Abstract
본원은 세포의 아미노산 결핍 조건에서 C/EBP-gamma 및 ATF가 이량체를 형성하여 자가포식 조절과 관련된 유전자 발현을 조절하는 신호전달체계를 표적으로 하는 자가포식 조절 방법, 또는 자가포식 조절제 스크리닝 방법을 개시한다. 본원에 따른 방법은 상기 기전 축을 중심으로 다양한 수준에서 자가포식 조절 문제로 인한 다양한 질환의 치료제 개발에 유용하게 사용될 수 있다.
Description
본원은 C/EBPgamma에 의한 특정 조건에서의 자가포식 조절을 규명한 분자 기전에 근거한 약물 스크리닝 기술과 관련된 분야이다.
자가포식은 세포내 항상성을 유지하기 위해 불필요한 단백질이나 손상된 소기관을 제거하는 작용으로 종간에 매우 잘 보존되어 있다. 영양 결핍 상황과 같은 세포 스트레스 상황에서 자가포식은 매우 높은 수준으로 증가하여 세포를 보호하는 역할을 수행한다.
이러한 자가포식은 불필요하거나 기능 장애가 있는 세포의 구성 요소를 제거하는 세포 내 분해 및 재활용 시스템이다. 영양 결핍과 같은 스트레스 상황에서 자가포식은 세포의 대사 항상성을 유지하기 위해 유도되고, 불충분한 영양소를 보충하기 위해 세포 내의 망가지고 불필요한 성분의 분해를 촉진한다. 따라서 자가포식이 제대로 일어나지 않는 경우 당뇨, 암, 신경퇴행질환과 같은 심각한 질병이 유발된다.
영양소 결핍은 영양소 유형에 따라 다른 메커니즘으로 감지된다. 아미노산 결핍은 GCN2(kinase general control nonderepressible-2)에 의해 인지되며, 이는 유리 아미노산이 낮은 상태일 때, 축적되는 모든 비하전 tRNA에 대해 높은 친화성을 갖는다. GCN2에 의해 일어나는 eIF2α(eukaryotic initiation factor 2α)의 인산화는 번역을 시작하기 위해 필요한 기능적 복합체의 형성을 감소시킨다. 그 결과 세포 내의 전체 단백질 번역이 억제되어 아미노산 결핍을 극복하는 과정이 진행되는 반면, mRNA의 하위 집합의 번역은 여전히 증가한다.
또한 전사 조절을 통해 아미노산 결핍을 극복하는 전략도 있다. mTORC1은 아미노산 및 자가포식의 가용성을 조정하는 핵심 허브이다. 아미노산 결핍 상황에서 mTORC1이 비활성화되어 ULK1 복합체가 활성화되며, 이 복합체는 자가포식을 시작하고 종료하는 데 중심적인 역할을 한다.
영양 결핍 상황을 극복하는 과정은 세포질에서의 변화뿐만 아니라 세포핵 내에서의 자가포식과 관련된 유전자 발현을 조절하는 것 또한 중요하다. TFEB, FOXO3a, p53, E2F1 및 CREB을 비롯한 많은 전사 인자들이 자가포식에 중요한 역할을 한다. 아미노산 결핍에 대응으로 TFEB은 핵으로 이동하여 불필요한 세포 소기관을 재활용하고, 아미노산의 가용성을 높이는 자가포식을 촉진한다. 포도당 결핍 상황에서 CARM1은 Pontin의 아르기닌 메틸화를 유도하며, 이는 FOXO3a의 TUDOR 도메인에 Pontin의 결합 능력을 부여한다. 따라서 포도당 결핍에 의해 형성된 FOXO3a-Pontin-Tip60 복합체는 리소좀 및 자가포식 유전자의 인핸서 활성화를 위해 작동한다.
그 동안 자가포식의 전사 조절은 영양 공급원을 구별하지 않고 주로 연구되었다. 자가포식의 전사 조절에 과정에 대한 이해를 향상시키고 이를 기반으로 한 약물 개발을 위해 영양소 결핍의 유형을 구분하여 자가포식 조절 연구 및 조절제의 개발이 필요하다.

본원은 아미노산 결핍 상황에서 자가포식 조절 기전 및 이에 기반한 자가포식 조절제 스크리닝 방법을 제공하고자 한다.
한 양태에서 본원은 아미노산이 결핍된 상태에서 특이적으로 자가포식을 조절하는 C/EBP-gamma (CCAAT Enhancer Binding protein)를 표적으로 하는 자가포식을 조절하는 물질을 스크리닝하는 방법을 제공한다.
일 구현예에에서 상기 방법은 아미노산이 결핍된 상태에서, C/EBP-gamma 및 ATF (Activating Transcription Factor) 단백질을 발현하는 세포를 배양하는 제 1 단계, 여기에서 상기 세포는, 상기 C/EBP-gamma 및 ATF 단백질이 이량체를 형성하고 상기 이량체가 결합할 수 있는 CARE (C/EBP-gamma-ATF Response Element, 반응요소) 및 상기 CARE에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 플라스미드를 포함하고; 상기 세포에 C/EBP-gamma 및 ATF의 이종 이량체 형성을 조절 또는 상기 이량체의 상기 CARE에 결합을 조절할 것으로 기대되는 시험물질을 처리하는 제 2 단계; 상기 세포에서 상기 리포터 유전자의 발현량을 측정하는 제 3 단계; 및 상기 측정결과 상기 시험물질로 처리되지 않은 대조군과 비교하여, 상기 시험물질이 처리된 세포에서 상기 유전자의 발현이 증가한 경우, 상기 시험물질을 자가포식을 증가시키는 후보물질, 또는 상기 유전자의 발현이 감소한 경우 상기 시험물질을 자가포식을 억제하는 후보물질로 선별하는 제 4 단계를 포함한다.

일 구현예에서 상기 방법에 사용될 수 있는 세포는 MEF (Mouse Embryonic Fibroblast) 또는 HepG2 세포를 포함한다.
자가포식은 영양분 결핍, 대사성 스트레스, 바이러스 감염, 노화 등 다양한 스트레스 상황에서 활성화되어 세포의 생존 및 항상성 유지에 중요한 역할을 하는 기작으로 자가포식 신호 조절에 문제가 생겼을 때 암과 퇴행성 뇌질환 등 여러 질환의 발생과 관련이 있다. 본원에서 규명된 아미노산 결핍 조건에서 특이적으로 작용하는 자가포식 조절 기전을 이용한 자가포식 조절제는 자가포식 조절 문제로 인한 다양한 질환의 치료제 개발에 유용하게 사용될 수 있다.

본원은 영양소 결핍의 유형에 의존하여 다르게 발현하는 유전자 및 자가포식 조절에 있어 C/EBPγ와 함께 ATF4가 아미노산 결핍 특이적으로 유도된 자가포식 과정에서 중요한 역할을 한다는 발견에 근거한 것이다.
본원에 따르면 C/EBPγ는 아미노산 결핍에 의해 유도된 자가포식 과정에서는 유전자 발현에 영향을 주었지만, 포도당 결핍에 의해 유도된 자가포식 과정에는 관여하지 않았다. 놀랍게도, C/EBPγ의 녹다운은 C/EBPγ 표적 유전자의 유도를 억제하고 아미노산 결핍 조건에서 자가포식 발생을 약화시켰다. 또한 본원에 따르면 C/EBPγ가 아미노산 결핍으로 유도된 자가포식 과정 동안 ATF4와 함께 유전자 발현을 조절한다는 것을 확인했다.
본원에서 자가포식(autophagy 또는 autophagocytosis)이란 라이소좀을 이용하여 세포내 불필요하거나 변성된 단백질을 포함하는 다양한 세포구성성분을 제거하는 이화작용을 일컫는 것이다. 자가포식 조절에 이상이 있는 경우, 변성단백질(misfolded protein)의 축적이 초래되어 이로 인해 신경변성질환이 발생하는 것으로 알려져 있다 (Komatsu et al., (2006), Nature, 441, 880-884). 또한 자가포식은 암세포에서 활성화 (Ding et al., (2009), Mol. Cancer Ther.,8(7), 2036-2045)되며, 자가포식 억제제가 항암 치료제로서 작용하는 것으로 알려져 있다 (Maiuri et al., (2007) Nat. Rev. Cell Biol. 8, 741-752). 아울러 암 및 신경변성 질환에 추가하여 자가포식은 간 질환, 심장 질환, 근육 질환 및 췌장 질환과 연관된 것으로 알려져 있다 (Levine and Kroemer, (2008), Cell, 132, 27-42; Fortunato and Kroemer, (2009), Autophagy, 5(6)). 따라서 본원에 개시된 기술을 이용한 자가포식 조절제, 조절방법은 자가포식 조절 이상으로 인한 다양한 질환의 치료 또는 예방에 효과적으로 사용될 수 있다.
이에 한 양태에서 본원은 C/EBP-gamma (CCAAT Enhancer Binding protein) 및 ATF (Activating Transcription Factor)를 표적으로 하는 자가포식을 조절하는 물질을 스크리닝하는 방법에 관한 것이다.
일 구현예에서 상기 방법은 아미노산이 결핍된 상태에서 C/EBP-gamma 및 ATF (Activating Transcription Factor) 단백질을 발현하는 세포를 배양하는 제 1 단계로, 상기 세포는, 상기 C/EBP-gamma 및 ATF 단백질이 이량체를 형성하고 상기 이량체가 결합할 수 있는 CARE (C/EBP-gamma-ATF Response Element) 및 상기 CARE에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 플라스미드를 포함하고, 상기 세포에 C/EBP-gamma 및 ATF의 이종 이량체 형성 또는 상기 이량체의 상기 CARE에의 결합을 조절할 것으로 기대되는 시험물질을 처리하는 제 2 단계; 및 상기 세포에서 리포터 유전자의 발현을 측정하는 제 3 단계를 포함하며, 상기 측정결과 상기 시험물질로 처리되지 않은 대조군과 비교하여, 상기 유전자의 발현 증가는 자가포식의 증가, 상기 유전자의 발현 감소는 자가포식의 억제를 나타낸다.

ATF (Activating Transcription Factor) 는 ATF 계열 단백질의 다양한 구성요소 중 하나로 오토파지 등 다양한 시스템에서 전사인자로 기능하는 것이 잘 알려져 있다. 특히, 아미노산 결핍 상황에서 빠르게 단백질로 발현하여 아미노산 결핍을 극복하기 위한 다양한 유전자들의 발현을 직접 조절하는 것으로 가장 잘 알려져 있다. 유전자 및 단백질 서열은 다음과 같이 공지되어 있다: Gene ID : 11911, NM_001287180.1, NP_001274109.1.
본원에 따른 방법은 진핵세포, 특히 동물세포, 예를 들면 포유류 유래 세포를 사용해서 수행될 수 있다. 일 구현예에서는 MEF (Mouse Embryonic Fibroblast) 또는 HepG2가 사용될 수 있다. 당업자라면 본원의 특징을 고려하여 적절한 세포를 선택할 수 있으며, 이러한 세포는 ATCC (American Type Culture Collection)과 같은 곳에서 입수가능하다.

일 구현예에서 본원에 따른 벡터는 CARE (C/EBP-gamma-ATF Response Element: 5‘ATTGCATCA3’) 및 상기 CARE에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 플라스미드이다. CARE는 프로모터에 C/EBP-gamma-ATF가 특이적으로 결합하게 되는 상기 서열을 의미하며, 프로모터의 업스트림에 위치할 수 있다.
본원에서 프로모터는 전사 조절 인자들이 결합하는 모든 DNA 염기서열 부위를 지칭하는 것으로, 일반적으로 전사를 조절할 대상이 되는 유전자의 유전정보를 지니고 있는 DNA 염기서열 앞부분에 위치한다. 진핵생물에서는 전사조절인자라고 하는 단백질들이 프로모터 부분에 결합함으로써 RNA 중합효소를 이끌고 오는 일에 관여한다. 프로모터는 이러한 전사 조절 인자들이 결합하는 모든 DNA 염기서열 부위를 지칭한다고 할 수 있다. 전사를 조절하는 DNA 염기서열은 프로모터 외에도 인핸서가 존재하나 프로모터가 전사시작지점으로부터 바로 앞쪽으로 수백 염기쌍 이내에 위치하는데 비해 인핸서는 프로모터로 부터 수천 염기쌍 이상 떨어져있다는 점에서 구분할 수 있다. 진핵세포에서 작동하는 프로모터는 HBV 프로모터, CMV(Cytomegalovirus) 프로모터, 아데노바이러스 후기 프로모터, 백시니아 바이러스 7.5K 프로모터, SV40(Simian Virus 40) 프로모터, SV40E1 프로모터, HSV(Herpes simplex virus)의 tk 프로모터, RSV(Respiratory syncytial virus) 프로모터, EF1(elongation factor-1 alpha) 알파 프로모터, 메탈로티오닌 프로모터, 베타-액틴 프로모터, 인간 IL-2(interleukin-2) 유전자의 프로모터, 인간 IFN(interferon) 유전자의 프로모터,인간 IL-4(interleukin-4) 유전자의 프로모터, 인간 림포톡신 유전자의 프로모터 또는 인간 GM-CSF(Granulocyte macrophage colony-stimulating factor) 유전자의 프로모터이다.
리포터 유전자는 외인성 유전자로 자연상태에서는 본원에 사용되는 조절서열과 연결되어 발견되지 않는 염기서열이다. 예를 들면 이러한 리포터 유전자는 LacZ, 클로람페니콜 아세틸 전이효소(CAT: chloramphenicol acetyltransferase), 레닐라 루시퍼라제(Renila luciferase), 반딧불이 루시퍼라제, 적색형광단백질(RFP), 녹색형광단백질(GFP), 분비성 태반 알칼라인 포스파타아제(secreted placental alkaline phosphatase, SEAP) 및 HSV-tk(Herpes simplex virus-thymidine kinase)를 들 수 있다.

단백질은 실질적으로 동일한 서열도 포함한다. 실질적으로 동일한이란, 단백질 및 핵산 서열 수준에 모두 적용되며, 참조 또는 기준이 되는 서열과 비교하여, 염기 또는 아미노산 잔기에 하나 이상의 치환, 결손, 또는 부가가 있을 수 있으나, 전체적으로 볼 때 기능에 차이가 없거나 기능을 나쁘게하지 않는 수준의 기능을 갖는 것을 의미한다. 상동성은 대상이 되는 서열을 최대한 대응되도록 얼라인하고, 당업계에서 통상적으로 이용되는 알고리즘을 이용하여 얼라인된 서열을 분석한 경우에, 최소 61%의 상동성, 보다 바람직하게는 70%의 상동성, 보다 더 바람직하게는 80%의 상동성, 가장 바람직하게는 90% 이상, 특히 95% 이상의 상동성을 나타내는 서열을 의미한다. 서열비교를 위한 얼라인먼트 방법은 당업계에 공지되어 있다. 예를 들면 Smith and Waterman, Adv. 본원에 따른 다양한 스크리닝 방법에서 사용되는 단백질의 양, 세포의 종류 및 시험물질의 양 및 종류 등은 사용하는 구체적인 실험방법 및 시험물질의 종류에 따라 달라지며, 당업자라면 적절한 양을 선택할 수 있을 것이다. 일 구현예에서 본원에 따른 방법은 상기 단백질을 발현하는 세포를 이용하여 수행될 수 있다. 세포를 이용하여 수행되는 경우 단백질은 이를 발현하는 세포로 제공될 수 있다.

본원에 사용되는 단백질은 당업계에 공지된 방법을 사용하여 제조될 수 있다. 특히 유전자 재조합 기술을 이용하는 것이다. 예를 들면 상기 단백질을 코딩하는 상응하는 유전자를 포함하는 플라스미드를 원핵 또는 진핵세포 세포 예를 들면 곤충세포, 포유류 세포에 전달하여 과발현 시킨 후 정제하여 사용할 수 있다. 상기 플라스미드는 예를 들면 본원의 예시적 구현예에서 사용한 것과 같은 동물 세포주에 트랜스팩션한 후, 발현된 단백질을 정제하여 사용할 수 있으나, 이로 제한하는 것은 아니다. 이 경우 단백질은 검출의 편이를 위해 다양한 표식물질, 예를 들면 바이오틴, 형광물질, 아세틸화, 방사선 동위원소와 같은 것으로 공지된 방법 또는 시중의 단백질 표지 키트를 사용하여 표지될 수 있으며, 표지된 물질에 적합한 검출기기를 사용하여 검출될 수 있다.
또는 스크리닝 단백질을 암호화하는 DNA 또는 RNA 서열을 적당한 숙주 세포에서 발현시켜 그 세포 파쇄물을 만들거나 상기 스크리닝 단백질의 mRNA를 시험관내에서 번역한 후 당업계에 공지된 단백질 분리 방법에 의해 스크리닝 단백질을 정제할 수 있다. 통상, 세포잔여물(cell debris) 등을 제거하기 위해 상기 세포 파쇄물 또는 시험관내 번역한 결과물을 원심분리한 후, 침전, 투석, 각종 컬럼 크로마토그라피 등을 적용한다. 이온교환 크로마토그라피, 겔-퍼미에이션 크로마토그라피, HPLC, 역상-HPLC, 프렙용 SDS-PAGE, 친화성 컬럼 등은 컬럼 크로마토그라피의 예이다. 친화성 컬럼은, 예를 들어, 항-스크리닝 단백질 항체를 이용하여 만들 수 있다.

또다른 구현예에서, 본원에 따른 방법이 사용될 수 있는 심혈관질환은 광동맥성심장병, 심근증, 고혈압성심장질환, 심부전, 폐성심, 심율동장애, 심장 내막염, 염증성 심비대증, 심근염, 심장판막증, 뇌혈관장애, 하지동맥질환, 선천성 심질환 및 심장 류머티즘으로 구성된 군으로부터 선택되는 심혈관질환이다.
또다른 구현예에서, 본원에 따른 방법이 사용될 수 있는 대사질환은 비만, 당뇨, 이상지방혈증, 지방간, 고혈압, 동맥경화, 고지혈증 및 고인슐린혈증으로 구성된 군으로부터 선택되는 대사질환이다.
또다른 구현예에서, 본원에 따른 방법이 사용될 수 있는 암은 뇌하수체선종, 신경교종, 뇌종양, 상인두암, 후두암, 흉선종, 중피종, 유방암, 폐암, 위암, 식도암, 대장암, 간암, 췌장암, 췌내분비종양, 담낭암, 음경암, 요관암, 신세포암, 전립선암, 방광암, 비호지킨성림프종, 골수이형성증을 포함한다.
이하, 본 발명의 이해를 돕기 위해서 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐 본 발명이 하기의 실시예에 한정되는 것은 아니다.
실시예
실험방법 및 재료
항체 및 시약
다음의 시판되는 항체를 사용하였다: 항-β-actin(A1978)(Sigma-Aldrich); 항-GFP(sc-9996), 항-ATF4(sc-390063) 및 항-C/EBPγ(sc-517003)(Santa Cruz biotechnology); 항-LC3(#2775)(Cell Signaling technology); 항-C/EBPγ(ab74045) 및 항-C/EBPβ(ab264306)(Abcam). 상업적으로 이용 가능한 형광 표지된 2차 항체가 사용됨: Alexa Fluor 488 donkey anti-rabbit IgG(A21206) 및 Alexa Fluor 594 donkey anti-mouse IgG(A21203)(Invitrogen). 종 및 용도에 대해 제조업체에서 권장하는 항체를 사용함. Bafilomycin A1(#11038)은 Cayman 제품이다.
세포 배양
MEF 세포는 5% CO2가 포함된 가습 인큐베이터에서 10% FBS와 ZelShield(Minerva Biolabs GmbH) 1% 항생제를 첨가한 Dulbecco’s modified Eagle’s 배지 (DMEM, Welgene)로 37℃에서 배양되었다. 연구에 사용된 MEF는 마이코플라스마 오염에 대해 정기적으로 테스트되었다. 아미노산 및 포도당 결핍의 경우, 세포를 PBS로 세척한 다음, 10% dialyzed FBS가 첨가된 모든 아미노산 및 포도당 고갈된 DMEM과 함께 배양하였다. 동시에 대조군 조건의 경우 세포를 일반 배지로 교체하여 배양하였다. Transfection은 Lipofectamine 3000(Invitrogen)으로 수행되었다.
면역형광
5 x 104 개 세포의 밀도로 1% 젤라틴이 코팅된 커버 슬립에서 키웠다. 커버슬립에서 성장한 세포를 PBS로 세척한 다음, PBS 중 2% 파라포름알데히드로 실온에서 10분 동안 고정했다. 고정된 세포는 실온에서 10분 동안 PBS-T(PBS중 0.5% Triton X-100)로 투과화 되었다. 1시간 동안 PBS-T에서 3% 소 혈청으로 blocking을 수행했다. 염색을 위해 세포를 항체와 함께 실온에서 2시간 동안 인큐베이션한 다음, 형광 표지된 2차 항체와 함께 1시간 동안 인큐베이션하였다. 염색이 끝난 세포는 공초점 현미경(Zeiss, LSM700)에 장착하고 시각화했다. 자가포식 연구를 위해 MEF를 GFP-LC3로 transfection시키고 커버슬립에 계대배양했다. 다음날, 세포를 보통의 배지 또는 아미노산/포도당 결핍 배지와 함께 배양하였다.
정량 RT-PCR
전체 RNA는 MEF세포에서 TRIzol (Invitrogen)을 이용하여 추출하고 분리되었다. RNA는 SuPrimeScript cDNA Synthesis Kit (GeNet Bio)을 사용하여 역전사되어 cDNA를 합성하였다. 얻어진 cDNA는 SYBR TOPrealTM qPCR 2X PreMix (Enzynomics)와 유전자 특이 프라이머로 PCR하였다. mRNA양은 ABI prism 7500 system 과 BioRad CFX384 system으로 검출하였다. mRNA의 양은 ddCt 방법을 사용하여 계산되었고 Hprt, Gapdh 및 β-actin이 대조군으로 사용되었다. 모든 반응은 3회 반복 수행되었다. PCR에 사용된 마우스 프라이머의 정보는 아래와 같다.

ChIP 실험
세포를 1% 포름알데히드에서 10분간 cross-link하고 차가운 PBS로 두 번 세척하였다. 세포를 1ml의 harvest buffer (0.1 M Tris-HCl [pH 9.4], 실험전에 10 mM DTT 추가)를 넣고 스크래퍼로 긁어서 1.5ml 튜브에 넣고 30℃에서 15분간 놓아둔 이후, 6000rpm에 3분 동안 원심분리하였다. 세포의 팰렛을 차가운 PBS로 세척한 이후, buffer I (0.25 % Triton X-100, 10 mM EDTA, 10 mM HEPES [pH 6.5], 및 0.5 mM EGTA) 과 buffer II (200 mM NaCl, 1 mM EDTA, 10 mM HEPES [pH 6.5], 및 0.5 mM EGTA)를 차례로 넣고 세척해 준다. ChIP lysis buffer (50 mM Tris-HCl [pH 8.1], 1 % SDS, 10 mM EDTA [pH 7.6], 실험 전에 protease inhibitor cocktail 추가)를 넣고 소니케이션을 통하여 크로마틴 절편을 만든다. 평균길이 250bp의 DNA 단편으로 만들어진 크로마틴 추출물을 dilution buffer (1 % Triton X-100, 2 mM EDTA, 150 mM NaCl, 20 mM Tris-HCl [pH 8.1], 실험 전에 protease inhibitor cocktail 추가)로 희석하고 항체를 넣어 4℃에서 밤새 면역침전 시켰다. 다음날 protein A/G sepharose 비드를 40ul씩 넣어주고 2시간동안 4℃에서 로테이션을 시켰다. 비드는 TSE I buffer (0.1 % SDS, 1 % Triton X-100, 2 mM EDTA, 20 mM Tris-HCl [pH 8.1], 및 150 mM NaCl), TSE II buffer (0.1 % SDS, 1 % Triton X-100, 2 mM EDTA, 20 mM Tris-HCl [pH 8.1], 및 500 mM NaCl), buffer III (0.25 M LiCl, 1 % NP-40, 1 % deoxycholate, 10 mM Tris-HCl [pH 8.1] 및 1 mM EDTA), 세번의 TE buffer (10 mM Tris-HCl [pH 8.0], 및 1 mM EDTA) 순서로 세척되었다. 그리고, elution buffer (1 % SDS, 0.1 M NaHCO3)를 사용하여 비드에서 용출시켰다. 용출된 추출물을 65℃에서 밤새 놔두어 reverse Cross-linking을 시키고 DNA는 QIA quick Gel Extraction Kit (QIAGEN)를 사용하여 정제되었다. 정제된 DNA는 quantitative RT-PCR을 통하여 분석되었다. 총 50ul의 DNA중 2 μl가 PCR에 사용되었다. 전자현미경
세포를 실온에서 1시간 동안 4% 글루타르알데히드와 1% 파라포름알데히드를 함유하는 0.1M 소듐 카코딜레이트를 사용하여 고정시켰다. 0.1M 나트륨 카코딜레이트로 3회 세척한 후 세포를 50% 에탄올에서 시작하여 100% 에탄올로 각 단계 20분씩 탈수했다. 그 후, 세포를 에탄올에 용해된 점진적으로 농축된 프로필렌 옥사이드와 함께 배양한 다음, 증가하는 농도의 Eponate 8 수지로 침투시켰다. 샘플을 65℃ 오븐에서 밤새 건조한 다음 Ultra microtom을 사용하여 절편을 만들고, 에너지 필터링 TEM 장치(LEO-192AB OMEGA, Carl Zeiss)로 절편을 관찰했다.
통계 및 재현성
모든 실험은 최소 3회 독립적으로 수행되었다. 통계 분석은 GraphPad Prism v5를 사용하여 수행되었다. 두 그룹 간의 비교에는 Student's t-test가 사용되었다. 사후 테스트를 통한 ANOVA 분석은 다중 샘플 비교 및 유의성 관계 식별에 사용되었다. <0.05의 p-값은 통계적으로 유의한 것으로 확인되었다.
실시예 1 C/EBPγ는 아미노산 유도 자가포식의 핵심 조절자임을 규명
본원에서는 우선 면역 블롯 분석을 수행하여 C/EBPγ 녹다운 후 자가포식 발생의 지표로서 지질화된 LC3-II 형성 수준의 변화를 확인하였다. 면역 블롯 분석은 아미노산 결핍(이하 AAS) 및 포도당 결핍(이하 GS) 에서 지질화되지 않은 LC3-I가 지질화된 LC3-II로 전환되었음을 보여주었다(도 1A). 그러나 MEF에서 C/EBPγ의 녹다운은 GS 조건이 아닌 AAS 조건 하에서 특이적으로 지질화된 LC3-II 수준을 약화시켰다(도 1A). 동시에, 자가포식 억제제인 Bafilomycin A1을 사용한 autophagic flux 분석은 LC3-II의 축적이 C/EBPγ 녹다운 MEF에서 대조군 MEF와 비교하여 AAS상황에서만 유의하게 약화되었음을 보여주었다(도 1B). 또한, 투과 전자 현미경 (TEM) 분석은 WT MEF에서 autophagosomes, autolysosomes 및 multilamellar body를 포함한 autophagic 소포의 수가 증가했지만 AAS상황에 대한 C/EBPγ 녹다운 MEF에서는 그렇지 않은 것으로 나타났다(보충 도 5). 그러나 C/EBPγ의 녹다운한 MEF에서 GS상황에서는 자가포식 과정에 전혀 영향을 미치지 않는 것으로 나타났다. 이러한 데이터는 C/EBPγ가 AAS 유도 자가포식 발생에서 중요한 역할을 한다는 것을 보여준다.
자가포식 과정에서 C/EBPγ의 역할을 추가로 확인하기 위해 녹색 형광 단백질(GFP) 태그된 LC3 양성 자가포식소체의 형성 양상을 확인하였다. GFP-LC3 점상 세포의 증가는 AAS에서 대조군 MEF와 비교하여 C/EBPγ 녹다운 MEF에서 유의하게 약화되었지만 GS가 유도된 자가포식의 경우에는 그렇지 않았다(도 1C). mCherry-GFP-LC3 리포터를 사용하여 LC3 반점 형성의 전체 수와 autophagic flux를 비교해보았다. GFP 신호는 산성 리소좀 조건에서 감쇠되고, 이 리포터는 리소좀 구획(노란색 점)과 산성 자가포식소체 소포(빨간색 점)와 융합되지 않은 자가포식 소포를 구분한다. AAS 시 빨간색 및 노란색 점의 전체 숫자는 대조군 MEF와 대조적으로 C/EBPγ 녹다운 MEF에서 현저하게 감소되었지만 GS는 그렇지 않았다(도 1D). 이러한 데이터는 AAS 유도 자가포식에 C/EBPγ가 필요함을 강력하게 나타낸다.
실시예 2. AAS 동안 C/EBPγ의 자가포식 기능을 위해서는 ATF4의 유도가 필요함을 규명
C/EBPγ 및 ATF4의 자가포식 과정에서의 역할을 추가로 조사하기 위해 잠재적 표적 유전자의 정량적 RT-PCR(qRT-PCR) 분석을 수행하고 들이 AAS 및 GS에 대한 자가포식에서 C/EBPγ 및 ATF4에 의해 공통으로 조절되는지 여부를 분석하였다. AAS와 GS는 모두 Map1lc3b, Bnip3, Vps26a, Gabarap, Gadd45b 및 Atg12를 포함한 자가포식 유전자의 mRNA 수준을 유도한 반면, C/EBPγ 또는 ATF4의 녹다운은 AAS에서 이들 유전자의 mRNA 수준을 유의하게 약화시켰지만 GS 조건에서는 그렇지 않았다. 도 2A, B). 이 데이터는 C/EBPγ와 ATF4가 AAS 유도 자가포식에 필요함을 나타낸다. 본원에서는 ATF4와 C/EBPγ 사이의 관계에 대해 더 의문을 제기했다. MEF에서 C/EBPγ의 녹다운하면 AAS 상황에서 ATF4의 발현에 영향을 미치지 않은 반면(도 2A), MEF에서 ATF4의 녹다운은 C/EBPγ 발현의 유도를 현저하게 약화시켰으며, 이는 ATF4가 C/EBPγ의 발현을 조절함을 나타낸다(도 2B). 본원에서는 또한 Cebpg 유전자 발현이 ATF4에 직접적으로 의존하는지 여부를 확인하기 위해 ChIP 분석을 수행했다. ChIP 분석은 ATF4가 AAS 3시간부터 Cebpg 프로모터에 결합하는 것을 보여주었다(보충 도 6). 그러나 C/EBPγ는 AAS 하에서 Cebpg 프로모터에 결합하지 않았다. 이러한 데이터는 ATF4가 C/EBPγ에 결합하지 않고 Cebpg 유전자 발현을 직접 조절함을 나타낸다.
실시예 3 C/EBPγ는 AAS 유도 자가포식 과정에서 표적 유전자의 전사 활성화를 위해 ATF4와 이종이량체를 형성하는 것을 규명
ATF4는 C/EBPγ와 이종이량체를 형성하여 C/EBP-ATF 반응 요소(CARE)에 결합한다. ATF4는 스트레스 반응 시 C/EBPγ에 결합하지만 지방 생성 동안에는 C/EBPβ와 결합한다. 면역 블롯 분석은 ATF4가 AAS상황에서 C/EBPγ와 결합하지만 C/EBPβ와는 결합하지 않는 다는 것과 GS상황에서는 ATF4가 C/EBPγ와도 결합하지 못하는 것으로 나타났다(도 3A).
C/EBPγ-ATF4 복합체가 AAS로 유도된 자가포식 유전자의 활성화를 위해 ATF4와 함께 표적 프로모터에 결합되는지 여부를 확인하기 위해서 항-C/EBPγ, ATF4 및 C/EBPβ 항체를 사용하여 C/EBPγ 표적 유전자의 CARE에 대한 ChIP 분석을 수행하였다. 그 결과 ChIP 분석은 C/EBPγ-ATF4 복합체가 AAS에서 Map1lc3b, Bnip3, Atg12, Gadd45b 및 Vps26a를 포함한 표적 유전자의 CARE에 결합되었지만 GS 조건에서는 그렇지 않은 것으로 나타났다(도 3B, C). 놀랍게도, MEF에서 C/EBPγ의 녹다운은 AAS에서 ATF4의 타겟 프로모터에 결합을 완전히 손상시켰지만 GS로 유도된 자가포식의 경우에는 그렇지 않았다(도 3B, C). 함께, 이러한 데이터는 C/EBPγ-ATF4 이종이량체가 autophagy 유전자의 CARE에서 결합되고 C/EBPγ가 ATF4와 함께 autophagy의 AAS 특이적 핵심 조절자로서 기능한다는 것을 나타낸다(도 4).
자가포식은 영양 및 에너지 결핍에 대한 반응으로 리소좀에 의해 세포질 단백질 및 세포 소기관의 분해하는 고도로 보존된 세포 내 과정이다. 따라서 AAS 및 GS를 포함한 영양소 결핍에 반응하는 세포의 능력은 대사 항상성과 생존력의 유지에 필수적이다. 영양소 결핍의 인지는 세포질의 영양소 공급원에 따라 다양한 신호 전달 경로를 유발한다. 또한 핵에는 영양소 결핍 의존적 전사 메커니즘이 있는 것으로 추측되고 있으나, autophagy 과정에서 핵 전사 메커니즘을 논의할 때 영양소 의존성은 이전까지 심각하게 고려되지 않았다.
본원에서는 자가포식을 유도하는 특정 상위 영양소 신호가 핵에서 autophagy의 특정 하위 조절자를 결정할 것이라고 가정했다. 다양한 상위 영양소 신호에 대한 반응으로 자가포식에서 핵 내에서 전사조절의 중요성을 확인하기 위해 본원에서는 GS 및 AAS에 의해 MEF에서 자가포식을 유도하고 각 조건에 대한 특정 전사인자를 식별하고자 했다. ATF4가 AAS 상황 하에서 자가포식 유전자를 활성화하기 위해 C/EBPγ의 도움이 필요하다는 것은 매우 흥미로운 결론이다. ATF4는 GS 및 AAS 유도 autophagy 모두에서 상향 조절되지만 ATF4는 AAS에서만 C/EBPγ의 발현을 유도한다. 또한, 본원의 결과는 ATF4 및 C/EBPγ의 이종이량체가 AAS 상황 특이적으로 자가포식 과정에 관여하는 유전자의 발현을 유도하지만, GS에 의해 유도된 자가포식을 유도하지 않는다는 것을 보여준다.
이전까지의 자가포식 과정에 대한 연구는 상위 신호 특이적 유전자 조절과 연관된 통제된 실험의 부족으로 인해 제대로 연구되지 않았다. 본원의 연구는 C/EBPγ를 AAS 특이적 전사인자로 식별하였고, 이 발견은 영양 결핍 유형 특이적 유전자 전사조절의 사례를 제공하며, 자가포식을 위한 상위 신호 특이적 전사 조절자가 추가로 존재할 수 있는 가능성을 함께 제시한다.
이상에서 본원의 예시적인 실시예에 대하여 상세하게 설명하였지만 본원의 권리범위는 이에 한정되는 것은 아니고 다음의 청구범위에서 정의하고 있는 본원의 기본 개념을 이용한 당업자의 여러 변형 및 개량 형태 또한 본원의 권리범위에 속하는 것이다.
본 발명에서 사용되는 모든 기술용어는, 달리 정의되지 않는 이상, 본 발명의 관련 분야에서 통상의 당업자가 일반적으로 이해하는 바와 같은 의미로 사용된다. 본 명세서에 참고문헌으로 기재되는 모든 간행물의 내용은 본 발명에 도입된다.
Claims (4)
- C/EBP-gamma (CCAAT Enhancer Binding protein)를 표적으로 하는 자가포식을 조절하는 물질을 스크리닝하는 방법으로, 상기 방법은
아미노산이 결핍된 상태에서 C/EBP-gamma 및 ATF (Activating Transcription Factor) 단백질을 발현하는 세포를 배양하는 제 1 단계로, 상기 세포는, 상기 C/EBP-gamma 및 ATF 단백질이 이량체를 형성하고 상기 이량체가 결합할 수 있는 CARE (C/EBP-gamma-ATF Response Element) 및 상기 CARE에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 플라스미드를 포함하고,
상기 세포에 상기 C/EBP-gamma 및 ATF의 이종 이량체 형성을 조절하거나 또는 상기 이량체의 상기 CARE에의 결합을 조절할 것으로 기대되는 시험물질을 처리하는 제 2 단계;
상기 세포에서 상기 리포터 유전자의 발현을 측정하는 제 3 단계; 및
상기 측정결과 상기 시험물질로 처리되지 않은 대조군과 비교하여, 상기 시험물질이 처리된 세포에서 상기 유전자의 발현이 증가한 경우, 상기 시험물질을 자가포식을 증가시키는 후보물질, 또는 상기 유전자의 발현이 감소한 경우 상기 시험물질을 자가포식을 억제하는 후보물질로 선별하는 제 4 단계를 포함하는, 자가포식 조절제 스크리닝 방법.

상기 세포에 C/EBP-gamma 및 ATF의 이종 이량체 형성을 조절할 것으로 기대되는 시험물질을 처리하는 제 2 단계;
상기 세포에서 자가포식의 활성과 함께 발현이 증가하는 자가포식 유전자의 발현을 측정하는 제 3 단계; 및
상기 측정결과 상기 시험물질로 처리되지 않은 대조군과 비교하여, 상기 시험물질이 처리된 세포에서 상기 유전자의 발현이 증가한 경우 상기 시험물질을 자가포식을 증가시키는 후보물질, 또는 상기 유전자의 발현이 감소한 경우 상기 시험물질을 자가포식을 억제하는 후보물질로 선별하는 제 4 단계를 포함하는, 자가포식 조절제 스크리닝 방법.
- 제 1 항 또는 제 2 항에 있어서,
상기 세포는 MEF (Mouse Embryonic Fibroblast) 또는 HepG2 세포인, 자가포식 조절제 스크리닝 방법.
- 제 2 항에 있어서,
상기 자가포식 유전자는 Map1lc3b (Microtubule-associated proteins 1A/1B light chain 3B), Vps26a (Vacuolar protein sorting-associated protein 26A) Gabarap(Gamma-aminobutyric acid receptor-associated protein), Gadd45b (Growth arrest and DNA-damage-inducible beta) 또는 Atg12(Autophagy related 12)를 포함하는, 자가포식 조절제 스크리닝 방법.
