Abstract
The invention provides gene encoding the hydantoinase protein separated from the Bacillus brevis having a high D-hydantoin hydrolysis activity under a high temperature and alkali condition. In order to recombine the gene, the Bacillus brevis isolate strain is cultured in a LB medium and makes a check the hydantoinase activity. The synthesized oligonucleotide based on N-terminal amino acid sequences of the protein is used in the separation of chromosome DNA from the Bacillus brevis in order to separate and clone the gene of the protein having the hydantoinase activity. The prepared O gt11 library is searched to clone the hydantoinase gene, the produced gene DNA is inserted in the Eco RI location of pUC19 of plasmid gene carrier to prepare recombinant DNA pHMY 180.
