Abstract

Przedmiotem zgłoszenia jest panel biosensorów ginekologiczno-onkologicznych do równoczesnego specyficznego oznaczania stężenia biomarkerów w płynach ustrojowych i homogenatach tkankowych techniką powierzchniowego rezonansu plazmonów w wersji Imaging w celu diagnostyki raka jajnika i innych schorzeń natury ginekologicznej. Panel zawiera płytkę szklaną pokrytą warstwą złota oraz siatkę polimeru tworzącą pęk miejsc aktywnych zawierających pięć różnych warstw receptorowych z przeciwciałami specyficznymi na dany marker oraz jedną warstwę odniesienia, przy czym warstwy receptorowe znajdują się na pięciu polach pomiarowych równomiernie rozłożonych na okręgu a warstwa odniesienia stanowi szóste pole pomiarowe usytuowane w środku. Dodatkowo każde z sześciu pól pomiarowych zawiera dwanaście punktów pomiarowych Wszystkie przeciwciała są połączone kowalencyjnie z linkerem — cysteaminą łączącą receptory z warstwą złota pokrywającą płytkę szklaną, a warstwa odniesienia zawiera zimmobilizowane BSA (BSA). Przy czym warstwy receptorowe pięciu pól pomiarowych równomiernie rozłożonych na okręgu stanowią królicze poliklonalne przeciwciało specyficzne na marker CA-125 (CA-125), królicze poliklonalne przeciwciało specyficzne na marker HE 4 (HE4), mysie monoklonalne przeciwciało specyficzne na marker CEA (CEA), królicze poliklonalne przeciwciało specyficzne na aromatazę (ARO) oraz mysie monoklonalne przeciwciało specyficzne na interleukine 6 (IL6).The subject of the application is a panel of gynecological and oncological biosensors for the simultaneous specific determination of biomarker concentrations in body fluids and tissue homogenates using the surface plasmon resonance technique in the Imaging version for the diagnosis of ovarian cancer and other gynecological diseases. The panel contains a glass plate covered with a layer of gold and a polymer mesh forming a bunch of active sites containing five different receptor layers with antibodies specific for a given marker and one reference layer, where the receptor layers are located on five measurement fields evenly distributed on a circle and the reference layer is the sixth field measurement located in the center. In addition, each of the six measurement fields contains twelve measurement points. All antibodies are covalently linked to the cysteamine linker that connects the receptors to the gold layer covering the glass plate, and the reference layer contains immobilized BSA (BSA). The receptor layers of the five measuring fields evenly distributed on the circle are rabbit polyclonal antibody specific for the CA-125 marker (CA-125), rabbit polyclonal antibody specific for the HE 4 marker (HE4), mouse monoclonal antibody specific for the CEA marker (CEA), a rabbit polyclonal antibody specific for aromatase (ARO) and a mouse monoclonal antibody specific for interleukin 6 (IL6).
