배지의 조성은 Peptene 5g, Glucose 10g, Na2HPO40.008g, NH4NO30.0016g, NaCl 23.4g, KCl 0.7g, MgCl2ㆍ6H2O 10.6g, CaCl21.1g, Na2SO43.9g, NaHCO30.2g, (NH4)2SO41.0g, K2HPO40.01g, Tris 6.05g, D.W.1ℓ. pH 7.8을 사용하여 미생물을 30℃에서 72시간 진탕배양시킨 후 배양액을 3~5℃에서 30,000~35,000 x g에서 10분간 원심분리하여 상층액과 침전물(미생물세포)로 분리한 미생물세포의 침전물에 동량의 증류수를 가한 다음 50~60℃에서 1~3시간 동안 강하게 교반시키면서 가열처리한 다음 35~45℃에서 30,000~35,000 x g에서 8~12분간 원심 분리하여 세포로부터 세포벽 부착 다당을 분리하여 최종 농도가 8~12%(Weight/Volume)가 되도록 세틸피리디니움 클로라이드(CPC, Cetylpyridinium Chloride)를 가하여 산성다당(Acidic poly-Saccharide)을 분리한 다음 이를 다시 10%의 염화나트륨(NaCl)용액에 완전히 재용시킨 후 2배의 냉각된 아세톤(Cold Acetone)으로 침전시켜 투석, 동결건조하여 분리한 것을 인산 완충용액(Phosphate Buffer)에 용해시킨 후 Sepharose CL-4B가 채워진 컬럼(Column)에서 켈크로마토그래피(Gelehromatography)를 행하여 완전정제하여서 되는 쥬글리아(Zoogloea sp.)가 생산하는 세포벽 부착 다당.The composition of the medium was Peptene 5g, Glucose 10g, Na 2 HPO 4 0.008g, NH 4 NO 3 0.0016g, NaCl 23.4g, KCl 0.7g, MgCl 2 6H 2 O 10.6g, CaCl 2 1.1g, Na 2 SO 4 3.9 g, 0.2 g NaHCO 3 , 1.0 g (NH 4 ) 2 SO 4 , 0.01 g K 2 HPO 4 , 6.05 g Tris, DW1 L.
