Calcium chloride can be used in the concentration range 100-400 mg per liter and magnesium chloride can be used in the range of 200-400 mg per liter with good results. (4) Potassium chloride was used in the concentration range 220-400 mg per liter with good results. (5) Sodium diphosphate was used in the concentration range 100-580 mg per liter with good results. (6) Sodium bicarbonate was used in the concentration range 2,900-4,200 mg per liter with good results. A minimum of 2,900 mg per liters was found to be necessary to prevent a decrease in pH with storage over 7 days. (7) A 2.5-7.5$ carbon dioxide atmosphere, depending on the amount of sodium bicarbonate added, was successfully used to maintain a constant pH value. (8) The addition of glucose (dextrose) was essential for maintaining good in vitro quality of the platelets, which is evident from the data in table 12. Dextrose was used in the concentration range 3,000-7,500 mg per liter with good results. (9) The pH value of the platelet storage medium was maintained in the range 6.8-7.4 measured at 22°C. A pH value above 7.4 resulted in clumping of the platelets, while a pH value below 6.8 caused swelling and loss of disc-shaped morphology.

The platelet concentrate was processed for storage for a 7 day period at 22°C either in CPD plasma or in the platelet storage medium using current licensed methods.

The percent recoveries and survivals were determined using the gamma function multiple-hit program. Paired t-tests were used to detect significant differences. In vivo platelet viability was evaluated by hypotonic shock response and degree of shape change with ADP. The results for the percent in vivo recovery and survival are shown in Tables 17 and 18, respectively.
