Cl.3 C12N 11/08 Creators of the invention: Dieter Kirstein, Peter Mohr, Marian Filipiak, Jerzy Krauze, Florian Schubert. Authorized by the patent: Akademia Ekonomiczna, Poznan (Poland) Method for the production of insoluble biocatalysts Invention It relates to a process for the preparation of insoluble biocatalysts by immobilizing biologically active compounds, especially enzymes, on new polymeric supports. These biocatalysts are intended for heterogeneous biocatalysis and affinity chromatography, especially in biotechnology and medicine. There are methods for the production of insoluble biocatalysts in which the immobilization of biologically active compounds occurs by adsorption, ion exchange, covalent bonding, gel, copolymerizations or physical demarcation, e.g. by means of membranes. Covalent bonding occurs after chemical modification or activation of the carrier or the immobilized compound. Suitable carriers are natural substances, such as minerals, chitin or organic polymers, and synthetic inorganic substances, for example glass. In addition to substances obtained by polymerization of monomers containing double bonds, polycondensation polymers such as polyamides, polyesters and the like or obtained by polyaditions such as polyurethanes are also used as carriers. For the polycondensation reaction, the diamines are treated with acids dicarboxylic compounds and their derivatives, for example polyamides as well as dialdehydes. Known polymers used as carriers require, as a rule, chemical modification in order to be able to attach biologically active compounds, or show only a slight hydrolytic stability. The aim of the invention is to develop a new one. a medium for immobilizing biologically active compounds, which is able to attach such compounds to the solution adsorptively or covalently with high efficiency without any further activation steps, with unmanageable loss of biological activity. According to the invention, this problem is solved by diamines of the general formula RiNH-R2 NHR3, in which Ri and R3 are optionally substituted hydrocarbon residues or hydrogen atoms, and R2 is an aliphatic, aromatic or heterocyclic residue. , is subjected to polycondensation in solution with an excess of benzoquinome, preferably in the form of its hydroquinone addition product - quinhydrone, especially at a mox ratio of about 1: 2. Both substances are dissolved, for example, in water, With buffers, alcohol, acetone or other solvents and stirred. The resulting water-insoluble polymer is filtered off, washed with solvents and suspended in water. Such polymeric carriers are used to immobilize biocatalysts. For the immobilization of biocatalysts on the medium of biocatalysts, especially the enzymes 128 365 ^ G | l ^ vOS), SN 128 365 and coH ^ W ^ M ^, this medium is treated with an aqueous, preferably buffer solution of the biocatalyst with a concentration of 1 - 50 mg / cm3 at the temperature 0-30 ° C, especially at room temperature. Due to the high activity of the carrier prepared in accordance with the invention, the immobilization of biologically active compounds can be achieved by simple filtration of solutions of these compounds through the carrier layer, the most convenient way to use for this purpose a column filled with a carrier and to act in the manner known from chromatography column The obtained insoluble biocatalysts show high specific activity, they are suitable for. transformations in the heterogeneous j phase "! f! o; separation processes carried out on the principle of affinity chromatographide. The invention is explained on the basis of examples. Example I; Preparation of the carrier 0.1 mole of p-phenylenediamine was mixed with 10 cm3 of water and a solution of 0.22 mole of p-benisoquinone in 10 cm3 of acetone was added. The mixture was stirred for 5 hours. 20 g of black was obtained. polymer in the form of a precipitate which was separated by filtering and then washed with acetone, a mixture of acetone and water (1: 2) and water. An aqueous suspension was then formed and stored in this form until Example II was used. Preparation of the carrier. 0.1 mole of p-phenyldiamine and 0.22 mole of quinhydron were dissolved separately in 20 cm3 portions of ethyl alcohol, then both solutions were combined, and after 2 hours the precipitated polymer (20 g) was filtered off, washed with alcohol and water, then an aqueous suspension was made and kept in this form until use. Example III. Preparation of the carrier. 0.1 mole of hexamethylenediamine and 0.22 mole of quinhydran were dissolved separately in 20 cm3 portions of ethyl alcohol, both solutions were combined, and the resulting polymer (20 g) was washed with ethyl alcohol and water, and an aqueous suspension was prepared in this form. stored until use. Example IV. Immobilization of a biologically active compound. 1 g of the carrier prepared according to Example 2 was mixed with a solution of 50 mg of glucose oxidase in 10 cm 3 of water and shaken for 2 hours at room temperature. The product was then drained, washed with water and with an acetate buffer (pH 5.6; J = 0.1). Immobilization efficiency 10—16%. and Example V. Immobilization of a biologically active compound. .1 g of the carrier obtained according to example II was shaken for 3 hours with a solution of 20 mg of catalase in 10 cm3 of phosphate buffer (pH 7.0; J = 0.1), then washed with this buffer and water (immobilization efficiency 10-30%) Example VI Biological immobilization of the active compound. 1 g of the carrier obtained according to Example 2 was shaken for 30 minutes with a solution of 30 mg of NADP in 10 cm 3 of water, the product was drained and washed with water (immobilization efficiency 20-40%). Application. 1 g of the product obtained according to example 4 and 1 g of the product obtained according to example 5 were mixed with an aqueous glucose solution (30 g / ml). Air was introduced into the suspension and the glucose was oxidized to glutenic acid by catalytic action of immobilized glucose oxidase. The hydrogen peroxide formed at the same time was decomposed by the action of immobile catalase. The rate of gluten acid formation was 1 X 10-4 moles / min. Example VIII. Use. 1 g of the product obtained according to example VI was treated on the column with a solution of glucose dehydrogenase (50 units / cm3) in Sdrensen buffer (pH 7; J = 0.05). The enzyme was specifically bound to the biocatalyst. The eluate from the column contained less than 5 units of glucose dehydrogenase per cc. The enzyme is eluted from the column with an aqueous solution of NADP (7 mg / cm3). Yield 75%. PL
