Abstract

The (-)-(R)- and (+)-(S)-enantiomers of 3-/ 2-4-(8-fluoro-10, 11-dihydro-2-methyl-dibenzo / b,f / thiepin-10-yl)-1-piperazinyl / ethyl / -2-oxazolidinone (I) and their salts are new cpds. The (+)-(S) enantiomer is prepd. (a) by resolving the racemate, esp. with (+) camphor-10-sulphoric acid or (b) by reacting the corresp. piperidine enantiomer with 2-(2-X-ethyl)oxazolidinone (X = leaving group). (+)-(S)-(I) has strong neuroleptic and CNS-depressant activities but low toxicity (LD50 (p.o. mice) >8g/kg (after 24 hrs.) and 2.1g/kg after 10 days) and minor cataleptic side effects (cataleptic ED50 in rats 34mg/kg i.p.). It is useful in treating acute or chronic schizophrenia and as a tranquiliser.
