Description

Opis wynalazkuDescription of the invention

The subject of the invention is a thiazole derivative, i.e. methyl 5-(1-(2-(4-(3,4-dihydro-2 H-benzo[b][1,4]dioxepin-7-yl)thiazol-2-yl)hydrazono)ethyl)-2-hydroxybenzoate, its use as a drug component and its use as a protein tyrosine phosphatase 1B (PTP1B) inhibitor, as well as a method for its preparation.

A, SanchezLópez E, Carrasco M, Verdaguer E, Fortuna A, Folch J, Bulló M, Auladell C, Camins A, Ettcheto M. Protein tyrosine phosphatase 1B (PTP1B) as a potential therapeutic target for neurological disorders. Biomed Pharmacother. 2022;155:113709. doi:10.1016/j.biopha.2022.113709). Protein tyrosine phosphatase 1B (PTP1B, EC 3.1.3.48) is a non-receptor class I tyrosine phosphatase encoded by the PTPN1 gene. It consists of 435 amino acids and has a molecular mass of 49.966 kDa. Overexpression of protein tyrosine phosphatase 1B is observed in numerous diseases. These include metabolic diseases such as type II diabetes, in which increased PTP1B expression leads to decreased insulin receptor sensitivity, and obesity, in which protein tyrosine phosphatase 1B influences leptin activity in the hypothalamus. Leptin is responsible for glucose stabilization and plays an important role in maintaining healthy weight. Furthermore, PTPN1 gene amplification is considered a marker of severe disease progression in breast, stomach, colon, and prostate cancers. Overexpression of PTP1B also plays a role in the progression of other types of cancer, such as ovarian cancer, pancreatic cancer, and glioblastoma multiforme. It adversely affects the risk of lung and brain metastases in breast cancer and liver metastases, which may accompany colorectal cancer. Recent reports also support the significant role of protein tyrosine phosphatase 1B in neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, and multiple sclerosis (Olloquequi J, Cano A, SanchezLópez E, Carrasco M, Verdaguer E, Fortuna A, Folch J, Bulló M, Auladell C, Camins A, Ettcheto M. Protein tyrosine phosphatase 1B (PTP1B) as a potential therapeutic target for neurological disorders. Biomed Pharmacother. 2022;155:113709. doi:10.1016/j.biopha.2022.113709). This is of great importance due to the fact that most of the above-mentioned neurodegenerative diseases, due to their extremely complex course, are treated only symptomatically.

Znalezienie skutecznego inhibitora białkowej fosfatazy tyrozynowej 1B stanowi zatem wyjątkowo atrakcyjny cel terapeutyczny, dający nadzieję na skuteczną terapię farmakologiczną schorzeń, które w dalszym ciągu nie posiadają efektywnych schematów terapeutycznych.Finding an effective inhibitor of protein tyrosine phosphatase 1B is therefore an exceptionally attractive therapeutic target, offering hope for effective pharmacological therapy for diseases that still lack effective therapeutic regimens.

Currently, there is no registered PTP1B inhibitor approved for the treatment of any of the above-mentioned conditions, as the molecules proposed for this use have not yet undergone clinical trials.

Jednakże możliwe jest porównanie skuteczności w aspekcie inhibicji białkowej fosfatazy tyrozynowej 1B, jaka charakteryzuje przedmiotowy wynalazek, względem inhibitora referencyjnego o dobrej przepuszczalności przez błony biologiczne, jakim jest (nazwa IUPAC): 3-(3,5-Dibromo-4-hydroxy-benzoyl)-2-ethyl-benzofuran-6-sulfonicacid-(4-(thiazol-2-ylsulfamyl)-phenyl)-amide (numer CAS: 765317-72-4) o udowodnionej aktywności względem PTP1B, wynoszącej IC50 = 4-8 μM.However, it is possible to compare the effectiveness in terms of inhibition of protein tyrosine phosphatase 1B, which characterizes the present invention, with respect to a reference inhibitor with good permeability through biological membranes, which is (IUPAC name): 3-(3,5-Dibromo-4-hydroxy-benzoyl)-2-ethyl-benzofuran-6-sulfonicacid-(4-(thiazol-2-ylsulfamyl)-phenyl)-amide (CAS number: 765317-72-4) with proven activity against PTP1B, amounting to IC50 = 4-8 μM.

Do fazy II badań klinicznych zgłoszony został m.in. inhibitor PTP1B o nazwie Ertiprotafib i o formule C31H27BrO3S. Charakteryzujące go IC50 zawierające się w przedziale od 1,6 do 29 μΜ było zależne od warunków prowadzonej analizy (Erbe DV, Wang S, Zhang YL, et al. Ertiprotafib improves glycemic control and lowers lipids via multiple mechanisms. Mol Pharmacol. 2005;67(1): 69-77. doi:10.1124/mol.104.005553). To date, few potential inhibitors have been approved for clinical trials. Ertiprotafib, a PTP1B inhibitor with the formula C31H27BrO3S, has been submitted for Phase II clinical trials. Its IC50, ranging from 1.6 to 29 μM, was dependent on the assay conditions (Erbe DV, Wang S, Zhang YL, et al. Ertiprotafib improves glycemic control and lowers lipids via multiple mechanisms. Mol Pharmacol. 2005;67(1):69-77. doi:10.1124/mol.104.005553). These values did not meet expectations, and ertiprotafib was found to be insufficiently selective for protein phosphatase 1B. Similarly, TTP814 was completed in Phase II of clinical trials - it was characterized by selectivity for PTP1B over TCPTP, but the study was discontinued in 2015. Ionis Pharmaceuticals, Inc. had two drugs in its product range: ISIS 113715 (ClinicalTrials.gov ID: NCT00330330) and ISISPTP1BRx (ClinicalTrials.gov ID: NCT01918865), which were also included in Phase II of clinical trials, however, these studies were also not continued, and sufficient data on the safety of the drugs were not provided.

Zastosowanie pochodnych tiazolu, jako selektywnych inhibitorów białkowej fosfatazy tyrozynowej 1B zostało już przedstawione w pracach naukowych na przykładzie JTT-551, opracowanego przez Japan Tobacco Company, który w badaniach przedklinicznych wykazał się wysoką aktywnością i selektywnością względem PTP1B, jednakże zaprzestano kontynuowania badań z uwagi na wykazywane przez niego działania niepożądane (Qian S, Zhang M, He Y, Wang W, Liu S. Recent advances in theThe use of thiazole derivatives as selective inhibitors of protein tyrosine phosphatase 1B has already been presented in scientific works on the example of JTT-551, developed by Japan Tobacco Company, which in preclinical studies showed high activity and selectivity towards PTP1B, however, further studies were discontinued due to its side effects (Qian S, Zhang M, He Y, Wang W, Liu S. The precipitate is filtered off using a vacuum filtration set with a Buchner funnel, and the product is purified by column chromatography using the eluent dichloromethane and methanol in a 95:5 ratio. Column chromatography is carried out in such a way that the product is dissolved in a small amount of dichloromethane and then introduced onto an adsorbent which is a silica gel with a bed diameter (230-400 mesh) placed in a cylindrical column. The flow of the eluent is forced by means of a pressure pump. As the components move down the column, they separate into individual fractions, which are analyzed by TLC. The solvent from the pure fraction is evaporated using a rotary evaporator, yielding pure product. 0.35 g of the compound (65.06% of theoretical yield) was obtained, with a melting point exceeding 260°C and noticeable decomposition. The correct structure of the compound and its purity were confirmed by analysis of 1H and 13C NMR and HRMS spectra.

Analiza prowadzona była w trzech niezależnych powtórzeniach dla 10 różnych stężeń (100 nM-500 μΜ) dla 5-(1-(2-(4-(3,4-dihydro-2H-benzo[b][1,4]dioksepin-7-ylo)tiazol-2-ylo)hydrazono)etylo)-2-hydroksybenzoesanu metylu o wzorze 1 i 10 różnych stężeń dla inhibitora referencyjnego (100 nM-50 μΜ).The assessment of potential biological activity is based on the ability of phosphatase (E12020, Invitrogen™, Thermo Fisher Scientific Inc.) to degrade the non-fluorescent substrate 6,8-difluoro-4-methylumbelliferyl phosphate (DiFMUP) (D6567, Invitrogen™, Thermo Fisher Scientific Inc.), specific for assessing phosphatase activity. Degradation of the substrate by the phosphatase releases the fluorophore 6,8-difluoro-4-methylumbelliferyl phosphate (DiFMUP). Its concentration is directly proportional to enzyme activity. The product has a fluorescence peak at an excitation wavelength of 358 nm and an emission wavelength of 455 nm; therefore, analysis of fluorescence, which translates to enzymatic activity, was performed under these conditions using a microplate reader (Agilent BioTek Synergy H1 Microplate Reader). A thiazole derivative, a non-competitive, allosteric inhibitor 3-(3,5-Dibromo-4-hydroxy-benzoyl)-2-ethyl-benzofuran-6-sulfonicacid-(4-(thiazol-2-ylsulfamyl)-phenyl)-amide (CAS number: 765317-72-4) (539741, Sigma-Aldrich), was used as a reference inhibitor. The analysis was performed in three independent replicates for 10 different concentrations (100 nM-500 μM) of methyl 5-(1-(2-(4-(3,4-dihydro-2H-benzo[b][1,4]dioxepin-7-yl)thiazol-2-yl)hydrazono)ethyl)-2-hydroxybenzoate of formula 1 and 10 different concentrations of the reference inhibitor (100 nM-50 μM).

Wyniki analizy dla inhibitora referencyjnego w trzech powtórzeniach znajdują się w Tabeli 1.The results of the triplicate reference inhibitor analysis are shown in Table 1.

W celu rozpoczęcia reakcji dodano 50 μΜ roztworu substratu. Doświadczenie dla wynalazku wykonano w pięciu powtórzeniach, a wyniki przedstawiono w Tabeli 1.After obtaining results indicating significant activity of methyl 5-(1-(2-(4-(3,4-dihydro-2H-benzo[b][1,4]dioxepin-7-yl)thiazol-2-yl)hydrazono)ethyl)-2-hydroxybenzoate of formula 1, the analysis was repeated only for it by performing the analysis in five independent replicates for 10 different, lower concentrations (1 nM-10 μM). The procedure for both analyses was identical. Reactions were carried out in an assay buffer consisting of 0.5 M sodium acetate solution (pH 5.0). Initially, different concentrations of the test compound (1 nM-10 μM) were incubated with phosphatase solution (10 mU of enzyme in a total reaction volume of 50 μM) at 25°C. To start the reaction, 50 μM substrate solution was added. A mixture of enzyme and substrate without inhibitor was used as a positive control. A reaction mixture with substrate, without enzyme, and without inhibitor was used as a negative control. The final volume of the reaction mixture was 100 μM. The measured sample contained 1% dimethyl sulfoxide and 1% N,N-dimethylformamide. The increase in fluorescence was monitored at an excitation wavelength of 358 nm and an emission wavelength of 455 nm in a 96-well plate for 30 min. The experiment for this invention was performed in five replicates, and the results are presented in Table 1.
