본 발명은 대장암 진단 또는 예후 예측용 바이오마커로서 HJURP(Holliday Junction Recognition Protein)의 용도에 관한 것이다.
Holliday Junction Recognition Protein (HJURP, hFLEG1로도 알려짐)은 동원체(centromeres)에서 histone H3-유사 변이체 동원체 단백질-A (CENP-A)의 통합 및 유지에 중추적인 역할을 하는 동원체 단백질이다. 이는 CENP-A의 특이 샤프론이며 복제된 동원체에서 새로 합성된 CENP-A 분자 및 뉴클레오솜과 통합한다. 이의 동원체 국소화는 세포 주기에 의해 조절되며, 및 동원체에서 이의 짧은 등장은 새로운 CENP-A 침적과 동시에 일어난다. 그러므로, HJURP는 동원체에서 CENP-A 침적 및 유지에 중요한 요소이다. 암의 개시 및 진행에서 히스톤 샤프론의 잘못된 조절의 역할이 연구되고 있다. 뿐만 아니라, HJURP가 폐암에서 과발현함이 보고되었으며, 루미날 A 유방암의 예후 마커로서 제시되고 있다. 그러나, 대장암에서의 이의 역할은 아직 알려지지 않았다.
대장암(CRC)은 가장 일반적인 악성종양 중 하나로서, 전세계적으로 사망의 주요 원인인 전이로 인한 암-관련 사망을 야기한다. CRC의 원격 전이는 진단시 환자 중 약 25%에서 나타났다. 원격 전이 대장암 환자에서, 평균 생존률은 10개월 미만이고, 화학요법을 받고 최상의 지원 치료를 받은 경우에는 24개월까지 늘어날 수 있다; 그러나, 원격 전이를 갖는 CRC 환자의 생존률은 여전히 낮다. 그러므로, CRC 전이의 초기 진단 및 예상이 CRC 환자의 생존률을 높일 수 있다. CRC의 초기 검출은 이의 임상조직학적 인자의 이해력을 높이고, 외과적 수술 단독 또는 화학요법 또는 방사선 요법과의 병행시 환자의 생존률을 증가시킬 수 있다. 그러나, 통상적인 조직학적 염색을 사용하여 재발, 전이, 화학요법에 대한 내성 및 감소된 생존률에 대해 높은 위험성을 갖는 환자를 예상할 수 있는 마커는 없다. 그러므로, 대장암에서의 신규한 예후 바이오마커의 확인은 효과적인 표적 치료를 개선시키는데 필수적이다.
이에, 본 발명의 발명자는 대장암에서 HJURP 발현의 병리학적 평가 및 4개의 대장암 세포에서 siRNA에 의한 HJURP 녹다운의 기능적 평가를 통해 예후 바이오마커로서의 효과를 확인하여 본 발명을 완성하였다.
이에 따라 본 발명은 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정할 수 있는 제제를 포함하는 대장암 진단 또는 예후 예측용 조성물 및 HJURP의 발현수준을 측정하여 전립선암의 진단 또는 예후 예측을 위한 정보를 제공하는 방법을 제공하는 것이다.
상기와 같은 본 발명의 목적을 달성하기 위해서, 본 발명은 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정할 수 있는 제제를 포함하는, 대장암 진단 또는 예후 예측용 조성물을 제공한다.
또한, 본 발명은 본 발명에 따른 대장암 진단 또는 예후 예측용 조성물을 포함하는, 대장암 진단 또는 예후 예측용 키트를 제공한다.
또한, 본 발명은 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계; 상기 발현수준을 정상 대조군과 비교하는 단계; 및 상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암이 있는 것으로 판단하는 단계;를 포함하는, 대장암 진단을 위한 정보 제공 방법을 제공한다.
또한, 본 발명은 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계; 상기 발현수준을 정상 대조군과 비교하는 단계; 및 상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암 악화 위험성이 높은 것으로 판단하는 단계;를 포함하는, 대장암 예후 예측을 위한 정보 제공 방법을 제공한다.
또한, 본 발명은 HJURP(Holliday Junction Recognition Protein)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는, 대장암 치료용 약학적 조성물을 제공한다.
본 발명은 HJURP를 바이오마커로 사용하여 대장암의 발생, 재발, 전이, 화학요법에 대한 내성 및 감소된 생존률에 대해 높은 위험성을 갖는 환자를 용이하게 예상할 수 있다.

본 발명은 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정할 수 있는 제제를 포함하는, 대장암 진단 또는 예후 예측용 조성물에 관한 것이다.
본 발명에서 사용되는 용어 "마커 또는 진단 마커(diagnosis marker)"란 전립선암을 가진 개체를 정상 세포 또는 정상 개체와 구분하여 진단할 수 있는 물질로, 정상 세포에 비하여 전립선암이 진행 또는 발병된 세포 또는 개체에서 증가 또는 감소를 보이는 폴리펩티드, 단백질 또는 핵산(예: mRNA 등), 지질, 당지질, 당단백질 또는 당(단당류, 이당류, 올리고당류 등) 등과 같은 유기 생체 분자들을 포함한다. 본 발명의 목적상, 본 발명의 전립선암 진단 마커는 정상 세포 또는 조직의 세포에 비하여, 전립선암 세포에서 특이적으로 발현 수준의 차이를 보이는 miRNA 또는 해당 miRNA의 단편이다.
본 발명에서 사용되는 용어 "miRNA 또는 이의 단편의 수준을 측정하는 제제"란, 시료에 포함된 해당 miRNA 또는 이의 단편의 발현 여부를 확인하는 방법에 사용되는 제제를 의미하는데, 바람직하게는 RT-PCR, 경쟁적 RTPCR(Competitive RT-PCR), 실시간 RT-PCR(Real-time RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블럿팅(Northern blotting), 유전자 칩 분석법 등의 방법에 사용되는 표적 유전자에 특이적으로 결합할 수 있는 프라이머, 프로브, 안티센스 올리고뉴클레오티드 또는 항체가 될 수 있으나, 특별히 이에 제한되지는 않는다.
본 발명에서 사용되는 용어 "프라이머"란, 짧은 자유 3 말단 수산화기(free 3' hydroxylgroup)를 가지는 핵산서열로 상보적인 템플레이트(template)와 염기쌍(base pair)을 형성할 수 있고 템플레이트 가닥 복사를 위한 시작지점으로 기능을 하는 짧은 핵산 서열을 의미한다. 프라이머는 적절한 완충용액 및 온도에서 중합반응(즉, DNA 폴리머레이즈 또는 역전사효소)을 위한 시약 및 상이한 4가지 뉴클레오사이드 트리포스페이트의 존재하에서 DNA 합성을 개시할 수 있다.
본 발명에서 사용되는 용어 "프로브"란, 유전자 또는 mRNA와 특이적 결합을 이룰 수 있는 짧게는 수 염기 내지 길게는 수백 염기에 해당하는 RNA 또는 DNA 등의 핵산 단편을 의미하는데, 올리고뉴클레오티드(oligonucleotide) 프로브, 단쇄 DNA(single stranded DNA) 프로브, 이중쇄 DNA(double stranded DNA) 프로브, RNA 프로브 등의 형태로 제작될 수 있고, 보다 용이하게 검출하기 위하여 라벨링될 수 있다.
상기 단백질의 발현 수준을 측정할 수 있는 제제는 상기 HJURP에 특이적으로 결합하는 항체, 엡타머, 올리고펩타이드 또는 PNA(Peptide nucleic acid), 또는 상기 단백질을 코딩하는 유전자에 특이적인 상보적 서열을 갖는 프라이머 또는 프로브를 포함할 수 있으나, 이에 제한되지 않는다.
또한, 본 발명은 본 발명에 따른 대장암 진단 또는 예후 예측용 조성물을 포함하는, 대장암 진단 또는 예후 예측용 키트에 관한 것이다.
상기 키트는 RT-PCR 키트, DNA 칩 키트 또는 단백질 칩 키트를 포함할 수 있으나, 이에 제한되지 않는다.
본 발명의 키트는 HJURP 유전자의 mRNA 또는 이로부터 발현되는 단백질의 검출여부를 확인함으로써 대장암의 진단과 예후를 예측할 수 있다. 본 발명의 대장암 진단 또는 예후를 예측하기 위한 키트에는 HJURP 유전자의 발현 수준을 검출하기 위한 제제(예컨대, 프라이머 또는 프로브) 또는 단백질을 특이적으로 검출할 수 있는 제제(예컨대, 항체 또는 압타머) 뿐만 아니라 분석 방법에 적합한 한 종류 또는 그 이상의 다른 구성 성분 조성물, 용액 또는 장치가 포함될 수 있다.
본 발명에서 HJURP 유전자의 mRNA 발현 수준을 측정하기 위한 키트는 RT-PCR을 수행하기 위해 필요한 필수 요소를 포함하는 키트일 수 있다. RT-PCR 키트는, 마커 유전자에 대한 특이적인 각각의 프라이머 쌍 외에도 테스트 튜브 또는 다른 적절한 컨테이너, 반응 완충액(pH 및 마그네슘 농도는 다양), 데옥시뉴클레오타이드(dNTPs), Taq-중합효소 및 역전사효소와 같은 효소, DNase, RNase 억제제, DEPC-수(DEPC-water), 멸균수 등을 포함할 수 있다. 또한 정량 대조구로 사용되는 유전자에 특이적인 프라이머 쌍을 포함할 수 있다.
또한, 본 발명의 키트는 DNA 칩을 수행하기 위해 필요한 필수 요소를 포함하는 진단 또는 예후용 키트일 수 있다. DNA 칩 키트는, 유전자 또는 그의 단편에 해당하는 cDNA가 프로브로 부착되어 있는 기판, 및 형광표식 프로브를 제작하기 위한 시약, 제제, 효소 등을 포함할 수 있다. 또한, 기판은 정량 대조구 유전자 또는 그의 단편에 해당하는 cDNA를 포함할 수 있다.
또한, 본 발명의 키트가 HJURP 단백질의 발현 수준을 측정하기 위한 키트일 경우, 항체의 면역학적 검출을 위하여 기질, 적당한 완충용액, 발색 효소 또는 형광물질로 표지된 2차 항체, 발색 기질 등을 포함할 수 있다. 상기에서 기질은 니트로셀룰로오스 막, 폴리비닐 수지로 합성된 96 웰 플레이트, 폴리스티렌 수지로 합성된 96 웰 플레이트 및 유리로 된 슬라이드글라스 등이 이용될 수 있고, 발색효소는 퍼옥시다아제(peroxidase), 알칼라인 포스파타아제(Alkaline Phosphatase)가 사용될 수 있고, 형광물질은 FITC(Fluorescein isothiocyanate), RITC(Rhodamine B isothiocyanate) 등이 사용될 수 있고, 발색 기질액은 ABTS(2,2'-아지노-비스(3-에틸벤조티아졸린-6-설폰산)) 또는 OPD(o-페닐렌디아민), TMB(테트라메틸벤지딘)가 사용될 수 있다.
또한, 본 발명은 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계; 상기 발현수준을 정상 대조군과 비교하는 단계; 및 상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암이 있는 것으로 판단하는 단계;를 포함하는, 대장암 진단을 위한 정보 제공 방법에 관한 것이다.
또한, 본 발명은 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계; 상기 발현수준을 정상 대조군과 비교하는 단계; 및 상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암 악화 위험성이 높은 것으로 판단하는 단계;를 포함하는, 대장암 예후 예측을 위한 정보 제공 방법에 관한 것이다.
상기 시료는 조직, 세포, 전혈, 혈청, 혈장, 타액, 객담, 뇌척수액 또는 소변 등을 포함할 수 있으나, 이에 제한되지 않는다.
또한, 본 발명은 1) 후보물질을 HJURP(Holliday Junction Recognition Protein) 또는 이의 단편을 발현하는 세포주에 처리하는 단계; 2) 후보물질이 처리된 세포주에서 상기 HJURP 또는 이의 단편의 발현 정도를 측정하는 단계; 및 3) 후보물질을 무처리한 대조군 세포주와 비교하여 HJURP 또는 이의 단편의 발현량이 감소된 후보물질을 선별하는 단계를 포함하는 대장암 치료제 후보물질의 스크리닝 방법을 제공한다.
또한, 본 발명은 1) 대조군인 대장암이 발병된 실험동물의 시료로부터 HJURP 또는 이의 단편의 발현 수준을 측정하는 단계; 2) 상기 실험동물에 대장암을 치료할 수 있을 것으로 예상되는 후보물질을 투여하는 단계; 3) 실험군인 상기 후보물질이 투여된 실험동물의 시료로부터 HJURP 또는 이의 단편의 발현 수준을 측정하는 단계; 및, 4) 대조군에서 측정된 HJURP 또는 이의 단편의 수준을 실험군에서 측정된 것과 비교하여, 대조군에서 측정된 것보다도 실험군에서 측정된 것이 낮은 수준을 나타낼 경우 후보물질을 선별하는 단계를 포함하는 대장암 치료제 후보물질의 스크리닝 방법을 제공한다.
또한, 본 발명은 HJURP(Holliday Junction Recognition Protein)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는, 대장암 치료용 약학적 조성물에 관한 것이다.
상기 활성 억제제는 HJURP에 특이적으로 결합하는 항체, 화합물, 펩티드, 펩티드 미메틱스 및 앱타머 등을 포함할 수 있으나, 이에 제한되지 않는다.
상기 발현 억제제는 HJURP 단백질을 코딩하는 유전자의 mRNA에 상보적으로 결합하는 안티센스 올리고뉴클레오티드, siRNA(small interfering RNA), shRNA(small hairpin RNA) 및 리보자임(ribozyme) 등을 포함할 수 있으나, 이에 제한되지 않는다.
이하 본 발명을 실시예에 의하여 더욱 상세하게 설명한다. 하기 실시예는 단지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 국한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
<실시예>
실시예 1. 실험
세포주 및 배양
HCT116, HT29, SW480, 및 SW620을 비롯한 인간 대장 세포주를 Korean Cell Line Bank (KCLB, Seoul, Korea)에서 구입하였다. 각 세포는 5% CO2,37°C의 완전 습식 인큐베이터 내에서 10% fetal bovine serum (FBS, Thermofisher) 및 1% penicillin/streptomycin (Gibco, UT, USA)가 보충된 RPMI1640 (ThermoFisher, Waltham, MA, USA)에 배양하였다.
siRNA에 의한 HJURP의 사일런싱(Silencing)
HJURP에 대한 특이 siRNA를 OriGene (Rockville, MD, USA, #SR310705)에서 구입하였다. 6-웰 플레이트에 웰 당 약 1 x 105 cells의 세포를 접종하고 24시간 동안 인큐베이션한 후, 트랜스펙션 12시간 전에 무혈청 배지로 배양하였다. 그런 다음, 세포를 무혈청 배지에서 HiPerFect transfection reagent (Qiagen, IN, USA, #301705)를 사용하여 HJURP-siRNA로 트랜스펙션시켰다. 36시간 후 세포를 수집하여 RNA 및 단백질을 분리하고 HJURP 녹다운을 확인하였다. 실험은 3회 반복 실시하였다.
역전사 PCR
RioEx reagent (Geneall, Seoul, Korea)를 사용하여 배양된 세포로부터 전체 RNA를 추출하였다. ReverTra Ace qPCR kit (Toyobo, Osaka, Japan)를 사용하여 전체 RNA로부터 cDNA를 역-전사하고 GAPDH를 내부 대조로 사용하였다. PCR 증폭 프로그램은 하기와 같으며, 총 35사이클을 실시하였다: pre-denaturation at 94℃ for 5 min, denaturation at 94℃ for 1 min, annealing of HJURP and GAPDH performed at 58 ℃ for 30 sec, and a final extension performed at 72 ℃ for 1 min. 사용된 올리고뉴클레오티드 프라이머는 다음과 같다 s: HJURP forward, 5′- CAGATGGGGTGGACAACAC-3′; HJURP reverse, 5′- TCTTCCATCCTGTAAGACGTG-3′; GAPDH forward: 5′- CTTAGCACCCCTGGCCAAG-3′; GAPDH reverse: 5′- GATGTTCTGGAGAGCCCCG-3′. 예상된 PCR 산물 크기는 HJURP가 100 bp이고 GAPDH가 154 bp이었다. PCR 산물을 2% 아가로스겔 상에 전기영동하고 에티디움 브로마이드 염색으로 10분간 시각화시켰다. 모든 분석은 3회 반복 실시하였다.
웨스턴 블롯팅 검정
Pro-PREPTM protein extraction solution(INtRON,Seongnam,Korea)에서 세포를 용균하였다. 단백질을 xMark Microplate Absorbance Spectrophotometer (Bio-Rad Laboratories, Inc., CA, USA)을 사용하여 bicinchoninic acid (BSA, Sigma, Yongin, Korea) 검정으로 정량하였다(30 μg/lane). 단백질을 10% sodium dodecyl sulfate-polyacrylamide gel (SDS-PAGE) 전기영동하고 Immobilon polyvinylidene difluoride membranes (Millipore, MA, USA)으로 옮겼다. 옮겨진 맴브레인을 5% 탈지유로 1시간 동안 차단하고 1:300로 희석된 항-인간 HJURP 폴리클로날 항체(Sigma-Aldrich, St. Louis, MO, USA)와 함께 4℃에서 밤새 인큐베이션한 후, 1:2000로 희석된 이차 항체와 함께 실온에서 2시간 동안 다시 인큐베이션하였다. Molecular Imager ChemiDoc XRS+ System (Bio-Rad Laboratories, CA, USA)을 통해 ECL western detection reagents (Advansta, CA, USA)으로 신호를 검출하였다.
세포 증식 검정
3-(4,5-dimethlthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) 검정을 96-well (3 x 104 cells/well)배양 플레이트에서 실시하였다. 세포를 37 °C 및 5% CO2에서 24, 48, 및 72 시간 동안 인큐베이션하였다. 그런 다음, 10 μL MTT (5 mg/mL)를 웰에 첨가한 후 2시간 동안 인큐베이션하였다. 상청액을 버리고 100 μL의 dimethyl sulfoxide (DMSO)를 각 웰에 첨가해 침전물을 용해시켰다. 570 nm에서 흡광도를 측정하여 생존도를 정량하였다
세포 침습 및 이동 검정
세포 침습 및 이동 검정을 Transwell Permeable Supports 6.5 mm Insert (Cosar, Corning, NY, USA) 및 성장 인자 감소 매트리겔(Matrigel) (BD Biosciences, San Jose, CA, USA)을 사용해 실시하였다. 지지체를 1 시간 동안 37℃에서 인큐베이션하고 매트리겔로 덮었다. 5 x 105 cells/well의 밀도로 세포를 첨가하고 10% FBS를 포함하는 750 μL 배양 배지를 하부 챔버에 첨가했다. 48시간 동안 인큐베이션한 후, 트랜스웰에서 배지를 제거하고 냉 PBS로 세척한 후 3.7% 파라포름알데하이드로 세포를 고정하였다. 세포를 0.5% 크리스탈 바이올렛으로 염색하고, 상부 맴브레인의 매트리겔을 제거하였다. 세포수를 현미경으로 계수하였다. 이동 검정은 매트리겔을 사용하지 않는 것을 제외하고는 유사한 방식으로 실시하였다.
소프트 아가 콜로니 형성 검정
6-웰 플레이트를 20% fetal bovine serum (FBS)가 보충된 RPMI1640 배지 중의 0.5% 아가로스로 코팅하였다. HJURP 녹다운 세포 또는 대조 세포(각 세포 수, 2.5 x 103 cells/mL)를 0.35% 아가를 함유한 RPMI1640 배지와 혼합하고 6-웰 플레이트 내 0.5% 아가 상에 덮어씌웠다. 플레이트를 콜로니가 형성될 때까지 37℃, 5% CO2에서 14일 동안 인큐베이션하였다. 그런 다음, 각 웰을 0.01% 크리스탈 바이올렛으로 염색하고 콜로니수를 계수하고 사진촬영하였다.
면역조직화학(Immunohistochemistry; IHC)
2002년 1월부터 2009년 12월까지 순천향대 천안 병원에서 수술을 받고 대장암이 있는지를 조직병리학적으로 확인한 환자로부터 162개 샘플을 수득하였다. 환자 데이터는 표 1에 나타냈다.
대장암 조직의 파라핀-침전 블록을 5 μM 두께 섹션으로 절단하고, 슬라이드를 수습하여 인간 HJURP 항체(Sigma-Aldrich, St. Louis, MO, USA)와 함께 4℃에서 밤새 인큐베이션하였다. 그런 다음 Tween® 20(TBST)과 Tris-buffered saline으로 세척하고 EnVision+ System-HRP Labelled Polymer Anti-Rabbit (Dako, CA, USA)을 사용하여 이차 항체와 함께 실온에서 30분간 인큐베이션하였다. 슬라이드를 70 μL 3′-3-diaminobenzidine (DAB, Dako, CA, USA)로 처리하고 현미경으로 관찰하였다. 면역조직화학적 파라미터를 반-정량 광학 분석에 따라 맹검 방식으로 3개의 독립 검사자(investigators)로 등급화하고 콘센서스 스코어(consensus score)를 각 시료에 대해 결정하였다. HJURP의 정량은 염색된 종양 세포의 백분율 및 강도를 평가하여 실시하였다. 염색 백분율은 다음과 같이 점수를 매겼다: 0, 0-5%; 1, 5-25%; 2, 25-50%; 3, 50-75%; 및 4, 75-100%. 염색 강도는 다음과 같음 방식으로 점수를 매겼다: 0, negative; 1, weak; 2, moderate; 3, strong. 그런 다음 최종 스코어를 저발현 또는 고발현 그룹으로 나누었다 (low: scores < 2; high: score ≥ 2).
통계 분석
통계 분석은 SPSS v19 program using Student’s T-test를 사용하여 실시하였다. 임상병리학적 변수의 95% confidence interval (CI) 및 hazard ratio (HR)를 단변량 및 다변량 Cox regression model을 사용하여 평가하였다. Kaplan-Meier 방법은 환자의 HJURP 발현 및 결과 사이의 관련성을 분석하기 위해 사용하였다. log-rank test는 전체 생존률을 평가하기 위해 사용하였다. 모든 테스트에서, P-value < 0.05는 통계학적으로 유의한 것이다.
실시예 2. 결과
대장암 환자에서 HJURP 발현 및 암-특이 사망 사이의 연관성
CRC 시료에서의 HJURP 발현을 평가하기 위해 IHC를 사용하였으며, 브라운 염색에 의해 확인되는 바와 같이 양성 발현이 세포질 및/또는 세포막에서 발견되었다(도 1). 전체적으로, 환자로부터의 162개 케이스의 CRC 시료를 사용하였다. 표 1에서 볼 수 있는 바와 같이, 높은 HJURP 발현이 44개 CRC 조직에서 관칠되었으며(27.2%), 118개 CRC 조직(82.8%)에서는 낮은 HJURP 발현을 나타냈다. 높은 HJURP 발현은 병리학적 인자와 유의한 관련성이 없었다. 도 1에서 볼 수 있는 바와 같이, 정상 상피에서는 약한 염색 또는 염색이 발견되지 않았으며, CRC 조직에서 HJURP의 중간 또는 강한 면역 염색이 발견되었다.
단변량 분석에 따르면 단계, 전이, 림프관 침습, 및 HJURP 발현은 대장암 환자의 5년 생존률에 유의한 영향을 갖는다. 유의한 인자에 대해 조정된 다변량 콕스 회기 분석(multivariate Cox regression analysis)을 5년 생존률을 평가하기 위해 사용하였다. 단계(HR = 1.771; 95% CI = 1.087-2.886; P = 0.022), 전이 (HR = 3.559; CI = 1.437-8.813; P=0.006), 림프관 침습 (HR = 1.573; CI = 0.954-2.592; P=0.076), 및 HJURP 발현 (HR = 1.880; CI = 1.167-3.027; P = 0.009)은 5년 생존률의 독립 예측 변수였다(표 2). 또한 HJURP 발현에 대해 양성 및 음성을 보이는 환자를 그룹화하여 콕스 회기 분석을 실시하였다. 콕스-회기 분석 결과는 두 그룹 사이에 유의차를 보였다(p < 0.05, Univariate; HR = 1.458; CI = 1.100-1.932; P = 0.009, Multivariate; HR = 1.724; CI = 1.032-2.880, p = 0.037). 이는 HJURP의 발현이 대장암 환자의 생존률을 감소시킴을 제시한다.
도 2에서 볼 수 있는 바와 같이,Kaplan-Meier로 HJURP의 높은 발현을 갖는 환자가 낮은 HJURP 발현을 갖는 환자에 비해 상당히 감소된 암-특이 생존률을 가짐을 나타내는 임상 병리학적 인자를 평가하였다 (log-rank test; P = 0.016).
siRNA에 의한 CRC 세포의 형질도입
대장암에서의 HJURP 발현을 평가하기 위해, 4개 CRC 세포주 (HT29, HCT116, SW480, 및 SW620)에서의 HJURP 발현 수준을 확인하고 인간 HJURP mRNA를 특이적으로 표적하는 siRNA로 이들 세포를 트랜스펙션시켰다. 그런 다음 RT-PCR 및 웨스턴 블롯팅으로 HJURP 발현의 하향조절을 확인하였다. HJURP mRNA 및 단백질 발현 수준 둘 다 대조 CRC 세포에 비해 HJURP-siRNA-트랜스펙션 세포에서 거의 검출되지 않았다 (도 3).
HJURP 발현의 siRNA-유도 하향조절에 의한 대장암 세포 증식의 감소
세포 증식에서 HJURP의 효과를 확인하기 위해, HJURP에 대한 RNA 간섭을 실시하였다. 트랜스펙션된 세포의 증식을 MTT 검정으로 확인하였다. HJURP 표적 siRNA가 처리된 세포는 대조 세포에 비해 낮은 비율로 증식하였다(도 4; p < 0.005). 증식 비율을 비교하였다: siRNA_HCT116 at 48 h (61%) 및 72 h (62%); siRNA_HT29 cells at 24 h (77%), 48 h (68%), 및 72 h (59%); siRNA_SW480 at 24 h (79%), 48 h (85%), 및 72 h (81%); siRNA_SW620 at 24 h (83%), 48 h (66%), 및 72 h (54%). 이러한 결과는 HJURP의 사일런싱이 세포 증식을 억제함을 제시한다.
HJURP의 하향조절에 의한 대장암 세포의 이동 및 침습의 억제
암 진행에서 HJURP의 기능을 특성화하기 위해, 트랜스웰(Transwell) 챔버 검정을 실시하여 세포 이동 및 침습을 확인하였다. 도 5에서 볼 수 있는 바와 같이, 대장암 세포에서 HJURP 발현의 하향조절은 대조 세포에 비해 세포 이동 및 침습을 상당히 감소시켰다. 이동 검정에서, HJURP 표적 siRNA를 처리한 그룹은 대조 세포(p < 0.05)에 비해 하부 챔버로의 이동이 상당히 감소하였다(36.3 ± 3.27%). 4개 그룹의 감소율은 하기와 같다: HT29, 36.3%; HCT116, 37.3%; SW480, 31.8%; 및 SW620, 39.6%. HJURP의 하향조절은 약 34.5 ± 7.96% (p < 0.05)의 대조세포에 비해 세포 침습을 상당히 억제하였다. 감소율은 하기와 같다: HT29, 33.3%; HCT116, 44.0%; SW480, 24.7%; 및 SW620, 36.2%. 이러한 결과는 HJURP가 대장암 세포 이동 및 침습을 약화시킴을 나타낸다.
HJURP에 의한 CRC 세포 앵커리지-독립 성장 감소
종양형성의 특징 중 하나인 앵커리지-독립 성장에서 HJURP가 필요한지 여부를 확인하기 위해(정상 및 바이러스-트랜스펙션 세포에서 앵커리지 및 성장 조절), 소프트 아가 콜로니 형성 검정을 실시하였다. HJURP 표적 siRNA가 트랜스펙션된 세포는 상당히 감소된 52.9 ± 10.15% (p < 0.05; 95.1 ± 8.41 colonies)의 콜로니 형성 효율을 보였다. 4개 세포주의 콜로니수는 하기와 같다: 4개의 대조 그룹 및 siRNA-처리 그룹 각각에서 HT29, 190 ± 7.51 및 102 ± 8.02; HCT116, 175 ± 5.51 및 87 ± 3.61; SW480, 161 ± 9.00 및 107 ± 8.50; 및 SW620, 211 ± 6.51 및 89 ± 9.61 (도 6). 이러한 발견은 HJURP가 대장암 세포의 앵커리지-독립 성장에서 중요한 참여인자임을 나타내는 것이다.
결론
본 발명은 악성종양에서 HJURP 발현의 기능 및 의의를 인 비트로에서 연구하였다. HJURP 기능은 동원체에서 CENP-A의 침적과 관련되어 있으며, 이의 활성화는 염색체 안정성을 위해 필요하다. 또한, 암세포의 불명화에도 관련되고, 이의 이상 상향조절이 여러 인간 암에서 보고되었다.
HJURP은 CENP-A과 상호작용하여 동원체에서 CENP-A를 국소화시키고 이의 특이 샤프론으로서 새로운 CENP-A 뉴클레오솜을 로딩한다. CENP-A가 표준 히스톤 H3와 다른 히스톤 H3 변이체이기는 하지만, 이의 염색질 결합은 척추동물 세포에서 DNA 복제와 관련이 없다. 새로 합성된 CENP-A는 동원체 내로, 특히 인간 세포에서 초기 G1 상으로 통합되다.
CENP-A는 동원체 형성 및 동원체 조립에서 중요한 역할을 한다. CENP-A는 유사분열 방추로의 염색체 부착의 복합적인 작업을 실행하며 이러한 부착이 정확히 이루어지도록 한다. 따라서, CENP-A의 이상 조절은 유사분열 진행에서의 시그널링 및 후기에서 방추체극으로의 염색체 이동의 조절을 지연시킨다. 인간 유방암에서, HJURP의 과발현은 Aurora kinases와 같은 유사분열 키나아제와 유사하여, 종양 발달의 특징인 유전적 불안정성을 유도한다.
대장암의 종양형성에서 HJURP의 분자 메커니즘은 아직 명확하지 않다. 그러나, HJURP가 p21, potent cyclin-dependent kinase inhibitor (CKI), ERK1/2, 및 GSK3β을 조절하여 세포 주기를 제어함이 보고되었다. 대장암에서 p21의 핵 국소화가 MAPK/ERK1/2 경로에 의해 조절되므로, 대장암에서의 HJURP의 종양형성 기능 또한 동일한 경로에 의한 것으로 여겨지고 있다.
본 발명에서는 HJURP의 고발현이 임상병리학적 인자와는 관련이 없으나, 림프절 전이, 신경 침습, 전이, 및 HJURP 고발현을 갖는 환자에서 전체 생존률이 낮음을 발견하였다. 또한, HJURP 고발현이 대장암 환자에서의 낮은 생존률과 연관되어 있음을 확인하였다. The Cancer Genome Atlas (TCGA) 데이터베이스 분석 결과는 HJURP 고발현 환자의 5년 생존률이 55%인 반면, HJURP 저발현 환자의 5년 생존률이 64%임을 보였다. 두 그룹 사이의 5년 생존률의 차이는 통계학적 유의성 없이 단지 9%였다.
또한, 본 발명에서는 HJURP의 과발현이 CRC 예후와 반비례적으로 관련되어 있어, 대장암 환자에서에서의 독립적인 예후 인자로서 사용될 수 있음을 입증하였다.
또한, 본 발명에서는 대장암 조직에서의 HJURP의 발현 및 4개 대장암 세포주에서 siRNA에 의한 HJURP 하향조절에 따른 세포 증식, 이동, 침습 및 콜로니 형성에 대한 효과에 초점을 맞추었다. 그 결과, siRNA는 4개 세포주에서 증식, 침습 및 이동 능력을 억제함을 확인하였다. 또한, 소프트 아가 콜로니 형성을 통해 인 비트로 종양형성이 siRNA로의 트랜스펙션 후 현저히 감소함을 확인하였다. 이러한 결과는 HJURP가 대장암에서 종양형성 유전자로서 작용함을 제시한다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (8)
- HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정할 수 있는 제제를 포함하는, 대장암 진단 또는 예후 예측용 조성물.
- 제 1 항에 있어서,
상기 mRNA 수준을 측정할 수 있는 제제는 상기 HJURP에 특이적으로 결합하는 프라이머, 프로브 또는 안티센스 올리고뉴클레오티드를 포함하는 것인, 대장암 진단 또는 예후 예측용 조성물. - 제 1 항에 있어서,
상기 단백질의 발현 수준을 측정할 수 있는 제제는 상기 HJURP에 특이적으로 결합하는 항체, 엡타머, 올리고펩타이드 또는 PNA(Peptide nucleic acid), 또는 상기 단백질을 코딩하는 유전자에 특이적인 상보적 서열을 갖는 프라이머 또는 프로브를 포함하는 것인, 대장암 진단 또는 예후 예측용 조성물. - 제 1 항 내지 제 3 항 중 어느 한 항에 따른 조성물을 포함하는, 대장암 진단 또는 예후 예측용 키트.
- 제 4 항에 있어서,
상기 키트는 RT-PCR 키트, DNA 칩 키트 또는 단백질 칩 키트를 포함하는 것인, 대장암 진단 또는 예후 예측용 키트. - 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계;
상기 발현수준을 정상 대조군과 비교하는 단계; 및
상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암이 있는 것으로 판단하는 단계;를 포함하는, 대장암 진단을 위한 정보 제공 방법. - 대상으로부터 분리된 시료에서 HJURP(Holliday Junction Recognition Protein)의 mRNA 또는 이의 단백질의 발현수준을 측정하는 단계;
상기 발현수준을 정상 대조군과 비교하는 단계; 및
상기 발현수준이 정상 대조군에 비해 과발현될 경우 대장암 악화 위험성이 높은 것으로 판단하는 단계;를 포함하는, 대장암 예후 예측을 위한 정보 제공 방법. - HJURP(Holliday Junction Recognition Protein)의 활성 억제제 또는 발현 억제제를 유효성분으로 포함하는, 대장암 치료용 약학적 조성물.
