Abstract
본 발명은 사카로마이시즈 세레비지애(Saccharomyces cerevisiae)의 SWI3 유전자와 유추 아미노산 서열에 있어 매우 높은 상동성을 나타내는 마우스 유래의 신규한 SRG3(SWI3-related gene)유전자, 그로부터 유추되는 아미노산 서열을 가지는 SRG3 단백질 및 전기 유전자로부터 발현되어 마우스의 SW12계 단백질과 복합체 SRG3 단백질에 관한 것이다. 본 발명의 SRG3 유전자는 1101개의 아미노산을 암호화하는 3303bp의 재방해독틀을 포함하고 있다. 아울러, SRG3 유전자로부터 유추되는 아미노산 서열을 분석한 결과, N-말단 부위는 아스파라긴산과 글루탐산이 풍부하여 산성을 띠고, 아르기닌과 라이신이 풍부한 염기성 부위는 C-말단의 로이신-지퍼 모티브와 인접한 곳에 위치하며, C-말단 부위는 프롤린과 글루타민이 풍부한 것으로 확인되었으며, 또한 N-글리코실화 부위 및 인산화 부위가 존재하는 것으로 확인되었다. 본 발명에서 분리한 마우스의 SRG3 유전자는 SRG3 단백질의 발현 및 분리등에 이용될 수 있으며, 나아가 사람의 SWI3계 유전자 분리에 이용될 수도 있다. 또한 SRG3 단백질을 T세포 분화의 주요 장소인 흉선에서 높게 발현되므로, 면역 조절인자의 탐색 및 개발에 이용될 수도 있다.The present invention relates to a novel SRG3 (SWI3-related gene) gene derived from a mouse, which shows a very high homology with the SWI3 gene of Saccharomyces cerevisiae in a similar amino acid sequence, and an amino acid sequence deduced therefrom SRG3 protein and a complex SRG3 protein expressed from an electric gene and expressed in mouse SW12 system protein.
