Claims (10)

  1. 침전법, 멤버레인 여과 및 크로마토그래피의 병용 도움으로 흉선으로 부터 폴리펩티드를 분리하고, 필요시 생성된 펩티드를 화학적 또는 효소적 방법에 의해 분해물로 분해시키고, 필요시 생성된 펩티드를 그의 N-아실 또는 N-아실글리실 유도체로 전환시키고, 필요시 생성물을 염으로 전환시킴을 특징으로 하여 면역계에 작용하며 분자량이 3480이고 205내지 10nm에서 최대 UV흡수를 나타내며 등전점이 3.95±0.15이고 아미노산 조성이 4아스파라긴산, 3트레오닌, 3세린, 6글루탐산, 1글리신, 4알라닌, 3발린, 1이소로이신, 1로이신, 5리신 및 1알기닌이고 필요시 N-말단이 탄소수 1내지 6의 아실그룹 또는 아실글리실그룹(여기에서 아실그룹은 1내지 6개의 탄소원자를 갖는다)을 가진 폴리펩티드, 그의 펩티드 분해산물 및 /또는 약물학적으로 허용할 수 있은 염을 분리하는 방법.
  2. 제1항에 있어서, 90% 이상의 순도를 가진 폴리펩티드인 방법.
  3. 제1항에 있어서, pH5.0내지 9.0에서 완충작용을 갖는 무기염의 수용액을 사용하여 흉선을 추출하고 불용성물질은 분리 제거하고 생성된 추출물로 부터 분리 공정에 의해 폴리펩티드를 분리시킴이 특징인 방법.
  4. 제1항에 있어서, 크로마토그래피단계가 양이온 및/또는 음이온 교환크로마토그래피, 비-특이성흡착제상에서의 크로마토그래피, 분배 크로마토그래피 또는 상기 방법을 2가지 이상 병용하는 것임이 특징인 방법.
  5. 제1항에 있어서, 펩티드는 용액에 남아 있는 반면 기타의 단백질은 침강 또는 여과로 분리제거할 수 있게 열변성시킨 추출물을 제4항에 따른 크로마토그래피에 사용함이 특징인 방법.
  6. 제4항에 있어서, 수-혼화성 유기용매를 제3항 또는 제5항의 추출물에 가하여 침전물로서 분리된 물질을 크로마토그래피에 사용함이 특징인 방법.
  7. 제4항에 있어서, 사용된 염에 따라 특정 농도에서 펩티드가 침전되고 침강법으로 농축되게 수-용성 무기 및 /또는 유기염을 가해 분열 침전시킨 추출물을 크로마토그래피에 사용함이 특징인 방법.
  8. 제4항에 있어서, 멤버레인 여과 및 /또는 젤여과로 분류한 결과 10,000달톤 이하의 분자량을 가진 펩티드만 여전히 함유한 추출물을 크로마토그래피에 사용함이 특징인 방법.
  9. 제4항에 있어서, 제5항 내지 제8항중의 어느 하나에 따른 방법을 2가지이상 병용하여 수득한 추출물-을 크로마토그래피에 사용함이 특징인 방법.
