Abstract

Se describe un método no invasivo para la detección genética antes de la implantación del embrión. Los ovocitos fertilizados se cultivan el día 1, seguido de la eliminación de las células de corona/cumulus residuales, lo que reduce la contaminación materna; aislamiento de ovocitos y cultivo individual desde los días 1-4 con suplemento de proteína sérica; llevar a cabo la ruptura de la zona con láser el día 4 permitiendo que el ADN libre de células embrionarias (cfADN) entre en el medio de cultivo; lavar los ovocitos con medio fresco para producir un medio que contiene cfADN del día 4; transferir al medio nuevo bajo aceite y cultivar hasta el día 5, 6 o 7 para formar un blastocisto expandido que se transfiere el día 5/6/7 a una gota de medio; exponer el blastocisto expandido a un pulso de láser para extruir el líquido de blastocoel que contiene cfADN embrionario, obteniendo el día 5/6/7 cfADN; y la modalidad de exámenes genéticos de cfADN utilizando la amplificación del genoma completo (WGA). El método se puede usar para la detección genética de embriones en busca de aneuploidía mediante la prueba de WGA para el número de copias de cromosomas completos.
