Abstract

The Present disclosure is related to a measuring instrument of diffusion coefficient of molecules marked with fluorescence in liquid samples. The instrument uses the FRAP technique and consists of a laser beam that passes through the window of an acoustic optical modulator controlled electronically to attenuate the intensity of the laser beam. As a result, it is possible to obtain, at will, either a very intense beam for bleaching or erasing the fluorescence (high power) or a low intensity excitation beam (low power) which allows the residual fluorescence to be monitored. With the aid of other optical elements, the light beams are directed towards the sample to excite or destroy the fluorescence. Subsequently, the instrument measures the recovery of fluorescence due to diffusion of the fluorescent molecules in the surrounding area by means of a photon detector, sending the signal to a high speed card in a computer. The fluorescence recovery time allows obtaining the diffusion coeffici ent of the fluorescent molecule. By means of graphic programming, it is possible to control the sequence to send signals to the devices and measurements can be recorded. Finally, the recovery is plotted and the corresponding equations for obtaining diffusion coefficients are applied.
