Disclosed are immunoglobulin single variable domain constructs such as ScFVs, VHHs and nanobodies comprising modifications to the C-terminal region that reduce protein interference, for example auto antibodies. In particular C-terminal extensions of the VTVSS sequence at the C-terminal end and point mutations at selected sites within the C-terminal region when the protein is folded.
