Abstract

The present invention relates to novel antibodies and fragments thereof that binds cannabinoid 1 (CB1) receptor. The antibodies and fragments thereof as disclosed herein include humanized antibodies that bind CB1 receptor. The invention also includes uses of the antibodies for treating a disease or disorder responsive to antagonism or agonism of the CB1 receptor.

Claims (2)

CLAIMS An isolated antibody or antigen binding fragment thereof that binds specifically to cannabinoid 1 (CB1) receptor, comprising a heavy chain CDR1, CDR2, and CDR3 according to SEQ ID NOs: 352, 353, and 354, respectively, and a light chain CDR1, CDR2, and CDR3 according to SEQ ID NOs: 355, 356, and 357, respectively. The isolated antibody or antigen binding fragment of claim 1, wherein the antibody or antigen binding fragment has at least one of the following properties: is an allosteric modulator; and inhibits CB1 receptor signaling activity. The isolated antibody or antigen binding fragment of claim 1 or 2, wherein the antibody is conjugated to an agent selected from an additional therapeutic agent, a cytotoxic agent, an immunoadhesion molecule, or an imaging agent. An in vitro method of antagonizing CB1, the method comprising contacting a cell expressing CB1 receptor with an antibody or binding fragment according to any one of claims 1- A host cell expressing the isolated antibody or fragment according to any of claims 1-3, wherein said host cell is not present in a human being. The isolated antibody or antigen binding fragment of claim 1, wherein one of light chain CDR1, CDR2 and CDR3 or the heavy chain CDR1, CDR2 and CDR3 may comprise one amino acid substitution, according to one of: the heavy chain CDR1 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 443-463; the heavy chain CDR2 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 464-577; the heavy chain CDR3 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 578-625; the light chain CDR1 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 626-661; the light chain CDR2 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 662-742; and the light chain CDR3 comprises an amino acid sequence selected from the group consisting of SEQ ID NOs: 743-824. The isolated antibody or fragment according to any one of claims 1-6, wherein the antibody or antigen binding fragment thereof has at least one of the following properties: is a humanized antibody; comprises IgG1 Fc region; comprises a modified Fc region. An isolated antibody or antigen binding fragment thereof that binds to cannabinoid receptor 1 (CB1), wherein the antibody or antigen binding fragment thereof comprises a heavy chain variable region amino acid sequence selected from the group consisting of SEQ ID NOs: 339-341, and a light chain variable region according to SEQ ID NO: 337. The isolated antibody or antigen binding fragment of claim 8, wherein the heavy chain variable region comprises SEQ ID NO: 341 and further comprising a heavy chain constant region comprising SEQ ID NO: 433, SEQ ID NO: 434, or SEQ ID NO: 435. The isolated antibody or antigen binding fragment of claim 8, wherein the heavy chain variable region comprises SEQ ID NO: 340 and further comprising a heavy chain constant region comprising SEQ ID NO: 433, SEQ ID NO: 434, or SEQ ID NO: 435. An isolated humanized antibody or antigen binding fragment thereof of any one of claims 1-10, wherein the antibody or fragment has at least one of the following properties: a binding affinity Kd for CB1 receptor of about 100 nM or less; CB1 receptor inhibiting activity that is at least 2 fold more potent relative to the corresponding non-humanized or chimeric antibody or fragment; exhibits reduced or absent brain penetration; exhibits reduced central nervous system (CNS) side effects relative to a small molecule CB1 receptor agonist or antagonist, for example AM6545, AM251, taranabant, or rimonabant. Use of the isolated antibody or antigen binding fragment of any one of claims 1-10 in the manufacture of a medicament for treating a disease or disorder selected from the group consisting of obesity, diabetes, dyslipidemia, metabolic diseases, fibrosis, non-alcoholic steatohepatitis (NASH), liver disease, primary biliary cirrhosis, renal disease, kidney fibrosis, chronic kidney disease, osteoporosis, atherosclerosis, cardiovascular disease, cancer, and an inflammatory disease. Use of the antibody or antigen binding fragment according to any one of claims 1-10 in the manufacture of a medicament for treating a disease or disorder is selected from the group consisting of pain, multiple sclerosis spasticity and glaucoma. The use of claim 12, wherein the disease or disorder is obesity. The use of claim 12, wherein the disease or disorder is liver fibrosis. The use of claim 12, wherein the disease or disorder is NASH. The use of claim 12, wherein the disease or disorder is kidney fibrosis. An isolated antibody or antigen binding fragment thereof as claimed in any one of claims
1-3 and 6-10, substantially as described herein and with reference to any example thereof. An in vitro method as claimed in claim 4, substantially as described herein and with reference to any example thereof. A host cell as claimed in claim 5, substantially as described herein and with reference to any example thereof. An isolated humanized antibody or antigen binding fragment thereof as claimed in claim 11, substantially as described herein and with reference to any example thereof. A use as claimed in any one of claims 1
2-17, substantially as described herein and with reference to any example thereof.
