Abstract

The present invention provides immunoresponsive cells, including T cells, cytotoxic T cells, regulatory T cells, and Natural Killer (NK) cells, expressing an antigen recognizing receptor and an inhibitory chimeric antigen receptor (iCAR). Particularly, the antigen binding receptor binds to a first antigen on a tumour tissue, activating the immunoresponsive cell whereas the iCAR binds to a second antigen that is not expressed on said tumor tissue to inhibit the cytotoxicity of the immunosuppressive cell. This achieves reduced off-target cytotoxic effects of the immunoresponsive cell during immunotherapy procedures.
