Abstract
Przedmiotem zgłoszenia są koniugaty 5-fluorouracylu i dichlorooctanu 2-hydroksyetylu o wzorze ogólnym 1, w którym n oznacza przynajmniej jedną grupę metylenową, korzystnie n oznacza 1-5 grup metylenowych oraz sposób ich otrzymywania, który polega na reakcji związku o wzorze 2 ze związkiem o wzorze 3 w obecności odczynnika sprzęgającego w rozpuszczalniku aprotonowym, w wyniku której otrzymuje się związek o wzorze 4, który po rozpuszczeniu w rozpuszczalniku protonowym przekształcany jest podczas katalitycznego uwodornienia w związek o wzorze 5, który po rozpuszczeniu w rozpuszczalniku aprotonowym w reakcji ze związkiem o wzorze 6 w obecności odczynnika sprzęgającego daje związek o wzorze ogólnym 1.The subject of the application are conjugates of 5-fluorouracil and 2-hydroxyethyl dichloroacetate of the general formula 1, in which n represents at least one methylene group, preferably n represents 1-5 methylene groups, and a method of obtaining them, which consists in reacting the compound of formula 2 with a compound of formula 3 in the presence of a coupling reagent in an aprotic solvent, which gives a compound of formula 4, which, after dissolution in a protic solvent, is transformed during catalytic hydrogenation into a compound of formula 5, which, after dissolution in an aprotic solvent, reacts with a compound of formula 6 in the presence of a coupling reagent gives a compound of general formula 1.

Adypinian benzylu otrzymano w reakcji monoadypinianu sodu z bromkiem benzylu. Kwas adypiniowy (12.23 g, 83.71 mmol) rozpuszczono w ługu sodowym (NaOH: 4.69 g, 83.71 mmnol; H2O: 42 ml) i mieszano przez 1 godzinę. Następnie oddestylowano rozpuszczalnik na wyparce obrotowej, a do suchej pozostałości dodano toluen (40 ml). Całość mieszano przez 2 min i ponownie oddestylowano rozpuszczalnik na wyparce obrotowej. Czynność powtórzono trzykrotnie. Do otrzymanego białego osadu dodano ponownie toluen (250 ml), bromek tetrabutyloamoniowy (2.5 g) oraz bromek benzylu (19.25 ml, 83.71 mmol). Całość ogrzewano w temperaturze wrzenia przez 6 godz., a następnie oddestylowano rozpuszczalnik na wyparce obrotowej. Otrzymaną pozostałość ostrożnie rozpuszczono w nasyconym roztworze wodorowęglanu sodu (200 ml) i frakcję wodną przemyto eterem dietylowym (2x50 ml), a następnie zakwaszono stężonym kwasem solnym do pH 3 i ekstrahowano octanem etylu (3x50 ml). Frakcje organiczne wysuszono siarczanem magnezu, oddestylowano rozpuszczalnik i otrzymano bezbarwny olej, który krzepnie w biały osad (6.9 g, 35%).Benzyl adipate was obtained by reacting sodium monoadipate with benzyl bromide. Adipic acid (12.23 g, 83.71 mmol) was dissolved in sodium hydroxide solution (NaOH: 4.69 g, 83.71 mmol; H2O: 42 mL) and stirred for 1 hour. The whole was heated at reflux temperature for 6 hours, and then the solvent was distilled off on a rotary evaporator. The obtained residue was carefully dissolved in saturated sodium bicarbonate solution (200 ml) and the aqueous fraction was washed with diethyl ether (2x50 ml), then acidified with concentrated hydrochloric acid to pH 3 and extracted with ethyl acetate (3x50 ml). The organic fractions were dried over magnesium sulfate and the solvent was distilled off to give a colorless oil that solidified into a white solid (6.9 g, 35%).

Do roztworu kwasu 6-((5-fluoro-2,4-diokso-3,4-dihydropirymidin-1(2 H)-ylo)metoksy)-6-oksoheksanowego (440 mg, 1.71 mmol) w mieszaninie bezwodnego acetonitrylu (17 ml) i DMF (5 ml) dodano dichlorooctan 2-hydroksyetylu (0.26 ml, 2.22 mmol), DCC (577 mg, 2.22 mmol) oraz DMAP (18 mg, 0.15 mmol). Całość mieszano w temperaturze pokojowej przez noc. Wytrącony dicykloheksylomocznik (DCU) odsączono przez warstwę Celitu, z przesączu oddestylowano rozpuszczalnik na wyparce obrotowej, a otrzymaną oleistą pozostałość oczyszczono na kolumnie chromatograficznej (heksan-aceton; 4 : 1, 3 : 1, 1 : 1, v/v).
