Przedmiotem wynalazku jest zastosowanie medyczne pochodnych 1-podstawionego 4-benzoilotiosemikarbazydu o wzorze ogólnym 1, przedstawionym na rysunku, gdzie R oznacza podstawnik etylowy lub bifenylowy.The present invention relates to the medical use of 1-substituted 4-benzoylthiosemicarbazide derivatives of the general formula I as shown in the figure, wherein R represents an ethyl or biphenyl substituent.
Pochodne 1-podstawionego 4-benzoilotiosemikarbazydu występują w znanym stanie techniki. Ich aktywność nie była dotychczas znana.1-substituted 4-benzoylthiosemicarbazide derivatives exist in the known art. Their activity has not been known so far.
Znane są z literatury [Molecules, 2010, 15, 4267] także pochodne 1,4-dipodstawionego (tio)semikarbazydu, które wykazują aktywność przeciwnowotworową wobec komórek białaczki mieloblastycznej HL60. Należą do nich pochodne semikarbazydu podstawionego w pozycji 1 podstawnikiem arylofuroilowym, zaś w pozycji 4 orto i/lub para podstawionym podstawnikiem benzoilowym. Znane są także pochodne tiosemikarbazydu wykazujące aktywność w stosunku do kilku rodzajów komórek nowotworowych: raka jamy ustnej (KB), raka jamy nosowo-gardłowej (CNE2), raka piersi (MCF-7), raka żołądka (MGC-803) oraz raka płuc (GLC-82), [Eur. J. Med. Chem. 2012, 154, 925].Derivatives of 1,4-disubstituted (thio) semicarbazide are also known from the literature [Molecules, 2010, 15, 4267], which show antitumor activity against HL60 myeloblastic leukemia cells. Nowotwory złośliwe, obok chorób układu krążenia, są głównymi przyczynami przedwczesnych zgonów wśród Polaków. Zgodnie z danymi podanymi przez Centrum Onkologii Instytutu im. M. Skłodowskiej-Curie, w 2006 roku do rejestrów nowotworów złośliwych wpłynęły informacje o prawie 126 tys. nowych zachorowań i stwierdzono ponad 91500 zgonów z tego powodu, zaś 390 tys. Polaków żyje z chorobą nowotworową. Najczęściej rejestrowanymi nowotworami u mężczyzn były nowotwory złośliwe płuca - 23.6%, jelita grubego - 12% i gruczołu krokowego - 11.2%. U kobiet zaś najczęściej rejestrowany był nowotwór złośliwy piersi - 21.5%, jelita grubego - 10.1% oraz płuca 8.2% (Świątek, Aptekarz Polski 2009, 31/9 online). Niezadowalające wyniki leczenia większości nowotworów złośliwych, wynikają zarówno z braku skutecznych metod, toksyczności, jak i z narastającej oporności wielolekowej. Ograniczenia te wymuszają poszukiwania innych, bardziej skutecznych i mniej toksycznych leków. Badania naukowe ukierunkowane są na modyfikację form farmaceutycznych obecnie stosowanych leków onkologicznych, poszukiwanie leków przeciwnowotworowych wśród leków obecnie stosowanych z innych wskazań oraz poszukiwanie nowych związków chemicznych pochodzenia naturalnego lub syntetycznych o działaniu przeciwnowotworowym. Pochodne o wzorze 1 mogą mieć zastosowanie jako lek do leczenia nowotworu piersi. Badane związki dodawano do podłoża hodowlanego i inkubowano przez 24 godziny. Po tym 3 czasie do podłoża dodawano 0,5 μ Ci [ H]-tymidyny (aktywność właściwa 6,7 Ci/mmol) i dalej inkubowano komórki przez 4 godziny, w inkubatorze w atmosferze zawierającej 5% CO2 (lub bez CO2 - komórki MDA-MB-231), w temperaturze 37°C. Dla porównania aktywności związków według wynalazku, wyznaczono wartości IC50 dla znanego i stosowanego leku - chlorambucylu. Wynosiły one odpowiednio 65 μM dla linii MCF-7 i 56 μM dla linii MDA-MB-231. Tak więc, związek 1b jest silniejszym inhibitorem biosyntezy DNA w komórkach linii MCF-7 niż chlorambucyl.
