Abstract

The antibody can be isolated from cell culture or ascites fluid in large quantities by affinity chromatography under mild conditions using the peptide bound to an immobilized substrate. The antibody has several specific uses for the isolation and characterization of protein C and as a model for the construction of Ca 2+ -dependent antibodies for the isolation of other proteins, as a diagnostic agent, and as a therapeutic agent to prevent activation of protein C. Protein C produced naturally or produced by expression of the recombinant gene. The benefits of the antibody in the purification of protein C include the specificity of protein C and not of activated protein C, and the unique Ca 2+ peptide binding specificity that allows the binding site to be protected when immobilized on the chromatographic substrate. In vivo, the antibody has been demonstrated to inhibit tumor growth. The antibody can also be used to promote coagulation in patients who have high levels of factor VTII inhibitors.
