Abstract

Un método para detectar múltiple y simultáneamente una combinación de bacterias Gram positivas y/o bacterias Gram negativas en una muestra, el método comprende el proporcionar ADN extraído de la muestra; preparación de una mezcla de reacción que incluye: el ADN proporcionado; uno o más pares de iniciadores de oligonucleótido de secuencia seleccionada del grupo que consiste de SEQ ID NO: 1 y SEQ ID NO: 2, SEQ ID NO: 4 y SEQ ID NO: 5, SEQ ID NO: 7 y SEQ ID NO: 8, SEQ ID NO: 10 y SEQ ID NO: 11,SEQ ID NO: 13 y SEQ ID NO: 14, SEQ ID NO: 16 y SEQ ID NO: 17 y sus combinaciones; y uno o más sondas de oligonucleótido de secuencia seleccionada del grupo que consiste de SEQ ID NO: 3, SEQ ID NO: 6, SEQ ID NO: 9, SEQ ID NO: 12, SEQ ID NO: 15, SEQ ID NO: 18 y sus combinaciones; amplificación, mediante PCR en tiempo real, del ADN proporcionado en la mezcla de reacción; y determinación simultánea de presencia o ausencia de las bacterias Gram positivas y/o bacterias Gram negativas en la muestra. El método permite identificar cualquier combinación de tres especies de bacterias Gram positivas (Listeria ssp., Listeria monocytogenes y Staphylococcus aureus) y tres especies de bacterias Gram negativas (Escherichia coli spp., Escherichia coli O157:H7 y Salmonella spp.).
