Abstract

Przedmiotem zgłoszenia jest sposób oceny jakości nasion pomidora, który polega na tym, że nasiona pomidora poddaje się obróbce wstępnej oczyszczania w ten sposób, że nasiona liofilizuje się znanymi sposobami, mieli i ekstrahuje poprzez wytrząsanie w kwasie mrówkowym o stężeniu od 1,9 do 2,1 mM, korzystnie 2 mM i sonikację, następnie wiruje i tak uzyskany supernatant zbiera się a resztę materiału poddaje ponownej obróbce wstępnej oczyszczania co najmniej jeden raz, następnie uzyskane supernatanty łączy się, miesza i rozcieńcza z octanem amonu o stężeniu od 49,9 do 50,1 mM, korzystnie 50 mM i pH od 4,49 do 4,51, korzystnie 4,5, w ilości od 1,99 do 2,01 ml, korzystnie 2 ml ocatanu amonu na 2 ml supernatantu, i tak przygotowane próbki supernatantu poddaje się oczyszczaniu na hybyrdowych kolumienkach SPE w ten sposób że supernatant przepuszcza się od 1 do 10 razy korzystnie 3 razy przez kolumienki SPE w porcjach po od 0,5 ml do 1,21 ml korzystnie 1,2 ml, przemywa hybrydowe kolumienki SPE kolejno octanem amonu oraz metanolem przez przepuszczenie roztworu przez złoże hybrydowych kolumienek SPE i wymywa nukleotydy ze złoża hybrydowej kolumienki SPE roztworem metanolu, wody i octanu amonu w stosunku od 1,9/6,9/0,9 do 2,1/7,1/1,1 korzystnie 2/7/1, przy czym czyszczenie hybrydowe prowadzi się w temperaturze od 18 do 30°C korzystnie 20°C utrzymując przepływ cieczy na poziomie nie większym niż 5 ml/min, korzystnie od 3 do 4 ml/min, następnie tak otrzymany i zebrany eluat frakcji nukleotydów odparowuje się w strumieniu azotu gazowego a pozostały osad rozpuszcza w roztworze acetonitrylu i wody w stosunku od 5% do 7% acetonitrylu korzystnie 6% acetonitrylu w wodzie, używając 50 do 250 µl, korzystnie 100 µl tego rozpuszczalnika i tak otrzymany osad poddaje się analizie wysokociśnieniowej chromatografii cieczowej w urządzeniu UHPLC, a następnie analizuje się otrzymane wykresy analityczne.The subject of the application is a method for assessing the quality of tomato seeds, which consists in that the tomato seeds are subjected to a pre-treatment of purification in such a way that the seeds are lyophilized by known methods, ground and extracted by shaking in formic acid with a concentration of 1.9 to 2, 1 mM, preferably 2 mM and sonication, then centrifugation and the supernatant thus obtained is collected and the rest of the material is re-pretreated for purification at least once, then the obtained supernatants are combined, mixed and diluted with ammonium acetate with a concentration of 49.9 to 50 .1 mM, preferably 50 mM and a pH of 4.49 to 4.51, preferably 4.5, in an amount of 1.99 to 2.01 ml, preferably 2 ml of ammonium acetate per 2 ml of supernatant, and supernatant samples prepared in this way subjected to purification on hybrid SPE columns in such a way that the supernatant is passed from 1 to 10 times, preferably 3 times, through SPE columns in portions of 0.5 ml to 1.21 ml, preferably 1.2 ml, the hybrid SPE columns are washed successively with acetate ammonium and methanol by passing the solution through a bed of hybrid SPE columns and elutes the nucleotides from the bed of a hybrid SPE column with a solution of methanol, water and ammonium acetate in a ratio of 1.9/6.9/0.9 to 2.1/7.1/1 ,1 preferably 2/7/1, with the hybrid cleaning being carried out at a temperature of 18 to 30°C, preferably 20°C, maintaining the liquid flow at a level not greater than 5 ml/min, preferably from 3 to 4 ml/min, then the eluate of the nucleotide fraction thus obtained and collected is evaporated in a stream of gaseous nitrogen and the remaining precipitate is dissolved in a solution of acetonitrile and water in a ratio of 5% to 7% acetonitrile, preferably 6% acetonitrile in water, using 50 to 250 µl, preferably 100 µl of this solvent and the precipitate obtained in this way is analyzed by high-pressure liquid chromatography in a UHPLC device, and then the analytical graphs obtained are analyzed.
