Abstract
본 발명은 코로나바이러스 NSP9이 처리된 편도 유래 질병 줄기세포 모델 및 이의 용도에 관한 것으로, 구체적으로 분리된 편도 조직 유래 줄기세포에 NSP9(Non structural protein 9) 단백질을 처리하는 단계를 포함하는 질병 줄기세포 모델의 제조방법 및 상기 방법으로 제조된 질병 줄기세포 모델을 이용한, 항바이러스제의 스크리닝 방법에 관한 것이다. 본 발명에서는 바이러스의 침습이 쉬운 구강 내 편도 유래 줄기세포를 활용하여 바이러스의 비구조단백질인 NSP9 단백질이 처리된 세포 독성이 유발된 질병 줄기세포 모델을 확립함에 따라, 코로나 바이러스의 비구조 단백질 중 하나인 NSP9의 세포 유해성 및 세포내 기전 연구에 유용하게 사용할 수 있을 뿐만 아니라 항바이러스제의 스크리닝 분야에도 유용하게 사용할 수 있다.
Description
본 발명은 코로나바이러스 NSP9 단백질이 처리된 편도 유래 질병 줄기세포 모델 및 이의 용도에 관한 것이다.
질병 발생에 영향을 주는 역학의 기본요인인 병인, 숙주, 환경 중 병인은 생물학적, 물리-화학적, 사회적 요인 등으로 세분할 수 있는데 이들은 질병 발생의 주요 요인이다. 병원체의 일반적인 침입 경로는 호흡기, 위장관, 피부 등이며 주로 호흡기를 통해 가장 많은 미생물이 흡입된다. 그중 바이러스는 곰팡이나 박테리아보다 훨씬 작고 살아있는 세포에 침투하는 전염성 미생물이다. 바이러스는 세포에 달라붙어 침투하고 유전물질을 세포 안으로 방출하여 세포를 제어하고 바이러스를 복제하게 만든다.
이러한 바이러스는 구조단백질, 비구조단백질, 조절단백질, 부속단백질 등 그 기능에 따라 분류될 수 있고, 바이러스의 유전체에 암호화되어있는 단백질의 수는 많지 않다. 이중, 대부분의 구조 단백질은 캡시드와 외피를 이루는데 사용되고, 비구조단백질은 감염된 세포 내에서 발현이 되며 바이러스 복제에 중요한 기능을 하여 면역을 조절하고 전사를 촉진하는 역할을 한다.
호흡기를 통한 바이러스는 주로 상기도감염으로 나타나며 감염된 사람과의 접촉이나 호흡기 비말을 통해 쉽게 전파된다. 상기도는 기도의 윗부분에 해당하며 인간의 호흡기 중에서 가장 침습이 쉬운 부위로 상기도 감염은 바이러스 감염 및 세균 감염이 주로 발생한다. 급성 호흡기 감염은 우리나라에서도 가장 많이 발생하는 질병 1위이며 기저질환이 없는 건강한 성인에서도 발생하며 대부분 바이러스로 인한 것으로 대증 치료가 원칙이다. 호흡기와 관련된 기관 중 바이러스가 침습할 수 있는 부분은 구강이며 구강 내 편도는 이러한 상기도 감염에 의한 유해성 및 영향을 가장 빠르게 나타낼 수 있는 기관 중 하나이다.
이에, 본 발명자들은 편도 유래 줄기세포를 이용하여 코로나 바이러스의 비구조단백질인 Non structural protein 9 (NSP9)과의 상호작용을 구체적으로 확인하여 신규한 유해성 및 영향을 조사하고, 항바이러스 후보물질 및 안전성 확인에 활용할 수 있는 NSP9이 처리된 바이러스 감염 편도 유래 줄기세포를 제조함으로써 본 발명을 완성하였다.
따라서 본 발명의 목적은 분리된 편도 조직 유래 줄기세포에 NSP9(Non structural protein 9) 단백질을 처리하는 단계를 포함하는, 질병 줄기세포 모델의 제조방법을 제공하는 것이다.
본 발명의 다른 목적은 본 발명의 방법으로 제조된 질병 줄기세포 모델을 이용한, 항바이러스제의 스크리닝 방법을 제공하는 것이다.
상기와 같은 본 발명의 목적을 달성하기 위해서, 본 발명은 분리된 편도 조직 유래 줄기세포에 NSP9(Non structural protein 9) 단백질을 처리하는 단계를 포함하는, 질병 줄기세포 모델의 제조방법을 제공한다.
본 발명의 일실시예에 있어서, 상기 NSP9 단백질의 처리는 0.2~0.3 ug/ml의 농도로 처리하는 것일 수 있다.
본 발명의 일실시예에 있어서, 상기 질병 줄기세포는 코로나바이러스 감염증을 나타내는 것일 수 있다.
본 발명의 일실시예에 있어서, 상기 NSP9 단백질은 처리된 줄기세포의 미토콘드리아 대사에 영향을 주는 것일 수 있다.
또한 본 발명은 본 발명의 방법으로 제조된 질병 줄기세포 모델을 이용한, 항바이러스제의 스크리닝 방법을 제공한다.
본 발명의 일실시예에 있어서, 상기 질병 줄기세포에 후보약물을 처리한 후, 미토콘드리아의 대사 변화를 분석하는 단계를 포함할 수 있다.
본 발명은 코로나바이러스 NSP9이 처리된 편도 유래 질병 줄기세포 모델 및 이의 용도에 관한 것으로, 본 발명에서는 바이러스의 침습이 쉬운 구강 내 편도 유래 줄기세포를 활용하여 바이러스의 비구조단백질인 NSP9 단백질이 처리된 세포 독성이 유발된 질병 줄기세포 모델을 확립함에 따라, 코로나 바이러스의 비구조 단백질 중 하나인 NSP9의 세포 유해성 및 세포내 기전 연구에 유용하게 사용할 수 있을 뿐만 아니라 항바이러스제의 스크리닝 분야에도 유용하게 사용할 수 있다.
도 1은 편도 조직 유래 줄기세포를 대상으로 NSP9 단백질을 농도별로 처리한 후, WST-8 키트를 이용하여 세포독성을 흡광도로 분석하여 나타낸 결과이다.
도 2는 NSP9 단백질을 농도별로 처리한 편도 조직 유래 줄기세포 실험군을 대상으로 살아있는 세포 및 죽은 세포를 분석한 결과를 나타낸 것이다.
도 3은 0.24 μg/ml의 NSP9가 처리된 편도 조직 유래 줄기세포 및 NSP9이 처리되지 않은 대조군에 대한 미토콘드리아 대사능을 비교 분석한 결과이다.

본 발명은 코로나바이러스 NSP9이 처리된 편도 유래 질병 줄기세포 모델 및 이의 용도에 관한 것이다.
현재 세계적으로 전염병 발생이 증가하고 있으며, 스페인 독감과 조류 독감 등 치사율이 높은 전염병 역시 매해 증가하는 양상을 보일 뿐만 아니라 최근 COVID-19의 엄청난 감염성으로 인해 바이러스 감염에 대한 연구 및 신속한 치료제의 개발이 매우 시급한 실정이다.
이에 본 발명자들은 생체외에서 바이러스 감염 및 항바이러스제를 신속하고 정확하게 발굴할 수 있는 플랫폼을 개발하기 위해 연구하던 중, 인간의 편도 조직으로부터 분리된 줄기세포를 대상으로 코로나바이러스의 비구조단백질 NSP9을 처리하여, 세포독성이 유발된 질병 줄기세포 모델을 확립하였다.
따라서 본 발명은 분리된 편도 조직 유래 줄기세포에 NSP9(Non structural protein 9) 단백질을 처리하는 단계를 포함하는, 질병 줄기세포 모델의 제조방법을 제공할 수 있다.
상기 편도 조직 유래 줄기세포는 인간의 편도 조직으로부터 분리된 줄기세포 일 수 있으며, 상기 NSP9 단백질은 0.2~0.3 ug/ml의 농도로 처리할 수 있다.
본 발명의 일실시예에서는 편도 조직 유래 줄기세포를 대상으로 NSP9 단백질을 농도별로 처리한 후, 세포 독성을 나타내는 상기 단백질의 농도를 확인한 결과, 0.2ug/ml, 바람직하게는 0.24 ug/ml 농도로 처리한 군이 다른 농도로 처리한 군에 비해 NSP9 단백질 처리로 인한 세포 독성이 유발되는 것으로 나타났다.
따라서 상기 단백질로 세포 독성이 유발된 세포는 바이러스 감염과 유사 또는 동일한 질병 세포모델에 해당된다고 볼 수 있으며, 구체적으로 상기 바이러스는 코로나바이러스 일 수 있다.
본 발명의 다른 일실시예에서는, NSP9 단백질이 편도 조직 유래 줄기세포에 미치는 영향을 분석하였는데, 그 결과, 미토콘드리아 대사에 영향을 미치는 것으로 나타났고, 구체적으로 Basal은 전체적인 미토콘드리아 대사능이 NSP9 처리에 의해 감소된 것으로 나타났고, proton에 의한 산소 소비율(Proton Leak), 최대 산소 소비율(Maximal Respiration), 잔존 산소량 소비율(Spare respiratory capacity), ATP 생성 비율 모두 NSP9 처리군이 대조군에 비해 감소한 것으로 나타나, NSP9이 세포 내에서 미토콘드리아 대사에 이상을 야기하여 세포의 독성을 유발할 수 있다는 것을 알 수 있었다.
또한 본 발명은 상기 본 발명의 방법으로 제조된 NSP9(Non structural protein 9) 단백질이 처리된 질병 줄기세포 모델을 제공할 수 있다.
나아가 본 발명은 상기 본 발명의 방법으로 제조된 질병 줄기세포 모델을 이용한, 항바이러스제의 스크리닝 방법을 제공할 수 있다.
항바이러스제의 스크리닝은 상기 질병 줄기세포에 후보약물을 처리한 후, 미토콘드리아의 대사 변화를 분석하는 단계를 통해 수행할 수 있으며, 구체적으로 후보약물 처리 시, 비정상적인 미토콘드리아의 대사를 정상화시키거나 개선하는 효과가 있는 경우, 상기 후보물질을 항바이러스제로 판단할 수 있다.
이외에도 상기 본 발명의 방법으로 제조된 질병 줄기세포 모델은 코로나바이러스의 NSP9 단백질의 세포 유해성 및 세포내 기전 연구에도 유용하게 활용할 수 있다.
이하, 실시예를 통하여 본 발명을 보다 상세히 설명하고자 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
<실시예 1>
편도 유래 줄기세포에 미치는 NSP9의 독성 농도 조사
본 발명자들은 편도 유래 줄기세포를 대상으로 NSP9를 농도별로 처리하여 줄기세포에 독성을 나타내는 NSP9의 농도를 분석하기 위한 실험을 수행하였다. 이를 위해 NSP9 단백질을 0.05, 0.1, 0.2, 0.4, 0.8 μg/ml 농도가 되도록 배지로 희석하여 편도 조직 유래 줄기세포에 처리하고 37℃, 5% CO2 배양기 내에서 4시간 동안 배양시켰다. 4시간 경과 후, 세포의 수에 비례하여 발색 반응이 나타나는 WST-8 kit 시약을 사용하여 동량의 시약을 넣어주고 배양기 내에서 4시간 반응시켰다. 반응 후 460nm의 파장에서 흡광도를 확인하고 이를 도표로 표시하였다.
그 결과, 편도 유래 줄기세포에 NSP9 단백질을 농도별로 처리한 결과, NSP9을 처리하지 않은 대조군에 비해 0.2 μg/ml의 농도에서 유의적으로 세포 독성이 유발되는 것으로 나타났다. 한편, 0.05 및 0.1 μg/ml 농도로 처리한 군과 0.4 및 0.8 μg/ml의 농도로 처리한 군에서는 세포 독성이 유발되지 않는 것으로 나타났다. 이러한 결과를 통해 본 발명자들은 시험관 내에서 편도 조직 유래 줄기세포에 NSP9 단백질을 0.2 μg/ml 농도로 처리할 때, 바이러스 감염과 같이 세포에 유해한 독성이 유발될 수 있음을 알 수 있었다.
<실시예 2>
NSP9 농도별 세포 독성 조건 확립
상기 <실시예 1>의 결과를 바탕으로, 농도별 세포 독성 조건을 확립하기 위해 살아있는 세포와 사멸한 세포를 각각 염색하여 확인하였다. 각 세포를 염색하기 위해 LIVE/DEAD kit를 활용하여 세포에 농도별로 NSP9 단백질을 37℃, 5% CO2 배양기 내에서 4시간 처리한 후 kit의 시약을 동량 사용했다. 시약의 반응시간은 30분으로 동일하게 진행하고 형광 현미경을 이용하여 무작위하게 5곳의 부분을 찍은 뒤, image J 프로그램으로 형광값을 수치화하여 도표로 나타냈다.
그 결과, 도 2에 나타낸 바와 같이, 살아있는 세포와 사멸한 세포의 두 도표에서 모두 NSP9 단백질을 모두 0.24 μg/ml의 농도로 처리한 군에서 가장 발현의 차이를 보이는 것을 확인할 수 있었다. 즉, 0.24 μg/ml 농도로 처리한 군이 다른 농도로 처리한 군에 비해 살아있는 세포가 가장 적고 사멸한 세포가 가장 많은 것으로 나타나, 편도 조직 유래 줄기세포에 NSP9 단백질을 0.2~0.3 μg/ml 농도로 처리하였을 때, 질병 세포모델의 제조가 가능하다는 것을 알 수 있었다.
<실시예 3>
NSP9 처리에 따른 줄기세포 내 미토콘드리아 대사능의 발현 비교분석
NSP9의 유효한 독성 농도 조건인 0.24 μg/ml를 처리한 군과 NSP9을 처리하지 않은 군에 대해 미토콘드리아 대사능을 비교 분석하였다. 미토콘드이라 대사능은 세포의 휴식기 때의 대사 능력, ATP 생성과 관련된 호흡 비율 등 각 구간별 세포내 미토콘드리아가 산소를 에너지로 변화시키는 대사 능력을 관찰하였다.
그 결과, 도 3에 나타낸 바와 같이, NSP9 단백질을 처리한 군은 이를 처리하지 않은 대조군에 비해 미토콘드리아의 대사 능력이 감소된 것으로 나타났다.
<실시예 4>
미토콘드리아 대사 항목별 수치 비교분석
상기 실시예 3의 결과를 바탕으로 본 발명자들은 NSP9 단백질이 편도 조직유래 줄기세포의 미토콘드리아에 미치는 영향을 보다 구체적으로 확인하기 위해, 각 세부 항목별 NSP9 에 따른 대사능의 변화를 분석하였다.
그 결과, 도 4에 나타낸 바와 같이, Basal은 전체적인 미토콘드리아 대사능이 NSP9 처리에 의해 감소된 것으로 나타났고, proton에 의한 산소 소비율(Proton Leak), 최대 산소 소비율(Maximal Respiration), 잔존 산소량 소비율(Spare respiratory capacity) 모두 NSP9 처리군이 대조군에 비해 감소한 것으로 나타났다. 한편, Non-mitochondrial oxygen consumption은 미토콘드리아 내에서의 산소 소비율이 아닌 다른 기관에서의 산소 소비율을 나타내는데 이 경우에는 NSP9 처리군이 대조군에 비해 높은 것으로 나타났다. 또한, ATP 생성 비율은 NSP9 처리군이 대조군에 비해 유의적으로 감소한 것으로 나타났다.
이상의 결과를 통해 본 발명자들은 코로나 바이러스의 비구조 단백질 중 하나인 NSP9을 처리하여 세포 독성이 유발된 질병 세포모델을 확립할 수 있었고, NSP9이 편도 유래 줄기세포의 미토콘드리아에 영향을 미치는 것을 알 수 있었으며, 본 발명에서 확립한 질병 세포모델을 이용할 경우, 항바이러스제를 스크리닝 하는 방법에도 유용하게 사용할 수 있음을 알 수 있었다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.

상기 NSP9 단백질의 처리는 0.2~0.3 ug/ml의 농도로 처리하는 것을 특징으로 하는 방법. - 제1항에 있어서,
상기 질병 줄기세포는 코로나바이러스 감염증을 나타내는 것을 특징으로 하는 방법. - 제1항에 있어서,
상기 NSP9 단백질은 처리된 줄기세포의 미토콘드리아 대사에 영향을 주는 것을 특징으로 하는 방법. - 제1항 내지 제4항 중 어느 한 항의 방법으로 제조된 질병 줄기세포 모델을 이용한, 항바이러스제의 스크리닝 방법.
- 제5항에 있어서,
상기 질병 줄기세포에 후보약물을 처리한 후, 미토콘드리아의 대사 변화를 분석하는 단계를 포함하는, 항바이러스제의 스크리닝 방법.
