Kość ludzka może być uważana za przykład kompozytu nieorganiczno-organicznego, zawierającego około 70% wagowych nieorganicznych kryształów, głównie hydroksyapatytu o wzorze chemicznym Ca5(PO4)3OH, oraz 30% wagowych macierzy organicznej, głównie kolagenu typu I [Osteoblast mineralization with composite nanofibrous substrate for bone tissue regeneration, Venugopal JR, Giri Dev VR, Senthilram T, Sathiskumar D, Gupta D, Ramakrishna S., Cell Biology International, 35, 2011]. Jako źródło jonów wapnia stosuje się najczęściej sole wapnia rozpuszczalne w wodzie, takie jak azotan lub chlorek, a jako źródło jonów fosforanowych stosuje się rozpuszczalne w wodzie fosforany amonu, sodu lub wapnia. Reakcję prowadzi się zwykle w środowisku zbliżonym do obojętnego, w temperaturze pokojowej lub podwyższonej.Water-soluble calcium salts such as nitrate or chloride are most commonly used as the source of calcium ions, and water-soluble ammonium, sodium or calcium phosphates are used as the source of phosphate ions. W reakcji według powyższego równania uzyskiwany jest hydroksyapatyt charakteryzujący się właściwym stosunkiem molowym atomów Ca/P równym 1,67, co jest wartością charakteryzującą hydroksyapatyt pochodzenia naturalnego [Calcium phosphates as substitution of bone tissues, VelletRegi M, Gonzalez-Calbet JM, Progress in Solid State Chemistry, 32, 2004]. Gęsty hydroksyapatyt znajduje szereg zastosowań w zastępowaniu kości i jest wykorzystywany do naprawy uszkodzeń kostnych w ubytkach dentystycznych i ortopedycznych, natychmiastowego zastąpienia zębów, powiększania krawędzi pęcherzykowych, jako materiał do pokrycia miazgi oraz w rekonstrukcji szczękowo-twarzowej [Synthesis of stoichiometric nano crystalline hydroxyapatite by ethanol-based solgel technique at low temperature, Kuriakose TA, Kalkura SN, Palanichamy M, Arviuoli D, Dierks K, Bocelli G et al., Journal of Crystal Growth, 263, 2004].The reaction according to the above equation yields hydroxyapatite with the appropriate Ca / P molar ratio equal to 1.67, which is a value characterizing hydroxyapatite of natural origin [Calcium phosphates as substitution of bone tissues, VelletRegi M, Gonzalez-Calbet JM, Progress in Solid State Chemistry, 32, 2004]. Niestety, kryształy strącanego w czysto wodnych roztworach soli hydroksyapatytu są stosunkowo duże, o rozmiarach rzędu kilkudziesięciu, a nawet do kilkuset mikrometrów. Tak otrzymywane kryształy są ponadto całkowicie nierozpuszczalne i nie ulegają przebudowie w organizmie ludzkim. Ilość 0,0075 moli (1,775 g) Ca(NO3)2 4H2O rozpuszczono w 25 ml wody demineralizowanej. Przygotowano 9% roztwór lecytyny w 37,5ml wody demineralizowanej. Roztwór lecytyny wkroplono do roztworu tetrahydratu azotanu wapnia. Wartość pH uzyskanej mieszaniny reakcyjnej ustalono na 10 oraz mieszaninę ustawiono w kąpieli wodnej na 60°C oraz mieszano. Do mieszaniny reakcyjnej wkroplono 15 ml roztwór 0,0046 moli (0,594 g) (NH4)2HPO4 w wodzie demineralizowanej o pH ustalonym na wartość 10 z szybkością 2 ml/h. Po wkropleniu mieszaninę reakcyjną mieszano przez 1,5 godziny w temperaturze reakcji - 60°C. Następnie mieszaninę naturalnie schłodzono do temperatury otoczenia, poddano wirowaniu, dekantacji. Uzyskany biały produkt przemyto pięciokrotnie heksanem w celu usunięcia pozostałości lecytyny. Morfologię uzyskanego produktu scharakteryzowano za pomocą elektronowej mikroskopii skaningowej (SEM) i przedstawiono na Fig. 6. Do mieszaniny reakcyjnej wkroplono 15 ml roztworu 0,0046 moli fosforanu sodu w wodzie demineralizowanej o pH ustalonym na wartość 9. Po wkropleniu mieszaninę reakcyjną mieszano przez godzinę w temperaturze reakcji 50°C. Następnie mieszaninę naturalnie schłodzono do temperatury otoczenia i poddano wirowaniu. Uzyskany biały proszek przemyto dwukrotnie etanolem 95% w celu usunięcia nadmiaru lecytyny. Uzyskano produkt o morfologii nanosfer o średnicach od około 30 nm do 110 nm.The lecithin solution was added dropwise to the calcium chloride solution.
