Abstract
Przedmiotem zgłoszenia jest sposób wytwarzania polilaktydowego substytutu kości gąbczastej, który charakteryzuje się tym, że poliaktydowy substytut kości gąbczastej zalewa się wodnym roztworem chlorowodorku chitozanu i/albo alginianu sodu i/albo hialuronianu sodu o stężeniu 0,5 - 3% w/v, ewentualnie z dodatkiem alkoholu C1-C3 w ilości do 5%obj. i utrzymuje się w warunkach próżni do 30 minut, w temperaturze 25 - 50°C, po czym nasączony substytut wyjmuje się z roztworu i zamraża w temperaturze -30 - 0°C w czasie od 5 h do 24 h, a następnie substytut poddaje się liofilizacji w czasie od 24 h do 48 h, w temperaturze -60-(-30)°C, przy ciśnieniu poniżej 120 mbar, po czym wytrąca się chitozan z substytutu liofilizowanego z chlorowodorkiem chitozanu za pomocą roztworu wodorowęglanu sodu w mieszaninie woda:alkohol C1-C3 i/albo sieciuje się alginian z substytutu liofilizowanego z alginianem sodu za pomocą wodnego roztworu chlorku wapnia o stężeniu 0,05 - 2 M, w warunkach próżni w czasie do 20 min, w temperaturze 15 - 30°C, po czym substytut płucze się i suszy.The subject of the application is a method for the production of polylactide spongy bone substitute, which is characterized in that the polylactide substitute of spongy bone is flooded with an aqueous solution of chitosan hydrochloride and / or sodium alginate and / or sodium hyaluronate at a concentration of 0.5 - 3% w / v, or by the addition of C1-C3 alcohol in an amount up to 5% vol. and kept under vacuum for up to 30 minutes at 25 - 50 ° C, then the soaked substitute is removed from the solution and frozen at -30 - 0 ° C for 5 h to 24 h, and then the substitute is subjected to lyophilization for 24 h to 48 h, at a temperature of -60 - (- 30) ° C, at a pressure below 120 mbar, after which chitosan is precipitated from a lyophilized substitute with chitosan hydrochloride using a sodium bicarbonate solution in a mixture of water: C1 alcohol -C3 and / or the alginate from the lyophilized substitute with sodium alginate is cross-linked with an aqueous solution of calcium chloride with a concentration of 0.05 - 2 M, under vacuum conditions for up to 20 minutes, at a temperature of 15 - 30 ° C, then the substitute is rinsed and dries.
