent to patent No. 45717 The patent is valid from February 17, 1961 The subject of patent No. 45717 is a method of coating tablets with coatings insoluble in gastric juice, which are obtained by spraying mixture of cellulose acetate phthalate and zein solutions. This method, which is very successful with tablets containing a wide variety of drugs, has failed in the preparation of sodium p-aminosalicylate tablets, isonicotinic acid hydrazide p-aminosalicylate tablets, and with tablets containing a mixture of these drugs. These drugs, when used orally in the treatment of tuberculosis, to have fiber insoluble in gastric juice are highly desirable. These drugs have an unpleasant taste and have an irritating effect on the tongue of the stomach, which causes a number of unpleasant- *) The owner of the patent stated that the authors of the invention are Waclaw Chrzaszcz, Barbara Chodkcwska, and Andrzej Mazur, MA. symptoms such as nausea, vomiting and diarrhea. Tablets of these drugs in the known embodiment are coated with a sugar coating, which only facilitates the intake of the drug, but does not prevent irritation of the gastric mucosa. In addition, the sugar contained in the tablets reduces the laceration, which in cases of tuberculosis requiring intensive nourishment, is a serious obstacle in the treatment. It turned out that the reduction in resistance of the zein acetylcellulose phthalate coating comes from the unexplained action of the acid amino salicylic acid residue. to cellulose acetate phthalate. It has been found that when a zein-acetylcellulose phthalate coating is used, a primer that isolates the drug from this coating, the coating obtains the very normal gastric resistance that it exhibits with other drugs, while the insulating layer may be completely non-resistant to gastric juice. It has proved to be particularly advantageous to use a mixture of talc, magnesium stearate, bonded with polymethyl methacrylate as the undercoat. For this purpose, the tablets of the above-mentioned drugs are moistened with a solution of polymethyl methacrylate in an organic solvent, and then the wet tablets are sprinkled with a mixture of talc and magnesium stearate and dried. The tablets prepared in this manner are coated in the manner according to the patent no. 45717 to the filaments of zein acetylcellulose phthalate. Sodium p-aminosalicylate (PAS-Naitrium) tablets with a diameter of 12 mm on both sides were moistened in a spreader with a 5% solution of methyl methacrylate in ethyl acetate and chloroform, and then the wet tablets were sprinkled with a mixture of talc and magnesium stearate. After drying at room temperature, the next day the tablets were sprayed with a solution of acetylcellulose phthalate with zein in acetone and ethanol. The tablets were finally colored yellow by spraying with a solution of cellulose acetate phthalate containing the dye. The weight of the individual tablets increased by about 50 mg with these treatments. The tablets so coated showed complete gastric juice resistance within three hours as required by the British Pharmacopoeia of 1958, while dissolving in intestinal juice within 30 to 60 minutes. PL
