En mutert mycobakterie valgt fra klassen bestående av nmtert M. bovis-BC, mutert M. tuberculosis og mutert M. lepra er beskrevet. Mutasjon av M.bovls-BCG, M.tubercu- losis eller M. lepra blir fortrinnsvis utført ved innskuddsmutasjon av et mycobakterielt gen. Innskudds- mutagenesen kan for eksempel bli oppnådd ved uriktig rekomblnasjon eller av et myobakterielt transposon. Slike muterte mycobakterier kan deretter bli trans- formert med en ekspressjonsvektor inneholdende et komplementgen for genet som er mutert og fortrinnsvis også et heterologt gen.
