The invention is within the field of autoimmune diseases and other immune system-mediated disease treatments that use collagen and/or derivatives thereof.
Mange immunsystem-formidlede sykdommer involverer en uønsket inflammatorisk respons, f.eks. sykdommer som inkluderer reumatoid artritt, kronisk hepatitt, Crohns sykdom, Although the immune system is essential for fighting infections, the immune response to the infections (foreign antigens) and the immune response to molecules produced in the body (autologous antigens) can result in numerous diseases, i.e. immune system-mediated diseases. Immune system mediated diseases can be either B cell mediated (ie antibody mediated) or T cell mediated. In addition, immune system-mediated diseases can be caused by immune complexes formed between antibodies and antigens (either foreign or autologous). Many immune system-mediated diseases involve an unwanted inflammatory response, e.g. diseases that include rheumatoid arthritis, chronic hepatitis, Crohn's disease,
psoriasis, vaskulitt og lignende.psoriasis, vasculitis and the like.
Existing therapies for immune system-mediated diseases, particularly immune system-mediated diseases that result in an unwanted inflammatory response, such as rheumatoid arthritis, are insufficient. Most immune system-mediated diseases are chronic conditions that require long-term administration of drugs. It is therefore important to use relatively non-toxic drugs. Many compounds are used for the treatment of autoimmune diseases, e.g. steroids and non-steroidal anti-inflammatory compounds, however, have produced toxic side effects that become apparent after long-term use. Immunosuppressive drugs have also been used to treat autoimmune responses. Such immunosuppressive drugs, e.g. cyclosporin A and azathioprine are relatively non-specific and have the negative effect of weakening the entire immune system, thereby making the patient susceptible to infectious diseases.
Oral administration of the compounds has been shown to induce immune tolerance with respect to the ingested compound and compounds structurally related to the ingested compound. It has been suggested that the phenomenon of oral tolerance induction may be adapted as a method of pretreatment of autoimmune disease. PCT publication WO 95/10301 describes the use of orally ingested cholera toxin conjugates to reduce a delayed hypersensitivity reaction and control experimental autoimmune encephalitis in mouse models.

The invention involves the use of one or more different collagens and/or collagen derivatives to induce oral tolerance in a specific population of immune cells to provide a method of treating immune system mediated diseases. The methods of using various collagens and/or collagen derivatives to treat immune system mediated diseases and composition for use of such methods are provided herein. The methods of the invention involve oral administration of a collagen or collagens found in a specific tissue to induce suppression (immunological tolerance) of inflammation against the tissue (or tissues) from which the collagen is found to occur naturally. The correspondence between a specific collagen type and its (eg, tissue type) can be found in various references, including Fukai et al., Methods of Enzymology, 245:3-28 (1994). Collagen (and collagen derivative) containing compositions for use in such methods are also described. The invention also provides many different types of mucosa-binding collagen conjugates and methods of their use in the treatment of immune system-mediated diseases.
A feature of the invention is to use minor collagens, i.e. comparable rare collagens found naturally associated with the main collagen, to induce immune tolerance to reduce the severity of an unwanted immune response, thereby providing the basis of a treatment of arthritis and related immune systems -mediated diseases; the invention also describes the use of the main collagen to induce immune tolerance. Cartilage contains different species of collagen that provide the main extracellular framework for surrounding tissues. Data that have been found suggest that type II collagen, a major collagen, may have the capacity to suppress inflammation in cartilage. The present invention is partly based on the realization that minor collagen(s) which naturally occur in combination with major collagen(s) may be important in inducing immune tolerance which is useful in the treatment of immune system-mediated diseases such as rheumatoid arthritis. A feature of the invention is to use minor collagens, alone or in combination with the main collagen, to induce immune tolerance which results in the alleviation of the disease process in several immune system mediated diseases.

The above-mentioned collagen types can also be obtained by recombinant expression techniques, as has been generally described in US Patent No. 5,405,757, PCT Published Patent Applications WO 93/07889 and WO 94/16570 and related patents and applications.
Another feature of the invention is to provide mucosa-binding collagen conjugates. The topical mucosa-binding collagen conjugates comprise a collagen component and a mucosa-binding component. The two components can be bound directly to each other or can be bound to each other through one or more intermediate binding molecules. Mucobinding collagen conjugates can be used to treat numerous immune system-mediated diseases. The treatment is carried out by inducing immune tolerance to one or more epitopes on the collagen component of mucosa-binding collagen conjugates.
The collagen component in the present mucosa-binding collagen conjugates may comprise one or more collagen or collagen derivative molecules. The collagen molecules of the collagen components of mucosa-binding collagen conjugates may be the same or different from each other. The collagen/collagen derivative molecules of the collagen component can be bound to each other, either directly or through binding intermediates. Alternatively, the collagen/collagen derivatives of the collagen components are not necessarily bound to each other, but can be bound directly to the mucosa-binding component of the present mucosa-binding collagen conjugates.
Mucobinding component comprises one or more molecules that have the ability to specifically bind to mucosal cells in a patient for treatment. Numerous different molecules can serve as the mucosa-binding component. These mucobinding molecules can be derived from the mucobinding structures of bacterial toxins, bacterial fimbriae, viral attachment proteins and plant lectins. Use of mucosa-binding structures from bacterial toxins is preferred. Particularly preferred are the B-subunits of cholera toxin and the B-subunit of heat-labile entertoxin from E. coli. The B-subunits of cholera toxin and the B-subunits of heat-labile enterotoxin from E. coli have the property that they bind specifically to ganglioside GM, in mucosal cells. When the toxins are used as mucosa binding structures, the toxins are preferably modified to substantially remove the destructive cytotoxic properties of the toxin. Inactivation of cytotoxic properties can be carried out in a number of ways which are well known in the art, such as e.g. includes denaturation, mutation and the like. Other molecules that specifically bind to ganglioside GM can be used as the mucosa-binding component of mucosa-binding collagen conjugates in the invention. Mucobinding molecules can be covalently bound to each other, either directly or indirectly, by means of an intermediate binding molecule(s). Alternatively, mucobinding molecules forming the mucobinding component can be associated with each other by means of intermolecular forces of attraction. The B subunits of cholera toxin form e.g. a 5-molecule "ring" structure with possibilities for connection. Examples of suitable toxin molecules include subunits S2, S3, S4 and/or S5 of Bordatella pertussis toxin, diphtheria toxin, diphtheria toxin B fragment, shiga toxin, shiga-like toxins, shiga toxin B subunit, shiga-like toxin B subunit, cholera toxin, E. coli heat-labile toxin. Examples of suitable bacterial fimbriae include E. coli K88, K99, 987P, F41, CF A/I, CFA/II, (CS1, CS2 and/or CS3), CF A/IV (CS4, CS5 and/or CS6), P fimbria, Vibrio cholera toxin co-regulated pili (TCP), mucus sensitive hemagglutinin (MSHA), fucose-sensitive hemagglutinin (FSITH), B. pertussis filamentous hemagglutinin and the like. Examples of suitable viral attachment proteins include influenza hemagglutinin, Sendai Virus hemagglutinin. Examples of suitable lectins include both plant lectins and animal lectins, soluble lactose-binding lectins, selectin, collectin, helix pomatin hemagglutinin, concanavalin A, wheat germ aggutinin, phytohemagglutinin, aurin ricin. Immunoglobulins specific for mucosal cell antigens can also be used as mucosal binding components.
The separate collagen compound and mucobinding component of mucobinding components can be bonded to each other in numerous ways. The two components can be linked together by chemical cross-linking agents such as N-succinimidyl (3-(2-pyridyl-dithio)propionate, dimethyl-3,3'-dithiobispropionimidate, 2-iminothiolan, N-succinimidyl-(4-azidophenyl )-1,3-dithioprionate, ethyl-4-azidophenyl-1,4-dithiobutyrimidate, diethylmalonimidate, 2-iminothiolan, N,N'-p-phenylenedimaleimide, and the like Alternatively, mucosa-binding collagen conjugates of the invention can be simple polypeptides which are fusion proteins formed between the collagen component and mucosa-binding molecular component. These fusion proteins can be produced by conventional in vitro genetic engineering techniques. Further features of the invention are to provide polynucleotide sequences that code for mucosa-binding collagen conjugate fusion proteins. A further feature of the invention is to provide host cells for the recombinant production of mucosa-binding collagen conjugate fusion proteins .
Nati. Acad. Sei. (USA), 81:7752-7756 (1984). Mucobinding molecular components can be obtained by purification from their naturally occurring organismal sources. Alternatively, mucosa-binding molecular components can be produced by recombinant DNA techniques. Methods for the purification of mucosa-binding molecules, or alternatively the recombinant production of such molecules which are suitable for use in the invention, are well known to persons with knowledge in the field. Recombinant production of cholera toxin B subunit has e.g. been described in Sanchez and Holmgren, Proe. Nati. Acad. Pollock. (USA), 86:481-485 (1989) and production of the B subunit of E. coli heat labile enterotoxin has been described in Hirst et al., Proe. Nati. Acad. Pollock. (USA), 81:7752-7756 (1984).

Purified collagens for use in the methods and compositions of the present invention can be isolated from animal or human tissue; the use of human collagens in the present compositions and methods is preferred when the subject to be treated is a human. In addition to obtaining collagens for use in the relevant compositions and methods from natural sources, the collagen for use in methods and compositions can be produced by recombinant DNA technology. A description of how to produce type II by recombinant DNA technology can be found, among other things, in PCT-published patent applications WO 93/07889 and WO94/16570, which are incorporated herein by reference. The recombinant production techniques described in these PCT publications can be quickly adapted to produce many different types of collagen, human or otherwise, by recombinant DNA techniques. Human collagens produced by recombinant DNA technology are preferably used in the compositions of the invention. Recombinant DNA technology avoids ethical and financial problems associated with obtaining sufficient amounts of human tissue.
In addition to using collagen obtained directly from natural sources, the method and compositions in the invention can include many different types of collagen derivatives. The collagen derivatives may differ from naturally occurring collagens in several aspects. The collagen derivatives can be non-glycosylated or glycosylated and different from naturally-occurring collagens. Desired glycosylation patterns can be produced by a wide variety of methods, including direct chemical modification and enzymatically catalyzed glycosylation and deglycosylation reactions. Desired glycosylation patterns can also be produced by inhibiting or omitting an enzyme that is necessary for the production of naturally occurring glycosylation patterns found on collagens. Collagen derivatives have a glycosylation pattern that differs from a corresponding naturally-occurring collagen, but are otherwise essentially the same in structure (with the exception that the amount of proline and/or lysine residues that have been hydroxylated) are referred to herein as "variably glycosylated collagens". . Collagen derivatives also include various fragments of naturally occurring collagens. Such collagen derivatives also include various fragments of naturally occurring collagens. Such collagen fragments can be produced by, among other methods, chemical or enzymatic cleavage of one or more peptide bands. The collagen derivative may also contain one or more amino acid residues which are different compared to corresponding amino acid residue positions in naturally occurring collagen. Collagen derivatives containing such amino acid substitutions can be produced by numerous methods including genetic engineering techniques and by in vitro peptide synthesis. Additional collagen derivatives can be produced by varying the amount of hydroxylysines and/or hydroxyprolines present in a given molecule, by the varied expression of lysine hydroxylases, and/or proline hydroxylases, where hydroxylase genes (recombinant or otherwise) and are expressed in a host cell for expression of recombinant collagen, or derivatives thereof. Collagen derivatives for use in the composition and method of the invention have the ability to reduce unwanted immune reaction in immune system-mediated disease that is relevant when administered to a patient either alone or as parts of a composition of the invention, including compositions comprising numerous collagen and/ or collagen derivatives. Collagen derivatives suitable for use in these methods and compositions of the invention for the treatment of rheumatoid arthritis can be determined by using in vitro tests of T cells from rheumatoid arthritis patients, where the tests can determine whether a given collagen derivative can stimulate suppressor T cells , induce clonal anergy, or include other forms of immune tolerance. Procedures for measuring antigen tolerance induction are well known to persons with knowledge in the field and can e.g. be found in Paul, W. E., author, Fundamental Immunology, 2nd Ed., Raven Press: New York (1994). Collagen derivatives for use in the treatment of other immune system-mediated disorders can be determined for suitable use in the treatment of a given immune system-mediated disease by in vitro tests on immune system cells isolated from patients suffering from the disease in question.
A collagen molecule, collagen derivative or mucosa-binding collagen derivative which is suitable for use in the method of the invention comprises at least one tolerance-inducing epitode. The term "tolerance-inducing epitope" as used herein refers to epitopes on collagen that have the property of inducing immune tolerance with respect to a pathogenic immune response in an immune system-mediated disease, independent of other parts of the collagen molecule that the "tolerance-inducing epitope" was derived from. Individual collagen molecules may comprise one or more tolerance-inducing epitopes. In addition, the tolerance-inducing epitope or epitopes on a given collagen molecule may vary according to the specific immune system-mediated disease to be treated with the collagen.
The invention also provides collagen derivatives consisting of amino acid residue sequences comprising one or more collagen tolerance-inducing epitopes; such novel polypeptides are referred to herein as "tolerance epitope polypeptides". Tolerance epitope polypeptides may comprise numerous identical tolerance-inducing epitopes. Tolerance epitope polypeptides in the invention may also comprise one or more non-identical tolerance-inducing epitopes derived from the same or different collagen molecules. Embodiments of the present tolerance epitope polypeptides include polypeptides in which the various epitopes are arranged in a regulated repeating pattern and the polypeptides in which the various tolerance-inducing epitopes are arranged randomly. Tolerance epitope polypeptides comprise many tolerance-inducing epitopes; while the tolerance epitope polypeptides may in fact be of any size, a person skilled in the art will appreciate that certain technical problems arise when synthesizing and using very large polypeptides, e.g. solubility, stability, etc. The tolerance epitope polypeptides in the invention preferably comprise 500 tolerance-inducing epitopes, more preferably in the range from 250 tolerance-inducing epitopes and further preferred in the range from 50 to 100 tolerance-inducing epitopes. Tolerance-inducing epitopes can be glycosylated or non-glycosylated, depending on whether the carbohydrate matrix is considered to be part of a tolerance-inducing epitope. Tolerance epitope polypeptides can be produced by conventional genetic engineering techniques such as those used e.g. is described in Goeddel Gene Expression Technology, Methods in Enzymology Volume 185, Academic Press (1991). Tolerance epitope polypeptides may comprise one or more non-collagen-derived amino acid residue sequences, whereby the collagen-derived tolerance-inducing epitope region on the tolerance epitope polypeptide serves as a carrier for the non-collagen-derived amino acid sequence so that the immune tolerance can also be induced against non-collagen-derived amino acid residue sequences. In other words, some embodiments of the present tolerance epitope polypeptides are polypeptides that act as "carriers" for inducing immune tolerance against non-collagen proteins that may be involved in the pathogenesis of immune system-mediated diseases. The present invention provides e.g. provide tolerance epitope polypeptides comprising regions of myelin basic protein that have been shown to be important in the development of pathogen immune responses in multiple sclerosis and multiple tolerance-inducing epitopes. The polynucleotide sequence encoding tolerance epitope polypeptides can be prepared, among other methods, by in vitro polynucleotide synthesis and manipulation of the previously isolated collagen-encoding polynucleotides.
Tolerance epitope polypeptides may be designed for the treatment of specific immune system-mediated diseases in accordance with information provided in Table 1. Tolerance epitope polypeptides may comprise tolerance-inducing epitopes derived from collagen or collagen found in a specific tissue to induce suppression (immunological tolerance). of inflammation against the tissue (or tissues) from which collagen is found to occur in nature. Tolerance epitope polypeptides which thus e.g. comprising tolerance-inducing epitopes of type II, type IX and type XI collagen may be used instead of (or in addition to) a mixture of type II, type IX and type XI collagens.
Visse underpopulasjoner av lymfocytter i en person som lider av (eller sannsynligvis utvikler) en spesifikk immunsystem-formidlet sykdom har reseptorer som har evne til Tolerance-inducing epitopes for a given collagen and a given immune system-mediated disease can be quickly determined by a person skilled in the field of immunology. Immune system-mediated diseases that can be treated by the present method include, but are not limited to, diseases listed in Table 1. The relevant methods of the invention comprise the steps of administering an effective amount of a composition of the invention, e.g. collagens, collagen derivatives, or mucosa-binding collagen conjugates. Preferred compositions for use in the treatment of specific immune system mediated diseases are set forth in Table 1, as described in the preceding section. In a preferred embodiment of the method, the composition administered to the subject comprises variable glycosylated collagens or mucosa-binding collagen conjugates, wherein the collagen component comprises variable glycosylated collagen molecules. The composition administered in the present methods is administered so that the active components, i.e. collagens and/or collagen derivatives, come into contact with lymphoid tissue in the intestinal tract, e.g. Peyer's patches or other similar sites, so that immune tolerance is induced. Such administration can be carried out by means of many possible methods, using formulations that include the relevant compositions designed for oral administration, i.e. the active components are not destroyed or inactivated in the mouth, stomach or other parts of the digestive system before in contact with appropriate intestinal lymphoid tissue. The treatment methods of the invention may also include the steps of administering additional pharmaceutical compounds for the treatment of immune system-mediated diseases, such as anti-inflammatory agents and the like.
The dosage at which the present composition is administered can vary within a wide range and will depend on various factors such as e.g. severity of the inflammation, age of the patient, etc., and must be adjusted individually. As a possible range for the amount of collagen(s) and/or collagen(s) derivatives that can be administered per day can be in the range from approx. 0.001 mg to approx. 200 mg. The amount of collagen and/or collagen derivatives administered is preferably low, thereby favoring the induction of immune tolerance by suppression rather than clonal anergy. The pharmaceutical compositions containing collagen(s) and/or collagen(s) derivatives can conveniently be formulated so that they provide doses within these ranges either as single dosage units or as multiple dosage units.
Optimal dosage of tolerance inducing compositions for use in the methods of the invention will vary in accordance with a number of factors. The terms "dosage" and "dose" as used herein may, unless otherwise indicated, refer not only to a single administration of the composition but may also be used to refer to the total amount of a given pharmaceutical composition administered over a selected period of time and involves many individual administrations. Factors affecting optimal dosage include the choice of collagen molecule or molecules (and/or collagen derivatives) administered to the patient, specific mucosa binding molecules selected, age of the patient, severity of the disease, other diseases that may be present in the patient, inert components of the formulation, excipients and the like. There can be considerable variation in the range of dosage that is effective in treating a given immune disorder. Different dosages of the same pharmaceutical composition can produce the desired tolerance effect by means of different mechanisms. Although implementation of the invention is not dependent on a particular theory of implementation, a person skilled in the art will better understand the invention and provide additional embodiments by assuming that there may be two primary mechanisms by which oral tolerance is mediated. Oral tolerance may be mediated by active cellular suppression in which regulatory T cells suppress the activation and proliferation of lymphocytes specific for the tolerized antigen. Other mechanisms of oral tolerance induction are clonal anergy in which T lymphocytes that have a suitable receptor are rendered unresponsive. In general, active suppression tolerances are favored with "low" doses of a tolerance antigen and clonal anergy is favored with corresponding "high" doses of the same tolerance antigen. An overview of principles and techniques for oral tolerance induction can be found in Weiner et al., Annual Review of Immunology: 1994, 809-835, Annual Reviews.
The present composition can be formulated as pharmaceutical compositions so that they are adapted to certain types of administration to mucosal surfaces, e.g. oral, topical and inhalation. The preferred form of formulation for oral administration is in a form where collagen and/or collagen derivatives in the composition contact small intestinal lymphoid tissue, e.g. Peyer's patches. Compositions in the invention can be administered topically, orally, intranasally, by injection or by inhalation in the form of pharmaceutical compositions comprising collagen(s) and/or collagen(s) derivatives in the form of the original compound or optionally in the form of a pharmaceutically acceptable salt therein, associated with a pharmaceutically acceptable carrier which may be a solid, semi-solid or liquid diluent or a digestible capsule, and such preparations comprise a further feature of the invention. Collagen(s) and/or collagen(s) derivatives and mucosa-binding collagen conjugates can also be used with the carrier material. Examples of pharmaceutical preparations include tablets, drops such as nasal drops, preparations for topical application such as ointments, gels, creams and suspensions, aerosols for inhalation, nasal sprays, liposomes, etc. Generally, collagen(s) and/or collagen(s) ) derivatives comprise between 0.05 and 99%, or 0.1 and 99% by weight of the preparation, e.g. between 0.5 and 20% for preparations intended for injection and between 0.1 and 50% for preparations intended for oral administration.
To produce pharmaceutical preparations in this form of the dosage unit for oral administration containing a compound of the invention, the active ingredient can be mixed with a solid, a powdered carrier, e.g. lactose, sucrose, sorbitol, mannitol, a stiffener such as potato starch, corn starch, amylopectin, laminaria powder or citrus pulp powder, a cellulose derivative or gelatin and may also include lubricants such as magnesium or calcium stearate or a Carbowax or other polyethylene glycol waxes and are compressed to form tablets Elelr cores for dragons. If dragees are necessary, the cores can e.g. be coated with concentrated sugar solutions which may contain gum arabic, talc and/or titanium dioxide, or alternatively with a film-forming agent dissolved in simple volatile organic solvents or mixtures of organic solvents. Dyes can be added to these coatings, e.g. to distinguish between different contents of active substance. For the preparation of soft gelatin capsules consisting of gelatin, and e.g. glycerol as a softening agent, or a similar closed capsule, the active substance can be mixed with a Carbowax or a suitable oil such as e.g. sesame oil, olive oil, or araki oil. Hard gelatin capsules may contain granules of the active substance with a solid, powdered carriers such as lactose, sucrose, sorbitol, mannitol, starches (eg potato starch, corn starch or amylopectin), cellulose derivatives or gelatin, and may also include magnesium stearate or stearic acid as lubricants.
The composition of the invention may also be formulated to provide a sustained release. By using several layers of active drug, separated by slowly dissolving coatings, sustained release can be achieved. Another way of producing sustained-release tablets is to divide the dose of active drug into granules with coatings of different thickness and press the granules into tablets together with the carrier substance. Collagen(s) and/or collagen(s) derivatives and mucosa-binding collagen conjugates can also be incorporated into slowly dissolving tablets that are made, e.g. of fat and wax substances or evenly distributed in a tablet of an insoluble substance such as a physiologically inert plastic substance.
In order to obtain dosage units of oral preparations -- tablets, capsules, etc. -- which are designed to prevent the release and possible decomposition of the active substance in the stomach acid, the tablets, dragees, etc. can be enteric coated, i.e. provided with a layer of stomach acid-resistant enteric film or coating which has such properties that it did not dissolve in the acidic pH values of stomach acid. The active substance will thus not be released until the preparation reaches the small intestine. As examples of such known enteric coatings, cellulose acetate phthalate, hydroxypropyl methylcellulose phthalates such as those sold under the brand names HP 55 and HP 50, and Edragit L and Eudragit S can be mentioned.
Liquid preparations for oral use can be in the form of elixirs, syrups or suspensions, e.g. solutions containing from approx. 0.1 to 20% by weight of active substance, sugar and a mixture of ethanol, water glycerol, propylene glycerol and optionally aroma, saccharin and/or carboxymethyl cellulose as a dispersant.
The term "purified" as used herein with reference to collagens denotes that the indicated molecules are present in the substantial absence of other biological macromolecules, e.g. polynucleotides, proteins and the like. The term "purified" as used herein preferably means at least 95% by weight, more preferably at least 99.8% by weight of the specified biological macromolecules present (but water, buffers, and other smaller molecules, especially molecules that have a molecular weight less than 1000 daltons may be present). The term "isolated" as used herein refers to protein molecule separated not only from other proteins present in the natural protein source, but also from other proteins, and preferably refers to a protein found in the presence of (if any) only a solvent, buffer or ion or other component normally present in a solution thereof. The terms "isolated" and "purified" do not include proteins present in their natural source.

Hver mus mottok 100 ug protein subkutant i halen. Groups of 10 DBA/1 mice were immunized with either human type II collagen (CU), mammalian cell derived recombinant type II collagen (rCII+), or insect cell derived recombinant type II collagen (rCII-). In each case, the collagen was dissolved in 0.01M acetic acid and then emulsified in a 1 to 1 ratio with complete Freud's adjuvant prior to immunization. Each mouse received 100 µg of protein subcutaneously in the tail. The two groups received two doses of 100 µg of rCII+ or rCII-, thus receiving a total of 200 µg of rCII+ or rCII-. In each case 200 µg total dose was administered, the first 100 µg dose of protein was administered as described above. The second 100 µg dose of protein was emulsified in incomplete Freud's adjuvant and administered three weeks after administration of the first dose.
Incidence of arthritis was measured and reported, as indicated in Table 2 at six weeks (post immunization).

kollagen etter intravenøs administrering av kollagencollagen after intravenous administration of collagen
Ovalbumin, human type II collagen (CU), mammalian cell derived recombinant type II collagen (rCII+), or insect cell derived recombinant type II collagen (rCII-) were administered intravenously to groups of 10 DBA/1 mice. The collagen (CU, rCII+ and rCII-) was dissolved in 0.001M acetic acid and dialyzed against PBS prior to administration. Either 33 µg or 33 µg of protein was administered daily for 3 days so that a total of either 100 µg or 1000 µg of protein was administered to each test mouse. Four days after administration of the last dose, the mice were then immunized with CU. Incidence of arthritis at six (6) weeks of immunization is shown in table 3.
The antibodies represent mean levels/group (expressed in units) against native human type II collagen using sera pooled four weeks (4) after immunization. The statistics are reported using Student's T test.

kollagen etter oral administrering av kollagencollagen after oral administration of collagen
Grupper av 10 til tolv DBA/1 mus ble oralt administrert enten ovalbumin, human type II kollagen (CU), pattedyrcelle avledet rekombinant type II kollagen (rCII+), eller Groups of 10 to twelve DBA/1 mice were orally administered either ovalbumin, human type II collagen (CU), mammalian cell derived recombinant type II collagen (rCII+), or
insektcelle avledet rekombinant type II kollagen (rCII-). Kollagenet ble oppløst i 0,01M eddiksyre og administrert fire (4) ganger pr. uke i to uker i totalt åtte doser. Enten 10 ug eller 1000 ug ble administrert daglig slik at mus mottok totalt enten 80 ug eller 800 ug insect cell derived recombinant type II collagen (rCII-). The collagen was dissolved in 0.01M acetic acid and administered four (4) times per day. week for two weeks in a total of eight doses. Either 10 µg or 1000 µg was administered daily so that mice received a total of either 80 µg or 800 µg
protein. Three (3) days after taking the last dose, the mice were then immunized with OI. Table 4 gives the incidence of arthritis at five weeks of immunization.
The reported statistical variance was calculated using a Fisher's exact test. Antibodies represent average levels per group (expressed in units) against native human II collagen using sera collected for four weeks after immunization and statistics were reported using Studenf's T test.
6.4. Human plateaggregeringsstudie6.4. Human platelet aggregation study
Type III recombinant collagen and human types I and III collagen were tested in blood samples from four donors and examined using routine platelet aggregation assay. All collagen types showed platelet aggregation activity. More specifically, the percent aggregation of 10 µg of recombinant type III collagen is equivalent to the percent aggregation of either 0.3 µg of type III or 0.6 µg of type I collagen.
