Abstract
본 발명은 인간알파 인터페론의 대량생산을 위해 살모넬라(Salmonell) 균주의 L-아라비노스(arabinose)오페론을 이용한 유도헝 프로모터가 포함된 유전자 재조합 대장균을 그라이셀롤과 효모추출물을 기본성분으로 하는 배지에서 배양하다가 일정한 시간에 L-아라비노스를 첨가하여 발현 유도를 시작한 후 배양액 내의 용준 산소량을 고갈 성분의 영양지표로 하여 배지를 공급함으로써 세포의 성장과 함께 지속적으로 재조합 유전자의 발현을 유도하여 발현된 재조합 인간 알파 인터페론이 세포내에 봉입체로서 다량 축적되게 하는 유가식 회분 발효방법을 제공한다.The present invention is to culture the recombinant E. coli containing the induced H. promoter using L-arabinose operon of Salmonell strain for mass production of human alpha interferon in a medium based on glycerol and yeast extract.
