Abstract

Se describen modificaciones a la somatotropina que muestran una mejora inesperada en la estabilidad conformacional y química sin pérdida de actividad biológica. Las alteraciones de estas características se obtienen mediante la modificación de las somatotropinas por mutagénesis dirigidas al sitio. Estos cambios aumentan la estabilidad de almacenamiento de estas proteínas. Ejemplos de modificaciones incluyen las somatotropinas porcinas en las que la histidina y la arginina substituyen a la glutamina y la histidina en las posiciones 19 y 20, respectivamente (Q19h,h2or), el ácido glutámico substituye a la fenilalanina en la posición 52 (F52E) y el ácido glutámico substituye a la leucina en la posición 137 (I137E). Alterations of these characteristics are obtained by modifying somatotropins by site-directed mutagenesis. These changes increase the storage stability of these proteins. Examples of modifications include porcine somatotropins in which histidine and arginine replace glutamine and histidine at positions 19 and 20, respectively (Q19h, h2or), glutamic acid replaces phenylalanine at position 52 (F52E) and glutamic acid replaces leucine at position 137 (I137E). Modified somatotropins are useful for increasing growth characteristics in animal species, eg, for increasing the ratio of lean to adipose tissue in pigs and milk production in cows.
