Abstract

Przedmiotem zgłoszenia jest układ enzymatyczny zawierający nanostruktury węglowe zmodyfikowane polielektrolitem kationowym, przy czym na nanostrukturach węglowych zmodyfikowanych polielektrolitem kationowym unieruchomiona jest dehydrogenaza cholesterolowa. Przedmiotem zgłoszenia jest również elektroda zawierająca podłoże przewodzące, na którym umieszczony jest wspomniany powyżej układ enzymatyczny, a także sposób wytwarzania tej elektrody i jej zastosowanie do oznaczania cholesterolu. Ponadto, przedmiotem zgłoszenia jest zastosowanie powyżej wspomnianej elektrody jako jednej z elektrod pracujących w bioczujniku zawierającym więcej niż jedną elektrodę pracującą i przystosowanym do oznaczania więcej niż jednego analitu. Kolejnym przedmiotem zgłoszenia jest bioczujnik do oznaczania cholesterolu zawierający wspomnianą powyżej elektrodę jako elektrodę pracującą. Na koniec, przedmiotem zgłoszenia jest sposób oznaczania cholesterolu obejmujący kontaktowanie próbki zawierającej cholesterol ze wspomnianym powyżej bioczujnikiem w obecności kofaktora oraz elektrochemiczne oznaczanie cholesterolu w próbce.The subject of the application is an enzymatic system containing carbon nanostructures modified with a cationic polyelectrolyte, wherein cholesterol dehydrogenase is immobilized on the carbon nanostructures modified with the cationic polyelectrolyte. The subject of the application also includes an electrode containing a conductive substrate on which the aforementioned enzymatic system is placed, as well as a method for manufacturing this electrode and its use for determining cholesterol. Furthermore, the subject of the application is the use of the aforementioned electrode as one of the working electrodes in a biosensor containing more than one working electrode and adapted to determine more than one analyte. Another subject of the application is a biosensor for determining cholesterol containing the aforementioned electrode as the working electrode. Finally, the subject of the application is a method for determining cholesterol, comprising contacting a sample containing cholesterol with the aforementioned biosensor in the presence of a cofactor and electrochemically determining cholesterol in the sample.
