US patent no. 3 522 34 5 concerns e.g. non-thrombogenic microcapsules which can be used when produced around enzymes or detoxification agents and placed in an extracorporeal anastomosis, to introduce oxygen, drugs, enzymatic substances and the like into the blood stream at adjustable rates.
I "Artificial Cells" av Thomas M.S. Chang (Charles C. In "Artificial Cells" by Thomas M.S. Chang (Charles C.
Thomas, Springfield, 111, 1972) a method for encapsulating biologically active materials in a semipermeable membrane is presented. The technique involves dissolving the material to be encapsulated and a monomer in water, and producing an emulsion with water as the discontinuous phase. When another monomer capable of polymerizing with the first is dissolved in the continuous phase of the emulsion, polymerization takes place at the interface between the two phases, and a membrane forms around the droplets of the solution, which are normally of colloidal size .
This method has disadvantages and limitations that limit its potentially wide application and commercial success. Since many biologically active substances, e.g. enzymes, are extremely labile, do they result in relatively harsh reaction conditions? which is necessary for polymerization, often in a very low yield of usable encapsulated material. The encapsulation of even relatively hardy enzymes, such as urease, is characterized by yields that are at best between 35 and 40%. During one or more steps in the method, much of the encapsulated material is denatured.
US patent no. 3,522,34 6 relates to microcapsules of regulated permeability. The method includes encapsulation of aqueous mixtures in membranes of adjustable size, thickness and permeability. Droplets of an aqueous mixture are dispersed in an organic, liquid medium which is not miscible with them and a component which is soluble in the organic liquid and which is capable of reacting with a component of the hydrophilic mixture is added to the dispersion, thereby obtaining a macromolecular membrane was formed by phase boundary coacervation, polymerization or condensation on the surface of the droplets. A polymer, a condensation product or a component thereof is usually added to the organic liquid and the membrane is formed around the droplets by reaction of the polymer, the condensation product or the component with a component in the dispersed droplets, which may be a precipitation, a condensation or polymerization catalyst, or a compound of the eventual condensation product. The effective pore size of the lean encapsulating membrane is a function not only of the membrane composition, but also of the thickness of the membrane and is chosen according to the intended use of the capsules. By suitable choice of operating conditions, the pore size can be varied, but the reaction condition which was necessary to ensure sufficiently strong, albeit selectively permeable membranes, is in any case difficult to regulate and in some cases unknown. The process also provides capsules that typically have membranes of random uniformity and porosity; and many are unusable for their intended purpose.
Det er derfor et mål ved foreliggende oppfinnelse å skaffe tilveie en fremgangsmåte hvorved labilt, biologisk materiale, f. eks. forskjellige enzymer og mange kjemisk virksomme substanser, It is therefore an aim of the present invention to provide a method whereby labile, biological material, e.g. various enzymes and many chemically active substances,
kan innkapsles i sin aktive tilstand, og å gi et høyere utbytte av anvendbar substans når den innkapsles på denne måten. can be encapsulated in its active state, and to give a higher yield of usable substance when encapsulated in this way.
In the method according to the invention, very strong semipermeable membranes are provided around any large number of labile biological materials.
With the help of the method, enzyme-containing microcapsules can be produced, where the microcapsule membrane has a more uniform semi-permeability, so that you get capsules with smooth walls without pore defects and with such a pore size that the encapsulated material cannot escape from the inside of the capsule , but can easily interact with substances in the environment where the capsules are placed.
Furthermore, with the help of the method, a number of different, chemically active substances can be encapsulated in a high-quality semipermeable membrane by using a number of different polymer systems and different interfacial polymerization techniques. The method has some variable parameters, which can be selected to be adapted to a particular substance to be encapsulated, vary the properties of the capsule membrane and maximize the yield of usable, encapsulated material.
Furthermore, with the method, a high concentration of hemoglobin can be encapsulated in a capsule which is permeable to oxygen, carbon dioxide and other small molecules.
It is also possible, with the help of the method, to regulate the pore size of the capsule membrane to a significant extent, as the method has wide application to a number of different encapsulation techniques.
Ved fremgangsmåten i henhold til oppfinnelsen fåes sterke By the method according to the invention, strong
semipermeable mikrokapsler med membraner som har porestørrelse som ligger innenfor et hvilket som helst av en rekke størrelsesområder, og således er selektivt permeable overfor molekyler som har dimen-sjoner som er mindre enn den øvre grense av det valgte område. semipermeable microcapsules with membranes having pore sizes that lie within any of a number of size ranges, and thus are selectively permeable to molecules having dimensions smaller than the upper limit of the selected range.
Grenseflatepolymerisasjonen for dannelse av den semipermeable membran omfatter følgende trinn: The interfacial polymerization to form the semipermeable membrane includes the following steps:
A) oppløsning av en diaminmonomer og nevnte kjernemateriale A) dissolution of a diamine monomer and said core material
i en vannfase; in an aqueous phase;
B) dannelse av en emulsjon i hvilken vannfasen fra trinn A B) formation of an emulsion in which the water phase from step A
er den diskontinuerlige fase, idet diaminet er svakt løselig i den kontinuerlige fase i emulsjonen og den kontinuerlige fase omfatter et hydrofobt løsningsmiddel; is the discontinuous phase, the diamine being slightly soluble in the continuous phase in the emulsion and the continuous phase comprising a hydrophobic solvent;
C) dannelse av en sterkt permeabel kapselmembran rundt C) formation of a highly permeable capsular membrane around
delene av den diskontinuerlige fase ved omsetning av diaminet med et disyrehalogenid som inneholdes i nevnte kontinuerlige fase, idet disyrehalogenidet er i stand til å danne en polymer ned nevnte diaminmonomer ved en polykondensasjonsreaksjon, og nevnte disyrehalogenid er sterkt løselig i nevnte hydrofobe løsningsmiddel; D) stansing av polymerisasjonsreaksjonen ved separering av det hydrofobe løsningsmiddel fra emulsjonen. the parts of the discontinuous phase by reacting the diamine with a diacid halide contained in said continuous phase, the diacid halide being able to form a polymer down said diamine monomer by a polycondensation reaction, and said diacid halide being highly soluble in said hydrophobic solvent; D) stopping the polymerization reaction by separating the hydrophobic solvent from the emulsion.
The method according to the invention is characterized in that the capsules produced in the above-mentioned step C) are subjected to the following steps: E) resuspension of the capsules in another hydrophobic liquid, the diamine monomer being practically insoluble in this; F) reacting a further amount of said diacid halide dissolved in said other hydrophobic liquid with said diamine monomer to effect further polymerization on said capsule membranes, so as to strengthen them, and to make them impermeable to the core material; and
G) stansing av polymerisasjonen. G) stopping the polymerization.
In more detail, the method according to the invention is carried out as follows: First, a hydrophilic solution is prepared, preferably with water, by dissolving the material to be encapsulated in water together with the diamine monomer. Since many monomer solutions have a pH value that would denature many labile biological substances, the monomer solution must often be buffered to a pH range between 5 and 9 before the labile biological material is added.
FYi hydrophobic, organic liquid having the following properties is then produced. First, the solubility of the other monomer to be used in the polymerization, namely a diacid halide, should be very high in the hydrophobic, organic liquid. Second, the hydrophobic liquid should have little affinity for the first monomer. Third, the hydrophobic solvent should be such a solvent that the hydrophilic solvent, when added, will form a good emulsion.
The two solvents are then combined and emulsified. The hydrophilic emulsifying agent is the discontinuous phase* Emulsification is accomplished by any of a number of well-known techniques, usually by means of an emulsifying agent. The size of the droplets that are produced determines the size of the microcapsules that are formed.
Mikrokapselmembranen dannes tvers over grenseflatesonen idet polymerisasjonen for-løper. When the desired droplet size range has been reached, the second monomer is added to the emulsion, and polymerization or condensation takes place at the interface of the two-phase system. Since the first monomer, which is dissolved primarily in the discontinuous phase, is poorly soluble in the continuous, hydrophobic, organic phase, some diffusion into the continuous phase takes place. The microcapsule membrane is formed across the interface zone as the polymerization proceeds. The membrane that forms limits further diffusion of the first monomer into the continuous phase, and the membrane properties are highly dependent on the detailed sequence of random collisions between the monomers.
The membranes produced in this way are usually macroporous, and successful semipermeable membranes are only obtained under special conditions that are difficult to regulate. It is believed that the problem is due to the presence of defects in the membranes that result in macroporous holes through the membranes after the reaction is stopped and the capsules are washed. This has previously been overcome by continuing the polymerization or condensation long enough to close these large holes, and at the same time the small pores. It is therefore assumed that acceptable capsules are only obtained over a narrow range of polymer formation, in which the holes are closed and the small pores are still open.
However, it has been shown that if such untreated microcapsules are separated from the continuous phase, suspended again in a quantity of such hydrophobic liquid where the solubility of the first monomer is greatly reduced, and then - exposed again to the second monomer - find the same polymerization turnover takes place at a much lower rate over a much narrower interfacial zone that lies only within the interstices of the untreated capsule membrane. This results in "patching" of the macroporous defects in the capsule wall, reduction of the pore size and strengthening of the membranes.
It is believed that this second polymerization reaction selectively bridges across the large pores where the first monomer diffuses close to the continuous phase, and that this "patching" takes place mainly without any significant increase in capsule wall thickness. In contrast, the continuous application of the first solvent increases the wall thickness and reduces the permeability of most membranes when achieving closure of the holes.
When carrying out the method according to the invention, it has been shown that this secondary polymerization technique can be used in a number of known encapsulation methods to produce semipermeable capsules and to regulate the pore size of these.
According to an embodiment of this two-stage polymerization, the amount of the second monomer in the continuous phase is regulated which, with conventional methods, would rapidly denature labile biological materials to be encapsulated. The technique to avoid such denaturation includes regulating the relative concentration of the solutes in the continuous and discontinuous phase so that the concentration of the other monomer in the continuous phase is kept lower than optimal values, and the solutes in the discontinuous phase are present in high concentrations. In this situation, the diffusion of the other monomer into the discontinuous phase is inhibited, and the denaturation of the labile biological material is minimized, but a macroporous, poorly produced and clearly unsatisfactory capsule is obtained. In the second polymerization step, however, the concentration of the second monomer in the continuous phase of the resuspension can be increased (since the encapsulated material is now protected by the untreated capsule membrane) to strengthen the capsule membranes and make them semipermeable.
Ifølge en utførelsesform av oppfinnelsen tilsettes According to one embodiment of the invention is added
Denne teknikk er også ganske nyttig for systemet hvor hydrolyse av den valgte annen monomer er en konkurrerende reaksjon med polymerisasjonen. the other monomer to the emulsion in the first polymerization step, not all at once to initiate the polymerization, but rather in portions, at specific time intervals during the polymerization. This technique maintains the concentration of the second monomer at the interface at a more constant optimal level throughout the formation of the first rough membrane and thus inhibits diffusion of the second monomer into the discontinuous phase and increases the yield of usable, labile, biological material. This technique is also quite useful for the system where hydrolysis of the selected other monomer is a competing reaction with the polymerization.
As mentioned, the first monomer is a diamine and the second monomer is a diacid halide; the initial hydrophobic solvent is a solution containing 80% cyclohexane and 20% chloroform; and in the resuspension the hydrophobic liquid is pure cyclohexane. The diamine is mainly insoluble in cyclohexane and slightly soluble in chloroform.
When encapsulating hemoglobin, it has been shown that if the red blood cells are dehydrated when exposed to hypertonic saline, a hemoglobin solution with concentrations up to 30 g/dl can be produced, and this high salt concentration inhibits the diffusion of water molecules into the continuous phase where they react with molecules of the other monomer and render them unusable.
Since it is well known that many labile, biological materials rapidly denature in strongly acidic or basic environments, at all stages of the new encapsulation process, the pH value in the immediate vicinity of the material to be encapsulated should at least be kept in the range of 5 to 9. This is achieved by bubbling CO2 through the diamine solution before it is added to the labile biological material. The diamine solution, which usually has a pH value of the order of 11.5, is transferred to diamine carbonate usually at a pH value of about 8.6-8.7. If it is saturated with C02 and stored under 100% C02 gas, the solution can be brought to a pH value of approx. 8.4.
In contrast to this, it is previously known to use the weak NaHCO^ - Na^jCO^ buffer system, i.e. salts of a strong base and a weak acid where the present invention has used the weak acid itself. The resulting pH value for the previously known buffered diamine hemolysate solution was thus equal to pH 11.
A principle advantage of the described technique for immobilizing biologically and/or chemically active materials for the production of solid or pseudo-solid reactors is that the materials are encapsulated in the main mass without direct competition with pollutants for available binding sites. Thus, highly active capsules can be produced from relatively poorly cleaned materials. In addition, the active materials are protected against attack by micro-organisms which are often responsible for the inactivation or inhibition of such reactor components.
Ved fremgangsmåten i henhold til oppfinnelsen anvendes In the method according to the invention is used
Chang, res then, the hydrophobic solvent being the continuous phase. The size of the colloidal droplets determines the size of the microcapsules to be produced. The emulsification can be achieved by one of the known emulsification techniques, such as e.g. using a mixing device, and is usually carried out by adding an emulsifier. Since the size of the droplets produced by a given technique will vary within a given range, a filter can be used to separate too large capsules produced from a certain batch to reduce the difference in capsule diameter. For å gjøre utbyttet av brukbart biologisk materiale som To make the yield of usable biological material which
For å lette egnet dispersjon og adskillelse bør de to løsningsmidler ha spesifikk vekt som er noe forskjellig fra hverandre. is enclosed in the capsule as large as possible, the reaction conditions must be carefully regulated so that the normally sensitive material will not denature to any significant extent during the polymerisation. It has thus been shown that a very critical factor when it comes to achieving a high yield of usable encapsulated material is the pH in the discontinuous phase. To ensure against denaturation of labile material, the pH must be constantly regulated during manufacture, usually within the range of 5 and 9. The solvents must be chosen, especially the hydrophilic one that dissolves the encapsulated material, from those that do not rapidly denature the material to be encapsulated. The choice will be possible for the person skilled in the art. To facilitate proper dispersion and separation, the two solvents should have specific gravities that are somewhat different from each other.
Polymerizations that take place in a pH environment that is unfriendly to the particular chemically active substance that one seeks to encapsulate should not be used for this substance. However, many monomers which in solution have a natural pH capable of denaturing most labile biological materials can be buffered to the above pH range without affecting their action. Other monomers can be used without buffering, depending on their natural pH and the properties of the material that is sought to be encapsulated. For strongly alkaline monomers, a method is preferred where CC₂ gas is introduced through the solution to bring the pH into the non-denaturing range.
Ved en utførelsesform av oppfinnelsen har det In one embodiment of the invention, it has
Fremgangsmåten gir således en semipermeabel membran av høy kvalitet ved anvendelse av en annen minimal mengde proved that a very strong capsule membrane has fewer defects and, by using a lower degree of polymerization, can be produced by a two-stage polymerization. In this respect, it has been shown that the capsule membranes produced by the above-mentioned one-step process are often macroporous, uneven and vary considerably in strength and thickness. If, however, the polymerization at the interface of the emulsified system is stopped by removing the continuous phase, and the capsules are then resuspended in fresh solvent with reduced solubility of the first monomer, a further portion of the second monomer added to the resuspension will cause: further polymerization which will strengthen the membranes, cause the largest pores in the membranes to be preferentially filled and thus avoid a macroporous state. The method thus provides a semipermeable membrane of high quality using another minimal amount
polymerisasjon, idet den sistnevnte utnyttes til maksimal ytterligere fordel for å fylle membrandefektene. polymerization, the latter being utilized to maximum additional advantage to fill the membrane defects.
Ytterligere forbedring av denne totrinns-polymerisasjon Further improvement of this two-stage polymerization
Det dannes således mer polymer for å fylle de store porene. makes it possible to regulate the strength, thickness and porosity of the membranes to a greater extent. If the hydrophobic solvent which forms the continuous phase of the emulsion is chosen so that the first monomer is little soluble in it, the monomer in the droplets will diffuse little into the continuous phase. This results in a relatively thick phase boundary and a correspondingly thick and exceptionally porous membrane, which is produced when the two monomers come into contact with each other. The greater the solubility of the first monomer in the continuous phase, the more macroporous the membrane usually becomes. If, on the other hand, the first monomer is much less soluble in the continuous, hydrophobic phase, the boundary between the phases will be sharp and a thin, dense and exceptionally microporous membrane will be obtained. When the partially prepared capsules are resuspended, a different solvent is used which has a different solubility for the first monomer than the solvent used as the discontinuous phase in the first polymerization. When a macroporous membrane has been produced in the first polymerization, a solvent is used in which the first monomer is mainly insoluble in the second polymerization and which has the effect of hardening the membranes. It is assumed that in the second polymerization step, the two precursors to the polymer meet only within the weak points of the defective membrane that has been produced until then, i.e. where there is preferential diffusion of the first monomer towards the solvent. More polymer is thus formed to fill the large pores.
In another embodiment of the invention, the concentrations of the monomers in the respective solvents can be advantageously further regulated. If e.g. the other monomer, which is dissolved in the hydrophobic, continuous phase of the emulsion, has that property
Denne teknikken gir ofte en meget porøs mikro-kapsel, men i det annet polymerisasjonstrinn kan konsentrasjonen av den annen monomer uten fare økes, og dersom man anvender et egnet løsningsmiddel, vil kapslene bli styrket og gjort mikroporøse ved that it denatures the material that one seeks to encapsulate, the first monomer, which is dissolved in the discontinuous phase, can be added at a high concentration in relation to the second, in the order of magnitude 100 to 1, to prevent any migration of the denaturing monomer into the droplets of solution during the first polymerization. This technique often gives a very porous micro-capsule, but in the second polymerization step the concentration of the other monomer can be increased without danger, and if a suitable solvent is used, the capsules will be strengthened and made microporous by
selektiv brodannelse tvers over de store porene med en tynn membran som forklart ovenfor, uten å påvirke det innkapslede materiale. selective bridging across the large pores with a thin membrane as explained above, without affecting the encapsulated material.
In yet another embodiment of the invention it has been shown
Det antas at reaksjonsproduktene ved en slik hydrolyse av den annen monomer bidrar til defekter i membranen dersom den ikke er fjernet i adskillelsestrinnet. stated that the concentration of encapsulated hemoglobin can be increased significantly if red cells used to prepare hemoglobin haemolysate are washed well in hypertonic salt solution. The relatively high salt concentration in the aqueous, discontinuous phase in the emulsion will additionally facilitate the interfacial polymerization of the first monomer by limiting the solubility of water in the organic phase containing another monomer, which will usually rapidly decompose when exposed to water. It is assumed that the reaction products from such hydrolysis of the other monomer contribute to defects in the membrane if it is not removed in the separation step.
De følgende eksempler illustrerer oppfinnelsen ytterligere. The following examples further illustrate the invention.
Eksempel 1 Example 1
Red cells from expired blood bank human whole blood were separated and then washed twice in twice their volume of hypertonic saline (1.5 g/dl) by centrifugation in a refrigerated centrifuge at 2800 xg for 5 minutes. Hypertonic saline was used to remove water from the cells in order to obtain the highest possible concentration of hemoglobin after dissolution of the cells ("lysis").

pakke cellene. As much salt solution as possible was removed from the packed cells which were then dissolved by vigorous shaking with 1 ml of anhydrous ether per 10 ml packed cells. This mixture was centrifuged at 1000 xg for 20 minutes and the cell fragments and ether that appeared on top were removed. The hemoglobin concentration was examined by measuring the absorbance of 10 µl of the hemolysate in 4.0 ml of isotonic saline at a wavelength of 540 nm. The absorbance readings from all correctly prepared hemolysate solutions were between 0.7 and 0.8, which corresponds to a concentration of 28-32 g/dl. Hemoglobin concentrations greater than 30 g/dl crystallize and precipitate when stored at refrigerated temperature (4°C). The hemolysate can be used for at least one week if it is cooled.
Hexanediamine carbonate solution was prepared by bubbling carbon dioxide into a 3M solution of 1,6-hexanediamine (Eastman Kodak Co.) until a pH of 8.5 was obtained. This required approx.

50 g of DBDSA in 800 ml of deionized water containing 50 ml of 6M NaOH. The solution was filtered to remove any remaining solids and DBDSA was precipitated by adding 70 ml of a 50% v/v HC1. After the crystals had settled, as much of the purple liquid as possible was poured off, and then 500 ml of 10% v/v HCl was added. The crystals were collected in a Buchner funnel and washed three times with deionized water, then washed twice with acetone and dried. The product was white or slightly pale red crystals and was stored in a brown bottle for protection from light. This compound is used to produce a membrane copolymer which has a negative charge to prevent clumping. 10 ml of hemolysate solution containing 30 g, dl of hemoglobin was mixed with 10 ml of 3M hexanediamine carbonate solution containing 2.88 g of DBDSA. These two monomers, when mixed, form the first reactant in this example. This aqueous solution was then added to 125 ml of pre-cooled hydrophobic organic liquid prepared from 100 ml of 20% chloroform-80% cyclohexane mixture and 25 ml of an emulsifier "SPAN-85" (Sorbitan Trioleate).
Emulsification was started in a pre-cooled mixer and continued for 1-2 minutes.
Terephthaloyl chloride, the other monomer, requires preparatory purification. Hot (almost boiling) cyclohexane was saturated with terephthaloyl chloride ("practical grade" - Eastman Kodak Co.) and the solution was quickly filtered. The clear liquid was collected and allowed to cool and crystallize in a closed vessel. The crystals were collected, avoiding the influence of air as much as possible, and dried in a vacuum since the compound reacts with moisture in the air. The stock solution of terephthaloyl chloride was prepared by dissolving 10.15 g of purified terephthaloyl chloride in 95 mL of 20% CHCl 3 - 80% CgH 12 , v/v. A 0.5M solution was obtained. This solution should be stored in a tightly sealed container and can be used for at least one month before it starts to form a scum.

Den overliggende organiske væsken ble deretter fjernet ved 20% chloroform - 80% cyclohexane was then added to the emulsion with slow stirring. The actual concentration ratio of diamine/diacid halide was 128 to 1, although some variation can be tolerated. After 3 minutes, the first polymerization was finished and the suspension was centrifuged for 15-20 seconds. at 400-500 xg. The overlying organic liquid was then removed by
aspirasjon. aspiration.
Om-røringen fortsatte i 3 minutter og deretter ble omsetningen stoppet ved tilsetning av 30 ml 50% "TWEEN-20" (sorbitanmonolaurat) pufret til nøytral pH med 0,3M NaHC03- The capsules were then resuspended in another liquid consisting of 100 ml of cyclohexane and 10 ml of sorbitan trioleate. While mixing slowly, an organic solution of 7 ml of 0.5 M terephthaloyl chloride in 20% chloroform and 80% cyclohexane was added. This hydrophobic liquid contains only 1.0 to 1.5% chloroform against 20% in the first organic stage. The reduced chloroform content reduces the solubility of diamine in the solvent and a thinner membrane is obtained and the holes in the original rough patch are patched. The diamine/diacid halide concentration ratio was 50 to 1. Agitation was continued for 3 minutes and then the reaction was stopped by the addition of 30 ml of 50% "TWEEN-20" (sorbitan monolaurate) buffered to neutral pH with 0.3 M NaHCO 3 -
Note that at no time was the hemoglobin-enzyme-hemolysate solution exposed to a pH less than 5 or greater than 9. The hexane-diamine solution has a natural pH of 11.5 and a pH of approx. 11 when mixed with hemoglobin. Bubbling of (X^ through this causes the production of I^CO^ and hexanediamine carbonate. CX^ acts as a buffer and is preferred over sodium salts or weak acids. Before buffering, the sorbitan monolaurate used usually has a pH of about 4.5. Longer-term effects in this acidic environment will denature the hemoglobin and enzymes. This can be avoided by buffering the sorbitan monolaurate to a pH close to 7. The purpose of the sorbitan monolaurate is to facilitate the transfer of microcapsules into an aqueous medium such as saline. (see "Artificial Cells ", Chapter 2). It should be noted that, in this step, sorbitan monolaurate tends to react slowly with terephthaloyl chloride to produce a sticky polymer coating and, in addition, to make the microcapsules fluted. A better alternative method is to remove as much of the solvent as possible by centrifugation and then apply the above-mentioned addition of "TWEEN-20" and subsequent separation.
Halve likevektstider (t 1/2) for de oppløste stoffer av forskjellige molekylvekter er angitt i tabell I. Microcapsules produced according to this method do not leak enzymes or hemoglobin. Half-equilibrium times (t 1/2) for the solutes of different molecular weights are given in Table I.
Utbytter bestemt for 3-glukuronidase lå. jevnt rundt ca. 50%. The activity of four enzymes, lactate dehydrogenase (LDH), glutamate oxaloacetate transaminase (GOT), urease and 3-glucuronidase, was measured before and after encapsulation. LDH and GOT are found naturally in the red cell hemolysate. Urease and 3-glucuronidase were added to the hemolysate in the form of lyophilized, partially purified enzyme, which is obtained from commercial sources. The result of the comparisons for LDH, GOT and urease is given in Table II. The effect was measured in international units (IU) per liter of sample under the experimental reaction conditions. Yields determined for 3-glucuronidase lay. evenly around approx. 50%.
Table III shows a comparison of the oxygen saturation present in the hemolysate solution used to produce microcapsules with encapsulated hemoglobin, at different G^ pressures.
Table IV shows the effect of varying the amount of acid halide in the discontinuous phase at each polymerization on the porosity of microcapsules prepared in a series of experiments, the porosity being measured indirectly by observing whether or not the encapsulated material leaks through the membrane. As the table clearly shows, the concentration of the second monomer in each polymerization step is essential for good capsule formation. The person skilled in the art will have little difficulty in determining the optimum concentration ratios to be used together with other polymer systems. The capsules in Table IV were prepared as indicated above, except that the emulsification was carried out in "SPAN 85" alone before the mixed organic solvent was added. Results of other variations in manufacturing are qualitatively similar.

Økning av reaksjonstiden ved første omsetning opp til 5 minutter hadde liten tilsynelatende virkning på mikrokapslene, the first turnover was red, showing that little or no denaturation had taken place, but was leaky and poorly made. Increasing terephthaloyl chloride concentration under these conditions produced microcapsules that were brown and show hemoglobin denaturation and leakage. Increasing the reaction time at first turnover up to 5 minutes had little apparent effect on the microcapsules,
men en økning i den annen reaksjonstid til mer enn 4 minutter gjorde mikrokapslene brune og viser hemoglobin-denaturering. but an increase in the second reaction time to more than 4 minutes turned the microcapsules brown and shows hemoglobin denaturation.
Reduksjon av den ene eller den annen reaksjonstid med ett minutt Reduction of one or the other reaction time by one minute
Når etanolen tas bort er det optimale annet reaksjonsvolum normalt 3-4 ml, heller enn 7. or more resulted in leaking microcapsules. The presence of minor amounts of ethanol, which is usually used as a stabilizer for chloroform, may necessitate adjustment of optimal volumes. To achieve the greatest reproducibility and most operationally reliable capsule formation, ethanol should be removed. Chloroform used in Table IV contains some ethanol. When the ethanol is removed, the optimal second reaction volume is normally 3-4 ml, rather than 7.
Reducing the amount of "TWEEN-20" used in the separation step resulted in poor separation of microcapsules from the organic solvent. Microcapsules allowed to remain in "TWEEN 20" for longer periods (> \ hour) turned brown. Microcapsules stored in unbuffered saline having a pH of 5.5 turned brown after a few days, while those stored in a solution buffered to pH 7.4 remained red for months.
Når t er veggtykkelsen og D er diameteren, er forholdet mellom veggvolum og kapselvolum gitt ved: Further examination of the capsules produced in this example shows that they have a ratio between wall volume and capsule volume of the order of 0.028. When t is the wall thickness and D is the diameter, the ratio between wall volume and capsule volume is given by:
The capsules have an average wall thickness (t) of 700Å or 0.07Mffi and an average diameter (D) of 15Mm. Consequently, R is 6 x 0.07/15 or 0.028. Since nylon has a density approximately equal to 1, 28 mg of nylon will be required to produce solid membranes per ml capsules. The total amount of diamine present, which can be calculated from the concentration used, is 1.5 mM/ml or 174 mg/ml. Since the nylon wall is 46% diamine by weight, 12.9 mg of diamine, or approx. 7.5% of the total quantity, be necessary to produce a solid nylon wall. However, since the acid chloride is the reaction-limiting component and the diamine/acid chloride ratio is at best 50/1, the upper limit for produced nylon is in the order of 2.0% of total diamine. This suggests that the fabricated membranes are almost sponge-like with an open cell structure.
Disse kapsler er ugjennomtrengelige for hemoglobin These capsules are impermeable to hemoglobin
(mw - 68.000), men gjennomtrengelige for insulin (mw 12.000, (mw - 68,000), but permeable to insulin (mw 12,000,
A comparison of the half-lives of insulin with those of low molecular weight substances as indicated in Table I shows a non-linear relationship. It is believed that this phenomenon is due to steric properties or the charge state of the insulin molecules.
E ksempel 2 Example 2
The procedure was the same as in Example 1, except that a solution consisting of 1 mM 4,4'-diaminostilbene-2,2'-disulfonic acid (Pfaltz and Bauer, Inc.) in isotonic, pH 7.4, phosphate -buffer plus 10 parts of 3M hexanediamine carbonate added to an equivalent volume of hemolysate was used instead of diamine-DBDSA in the hemolysate solution. The manufactured capsules have the same properties as those in example 1.
Eksempel 3 Example 3
The procedure is the same as stated in Example 1, except for the following. DBDSA is not added to the hemolysate solution, and the amount of acid chloride used in both polymerization steps, i.e. 0.35 ml of acid chloride per ml hemolysate-diamine solution, is the same. The acid halide is added in equal portions at each polymerization at intervals of 30 seconds, instead of being added all at once at the beginning of the polymerization. This method keeps the local concentration of acid chloride low and provides a high quality membrane. Percentage yield is comparable to that stated in example 1.
Eksempel 4 Example 4
De fremstilte kapsler har lignende egenskaper som de i Capsules of terephthaloyl chloride and hexanediamine containing 2,5-diaminobenzenesulfonic acid were prepared using the same procedure as in Example 1, except that 2,5-diaminobenzenesulfonic acid was added to hexanediamine instead of DMDSA. The manufactured capsules have similar properties to those in
eksempel 1. example 1.
Eksempel 5 Example 5
Sebacyl chloride-hexanediamine carbonate microcapsules were prepared using the procedure of Example 1, except that sebacyl chloride was used instead of terephthaloyl chloride and DBDSA was not used. This method produces microcapsules with similar properties to the microcapsules in example 1.
Eksempel 6 Example 6
Capsules of sebacyl chloride-lysine were prepared according to the procedure given in Example 5, except that either 3M lysine or a solution consisting of equal amounts of 3M hexanediamine carbonate and 3M lysine was used instead of 3M hexanediamine carbonate. A few microcapsules produced by this method were leaky and unsatisfactory. Others showed similar properties to those produced in Example 1.
Eksempel 7 Example 7
1,5 ml bovin-albumin (30%) ble tilsatt til 1 ml av DBDSA-heksandiaminkarbonatløsningen i eksempel 1 sammen med 0,4 ml 36% KC1 og 0,1 ml 2,9M tetraetylenpentamin (på forhånd justert til 1.5 ml of bovine albumin (30%) was added to 1 ml of the DBDSA-hexanediamine carbonate solution of Example 1 along with 0.4 ml of 36% KCl and 0.1 ml of 2.9 M tetraethylenepentamine (adjusted beforehand to
Det tilsettes 0,4 ml tereftaloylkloridløsning (0,5M i heksan-kloroform fra eksempel 1) i 0,1 ml porsjoner hvert 30. sekund, etterfulgt av ytterligere tilsetninger av 0,1 ml hvert minutt i de neste 6 minutter. pH 9.5 with HCl). 4.0 ml of "SPAN 85" and 10 ml of the hexane-chloroform solution from Example 1 were then added to the mixture and an emulsion was obtained, within 1-2 minutes, with vigorous stirring using a magnetic stirrer . 0.4 ml of terephthaloyl chloride solution (0.5M in hexane-chloroform from Example 1) is added in 0.1 ml portions every 30 seconds, followed by further additions of 0.1 ml every minute for the next 6 minutes.
After centrifugation to separate the capsules, the supernatant was discarded and the untreated microcapsules were resuspended in cyclohexane containing 5% "SPAN 85". 0.7 ml of terephthaloyl chloride was added and the suspension was stirred for 3 minutes. After separation of the phases by centrifugation, the microcapsules were washed once in pure cyclohexane and subjected to final washing and recycling as indicated in example 1.
Labile, biological materials, including enzymes, can be dissolved in albumin solution if desired. Tetraethylenepentamine helps to crosslink the polymer to produce strong, semipermeable membranes with uniform pore size.
