Abstract

Description
본 발명은 암의 예방 및/또는 치료, 구체적으로 교모세포종의 치료에 사용하기 위한 miR-17 모방체 및 miR-340 모방체의 조합물에 관한 것이다. 구체적으로, 상기 조합물은 miR-222 antagomiR을 추가로 포함한다.
교모세포종 (Glioblastoma: GBM)은 치명적인 악성 뇌종양인 등급 IV 성상세포종으로, 성인에서 가장 흔한 원발성 뇌종양이다. 오늘날, 수술, 방사선요법, 및 테모졸로미드 (temozolomide)를 사용한 화학요법이 GBM 환자의 치료 표준으로 남아 있다 (Stupp, et al. (2005). miRNAs는 매우 중요한 다양한 세포 과정에서 중요한 역할을 하기 때문에, 개인 맞춤형 암 치료에서 miRNAs의 사용은 매우 매력적이다.
그러나, GBM 공격성의 새로운 동인, 및 GBM 치료 전략에 대한 임상적 및 생물학적으로 관련된 miRNAs를 확인하기 위한 더 많은 조사가 분명히 필요하다.
본 발명의 발명자는 GBM 다중표적화 치료 전략에 사용될 수 있는 임상적으로 관련된 miRNAs로서 miR-17 및 miR-340을 확인하였다. 이들은 miR-17 및 miR-340의 이소성 발현 (ectopic expression)이 종양 공격성을 인 비트로 및 인 비보 억제하였음을 발견하였다.

용어 "마이크로RNA (microRNA)" 또는 "miRNA" 또는 "miR"은 유전자 발현의 전사 및/또는 전사후 조절에서 기능하는 작은 비-코딩 RNA를 지칭하기 위해 본원에서 상호교환적으로 사용된다. 다양한 구체예에서, miRNAs는 가이드 가닥 (guide strand) 및 패신저 가닥 (passenger strand)으로 처리되는 듀플렉스 (duplex)를 포함하는 헤어핀 구조를 갖는다.

이전에 설명한 바와 같이, 일부 과발현된 miRNAs는 "종양발생 (oncogenic)" 효과를 발휘하는 것으로 보고된 반면에, 일부 하향조절된 miRNA는 "종양 억제 (tumor suppressor)" 효과를 보여주었다. 특정 miRNA 변경은 각각 표적화된 miRNAs의 상향조절 또는 하향조절을 유도하는 "모방체 (mimics)" 또는 "antagomirs"로 지칭되는 올리고뉴클레오티드 서열을 사용하여 구체적으로 표적화될 수 있다. 그러므로, 종양에서 변경된 miRNAs를 확인함으로써, 본 발명자는 암 환자, 구체적으로 GBM 환자의 개인 맞춤형 의학을 위한 표적 요법을 시행할 수 있었다.

본원에서 miR-17 모방체, miR-340 모방체 및 miR-551b 모방체에 사용된 "모방체 (mimic)"는 이러한 miRNAs를 모방하도록 디자인된 작은, 합성, 이중가닥 RNA 분자를 의미하며, 상기 언급한 바와 같이, 이는 표적화된 miRNAs의 상향조절을 유도할 수 있다. 상기 모방체는 해당 miRs의 세포내 기능을 증가시키거나 또는 대체하며, 모방하는 miRNA에 의해 발현이 하향조절되는 유전자 산물의 발현에 대응할 수 있다. miRNA 모방 기술은 내인성 miRNAs를 모방하여 이의 표적에 특이적으로 결합하고, 이에 의해 표적 유전자의 번역 억제를 유도하는데 사용되는 miRNA-기능 획득 전략 (miRNA-gain-of-function strategy)에 속한다.

DNA-기반 miRNA 모방체, RNA-기반 miRNA 모방체, miRNA를 코딩하는 폴리뉴클레오티드 또는 이의 전구체를 예로 들 수 있다.
DNA-기반 miRNA 모방체는 성숙한 miRNA 서열의 DNA 버전 또는 이의 기능적으로 동등한 변이체를 포함하는 DNA 올리고뉴클레오티드에 해당한다.

물질 및 방법
GBM 세포주 및 환자-유래 모델

세포 이동

루시퍼라제 분석

본 발명의 연구 결과를 임상에 적용하기 위해, 본 발명자는 인 비보에서 Ge518 종양 성장에 대한 조합 치료의 효과를 조사하고자 하였다. 이를 위해, 본 발명자는 누드 마우스의 옆구리에 Ge518 세포를 피하 주사하였다 (도 10의 A). 종양이 150 mm2에 도달하면, 마우스를 miR-Combo로 치료하였다. 본 발명자는 인 비트로에서 밝혀진 바와 같이, 종양 성장의 유의미한 지연을 보여주었고, 이는 miR-Combo가 GBM 성장 및 종양형성 능력에 효율적으로 영향을 미치는 것을 나타낸다 (도 10의 A). 본 발명자는 또한 miR-Ctrl과 비교하여 miR-Combo에서 세포 증식의 마커인 Ki-67 발현의 유의미한 감소를 발견하여, GBM 세포 증식에 대한 조합 전략의 효과를 확인하였다 (도 10의 B-C). 잘 설명된 miRGE 렌티벡터 시스템 (Myburgh, et al. (2014). Optimization of Critical Hairpin Features Allows miRNA-based Gene Knockdown Upon Singlecopy Transduction. Mol Ther Nucleic Acids 3: e207.)을 사용하여, 본 발명자는 GBM PDCs에서 miR-17-3p, 및 miR-340 모방체들, 및 miR-222 antagomiR을 동시에 발현시켰다.
그 다음에, Tet-On 시스템을 사용하여, GBM 세포를 독시사이클린으로 처리하여 miRNA 조절을 수행하였다. 도 11의 A에 나타낸 바와 같이, 독시사이클린 처리는 Ge518, Ge738, 및 Ge970.2 PDCs 모두에서 세포 생존율의 유의미한 감소를 유도하였다.

물질 및 방법
세포 형질감염
miR-17-3p, 및 miR-340-5p 모방체들, 및 miR-222-5p antagomir를 제조자의 프로토콜에 따라 최종 농도 5 nM으로 리포펙타민 RNAimax (Invitrogen)를 사용하여 형질감염시켰다. 비-표적화 스크램블 miRNA (Life Technologies)를 대조군으로서 사용하였다.
세포 생존율 분석
세포 생존율 분석은 제조자의 프로토콜에 따라 CellTiter-Glo 분석 키트 (Promega)를 사용하여 수행하였다.
