Abstract
본 명세서에서는 섬유아세포 증식의 유도 및 단백질의 분비 혹은 생산 증가를 위한 소분자가 제공된다. 본 명세서에 기재된 소분자는 피부 재생의 촉진을 위하여 이용될 수 있다. 또한, 본 명세서에서는 피부 재생 및 상처 치유를 촉진시키는 방법들이 제공된다. 본 발명은 일반적으로 피부 재생 및 상처 치유에 관한 것으로, 더욱 구체적으로는, 인간 피부의 섬유아세포 증식 및 증가된 단백질 생산의 소분자-매개 유도에 관한 것이다
Description
본 발명은 일반적으로 피부 재생 및 상처 치유에 관한 것으로, 더욱 구체적으로는, 인간 피부의 섬유아세포 증식 및 증가된 단백질 생산의 소분자-매개 유도에 관한 것이다.
척추동물의 신체에서 가장 큰 기관인 피부는, 복합적 신경 및 혈액 공급부를 지니는 진피와 표피로 구성되어 있다. 제3층인 하피(hypodermis)는 주로 지방과 소성 결합 조직층으로 구성되어 있다. 이들 세 층은 상처 등과 같은 임의의 기계적 손상으로부터 신체를 보호하는 데 있어서 중요한 역할을 한다.
표피 바로 밑에 위치한 진피는, 피부의 커다란 부분을 구성하며, 일부 엘라스틴과 글라이코사미노글라이칸(GAG)을 지니는 콜라겐으로 구성된다. 진피에 존재하는 주된 세포 유형은 섬유아세포인데, 이는 상처 치유 과정에서 중요한 역할을 하는 프로테아제 및 콜라게나제 등과 같은 리모델링 효소를 생산할 수 있다. 상처 치유는 순차적 기전이고, 더욱 구체적으로는 이벤트-구동 과정이며, 이에 따라서 하나의 세포 유형 셋 오 프 캐스케이드(set off cascade)로부터 다른 세포 유형으로 신호전달하여, 치유 단계들을 통해 상처를 진전시킨다. 성인 상처 치유는 본질적으로 회복 과정이며, 이는 통상적으로 흉터 형성을 보인다. 조직 회복은 상처 부위에 피브린 응괴 침착과 함께 즉시 시작되어, 손상된 혈관으로부터의 출혈을 방지한다. 순환하는 혈소판은 이어서 상처 부위에 응집되고, 각종 염증 매개체, 예컨대, PDGF, TGF-α 및 TGF-β, 표피 성장 인자(EGF) 및 FGF가방출된다. 이들 분자는 또한 상처 회복 과정에서 하류에서의 주된 역할을 하는 것으로 여겨진다.상처에 대한 성인 조직의 염증 반응은 호중구의 조기 유입을 특징으로 하며, 그 개수는 꾸준히 증가하여 상처후 24 내지 48시간에 최대에 도달한다. 호중구 개수가 감소되기 시작함에 따라서, 대식세포가 상처 부위를 차지하여 다시 이주시킨다. 재상피화가 또한 동시에 일어나고, 각질형성세포들이 상처 가장자리의 기저세포 및 진피내의 깊은 곳으로부터 과립화된 조직을 가로질러 이동한다. 각질형성세포들은 피부의 장벽 특성을 재확립시키자마자, 이들은 접촉 저해 시 기저 세포 표현형을 되찾아 층상 편평 각질화 표피 내로 분화시킨다.
염증 반응 및 상피화의 최종 단계는 섬유아세포 및 내피 세포의 이동 그리고 과립화 조직의 형성과 동시에 일어난다. 혈관신생과 섬유증식이 이어서 일어나고, 섬유아세포는 지배적인 세포형으로 되어, 콜라겐과 ECM 아래쪽에 놓인다. 콜라겐의 리모델링은 섬유아세포 및 대식세포에 의해 생산되는 기질 금속단백분해효소를 이용해서일어난다.
피부 섬유아세포는, 세포 복제를 겪기 위하여 훨씬 더 높은 농도의 섬유아세포 성장 인자(FGF)를 필요로 하고,통합된 조직 내로 표피의 중층편평 상피 세포를 조직하는 ECM의 생성을 담당한다. 또한, 피부 섬유아세포는, 신체의 근막에 피부를 고정시키는, 결합 조직의 긴 섬유상 띠를 형성한다. 피부 섬유아세포 없다면, 상처 부위는세포외 기질을 재생할 수 없고, 표피 피부 세포는 상처 부위 위에 증식될 수 없다. 따라서, 피부 섬유아세포가없다면, 피부는 상처로부터 적절하게 회복될 수 없다.
인간에서, 피부는 체질량의 대략 1/10을 나타내고, 이 주요 기관의 일부에 대한 외상, 질환, 화상 혹은 수술 등과 같은 손상은 극적인 결과를 지닌다. 조직-공학적 조작된 피부 대체물은 급성 및 만성 피부 처와 싸우는데용된다. 조직 공학은 병든 혹은 손상된 조직 혹은 기관을 대체하기 위하여 새로운 생리학적 물질을 재생시키는 것을 목표로 한다. 이를 달성하기 위하여, 세포의 공급원이 요구될 뿐만 아니라, 세포가 지원될 수 있는 인조 세포외 기질(ECM)도 요구된다.
콜라겐 및 파이브로넥틴 등과 같은 천연 생체고분자는 세포가 유착될 수 있는 바이오소재의 잠재적인 공급원으로서 연구되어 왔다. 조직 공학에 이용되는 제1세대의 분해성 고분자는 다른 수술 용도로부터 개조되었고, 기계적 특성 및 분해 특성의 점에서 결점을 지닌다. 이것은 세포 및/또는 분자를 인시츄로 전달하는 방식, 소위 스마트 기질 기술(smart matrix technology)로서 주로 합성 분해성 겔의 발달을 초래하였다. 조직 또는 기관 회복은 통상 흉터의 생성을 초래하는 섬유 형성 반응을 수반한다. 그러나, 특정 포유류 조직은, 흉터 형성 없이 완벽한 재생 능력을 지니는데; 양호한 예는 MRL/MpJ 마우의 귀와, 배아 혹은 태아 피부를 포함한다. 이들 모델시스템의 연구는, 이러한 완벽한 재생을 달성하기 위하여, 염증반응이 섬유형성 및 흉터 형성의 정도를 감소시키도록 개조된 것으로 드러났다.
피부 섬유아세포는 ECM 침착, 상피-간엽성 상호작용 및 상처 치유에서 주요 역할을 하는 세포의 동적 모집단이다. 섬유아세포는 실험실에서 용이하게 배양되고, 조직-공학적 조작된 피부 대체물 내로의 섬유아세포의 혼입은증상을 보이는 통증 완화, 급성 및 만성 상처의 더욱 신속한 치유, 더 적은 흉터 형성 및 더 양호한 성형 결과를 포함하는 고무적인 결과를 산출해왔다. 그러나, 현재, 손상되지 않은 피부의 해부학, 생리학, 생물학적 안정성 혹은 미학적 속성을 완벽하게 복제하는 생체공학적으로 조작된 피부의 모델은 없다.
각종 조직-공학적 조작된 피부 산물은 특정 임상 적용을 위하여 특정 산물의 동정을 가능하게 하므로 다기관 임상 시험에서 비교될 필요가 있다. 이것은 지정된 산물의 증가된 사용에 따른 그들의 비용의 저감을 잠재적으로유도할 수 있었다. 또한, 조직-공학적 조작된 피부 대체물과 사이토카인 및 성장 인자와의 조합은 장래에 항균성 유익을 위하여 디펜신을 혼입시킬 가능성을 제공할 뿐만 아니라 상처 치유를 증대시키는데 사용될 수 있다.__
따라서, 재료 기술이 지능적 설계에 기초할 뿐만 아니라 재생 과정에 내포된 분자에도 기초하는 피부 대용물을생성하는 신규한 접근법에 대한 필요성이 존재한다.
본 발명은 소정의 소분자가 섬유아세포 증식을 유도하고/하거나 단백질 생산을 증가시킨다는 중대한 발견에 기초한 것이다.
본 명세서에서는 대상체에서 섬유아세포 증식을 유도하는 방법이 제공된다. 상기 방법은 섬유아세포 증식을 유도하는 소분자의 유효량을 대상체에게 투여하는 단계를 포함한다. 소정의 양상에 있어서, 소분자는 SL 327, ABT702, 페노밤(fenobam), SX 011, 피소스티그민 헤미황산염(physostigimine hemisulfate) 또는 NBQX이다. 또 본 명세서에서는 대상체에서 단백질 분비를 증가시키는 방법이 제공된다. 상기 방법은 단백질 분비를 증가시키는 소분자의 유효량을 대상체에게 투여하는 단계를 포함한다.

피부 섬유아세포는 피부 장벽 보전력 및 상처 치유를 촉진시키는 결합 조직 섬유 및 성장 인자의 형성에 기여하는 콜라겐의 생산을 담당하는 피부 세포이다. 정상의 상처 치유에 있어서, 섬유아세포는 상처액에 존재하는 각종 인자에 반응하여 새로운 피부층을 증식시키고 재생시키기 위하여 주변의 미손상 조직으로부터 과립화 조직으로 동원된다. 정상의 노화 동안, 피부 섬유아세포는 정상의 상처 치유에 관여하는 성장 인자(예컨대, EGF,FGF7/KGF, TGFβ, 콜라겐)를 증식시키는 능력 및 이러한 성장 인자를 분비시키는 능력을 둘 다 소실한다. 섬유아세포 증식을 촉진시키고 단백질(예컨대, EGF, FGF7/KGF, TGFβ, 콜라겐)의 분비를 증가시키는 소분자를 식별하도록 설계된 고속-처리 선별이 본 명세서에 기재되어 있다.
성장 인자는 세포 성장, 증식 및 세포 분화를 자극시킬 수 있는 자연 발생적 물질이다. 성장 인자들은 각종 세포 과정을 조절하기 위하여 중요하다. 성장 인자들은 전형적으로 세포들 간의 신호전달 분자들로서 작용한다.그들의 예는 그들의 표적 세포의 표면 상의 특정 수용체에 결합하는 호르몬 및 사이토카인이다. 이들은 흔히 세포 분화 및 성숙을 촉진시키며, 이는 성장 인자들 간에 다양하다. 예를 들어, 골 형성 단백질은 골 세포 분화를자극하는 한편, 섬유아세포 성장 인자와 혈관 내피 성장 인자는 혈관 분화(혈관신생)를 자극시킨다. 성장 인자및 성장인자 계열의 예는, 아드레노메둘린(AM), 안지오포이에틴(Ang), 자가분비 운동 인자, 골 형성 단백질(BMP), 뇌 유래 신경영양 인자(BDNF), 표피 성장 인자(EGF), 에리트로포이에틴(EPO), 섬유아세포 성장 인자(FGF), 신경교 세포주 유래 신경영양 인자(GDNF), 과립구 집락 자극 인자(G-CSF), 과립구 대식세포 집락 자극인자(GM-CSF), 성장 분화 인자-9(GDF9), 간세포 성장 인자(HGF), 간세포-유래 성장 인자(HDGF), 인슐린 유사 성장 인자(IGF), 이동 자극 인자, 마이오스타틴(GDF-8), 신경 성장 인자(NGF) 및 기타 뉴트로핀, 혈소판 유래 성장 인자(PDGF), 트롬보포이에틴(TPO), 형질전환 성장 인자 알파(TGF-α), 형질전환 성장 인자 베타(TGF-β), 종양 괴사 인자-알파(TNF-α), 혈관 내피 성장 인자(VEGF), Wnt 신호전달 경로, 태반 성장 인자(PlGF), 소태아 성장호르몬(FBS), IL-1, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, bFGF 및 VEGF를 포함하지만, 이들로 제한되는 것은 아니다.
섬유아세포는 위에 있는 표피에 확산되어 측분비 방식으로 각질형성세포에 영향을 미쳐 자가분비 효과 및 측분비 효과를 지니는 성장 인자 및 사이토카인을 방출하는 가용성 인자들을 분비한다. 자가분 활성은, 섬유아세포 증식뿐만 아니라 콜라겐 합성을 촉진시키는 결합 조직 성장 인자의 분비와, 형질전환 성장 인자(TGF)-β-유도 합성을 포함한다. 측분비 활성은, 구체적으로 각질형성세포 성장 인자(KGF), 과립구-대식세포 집락 자극 인자, 인터류킨(IL)-6 및 섬유아세포 성장 인자(FGF)-10의 섬유아세포 분비를 통해서, 각질형성세포 성장 및 분화에 영향을 미친다. 반응 시, 각질형성세포는 IL-1 및 부갑상샘 호르몬-관련 펩타이드를 합성하고, 이어서, KGF를 생성하도록 섬유아세포를 자극시키고, 이에 따라서 이중 측분비 루프가 존재한다. 또한, 단독으로 배양된 각질형성세포는 IL-1을 상대적으로 약하게 발현시키지만, 섬유아세포와 공동 배양된 경우에는 IL-1 및 c-Jun의 증가된 발현을 보인다. 따라서, 피부 기질 내에 혼입된 섬유아세포는 ECM 및 자극 성장 인자를 생산하는데 역할을하며, 이는 표피 형성을 지원하고 상처 치유를 용이하게 하기 위한 최적 환경을 제공한다.
섬유아세포 밀도는 정상의 표피 형태학의 발달 및 각질형성세포 분화를 위하여 고려되는 중요한 인자이며, 그최적 밀도는 여전히 확립될 필요가 있다. 섬유아세포는 또한 부분적으로 콜라겐 제IV형 및 제VII형, 라미닌 5및 니도겐을 생성함으로써 기저막 형성에 기여하지만, 또한, 사이토카인의 분비를 통해서 각질형성세포를 자극시켜 기저막 성분을 생성한다. 섬유아세포에 의해 분비된 TGF-β는 각질형성세포에 의해 콜라겐 제IV형 및 제VII형의 합성을 유도한다.
신생혈관형성 및 림프관신생은 또한, 정상의 피부 항상성 및 상처 치유의 유지를 위하여 중요한 과정이며, 이를위하여 섬유아세포는 중요한 측분비 역할을 한다. 혈관 내피 성장 인자(VEGF) 계열의 구성원은 VEGF-A, -B, -C및 -D를 포함하며, 이들은 정상의 인간 섬유아세포에 의해 생산되고 특정 수용체를 통해서 혈관 및 림프관 내피세포 증식을 조절함에 있어서 중요하다. VEGF-A는 혈관 형성 가능한 상주 내피세포와 내피 전구 세포의 활성화에 관여하는 것으로 잘 알려져 있고; VEGF-B는 내피 세포를 위하여 덜 유사분열성인 한편, VEGF-C 및 -D는, 혈관신생을 매개하기 위하여 VEGF 수용체 2(VEGF-R2)에 결합하고 그리고 림프관신생에 영향을 미치기 위하여VEGF-R3에 결합하는, 동일한 수용체 특이성을 지닌다.
FGF-1은 상처를 발달시켜 효과적으로 봉합하기 위하여 신체의 자체 유착 조직을 촉진시킴으로써, 감염을 방해하고 흉터 형성을 경감시킨다. 섬유아세포 활성을 자극하기 위하여 FGF를 이용하는 것은 결합 조직을 구성하는콜라겐의 강인한 성향으로 인해 조직을 봉합하는 효과적인 수단이다. 조직들을 함께 봉합하기 위하여, 인간 신체는 콜라겐과 엘라스틴을 이용해서 우수한 전단 강도를 얻는다. 강력한 소섬유로 묶인 콜라겐 가닥들을 포함하는 제I형 콜라겐은 그의 구조적 보전성을 증가시키는 독특한 3중 나선 구조를 지닌다.__ 이하의 실시예는 예시하기 위하여 의도된 것이므로 본 발명을 제한하는 것은 아니다. 실시예 1 인간 피부 섬유아세포(hDF)의 증식을 위한 고속-처리 증식 선별 검정
증식 검정을 위하여, 인간 피부 섬유아세포(hDF)를 DMEM(깁코사(Gibco)) 15% FBS(하이클론사(HyClone)) 중1,000 세포/웰에서 96 웰 흑색/투명 바닥 조직-배양 처리 플레이트(코닝사(Corning))에 평판 배양하였다. 그 다음날에 세포를 2.5μM 최종 농도의 소분자 라이브러리(토시스(Torcris) 1120 생물학적 활성 화합물)로 처리하였다. 처리 3일 후, 세포를 고하고, DAPI로 염색하여, 로쉐 셀라비스타 하이-컨텐트 이미징 시스템(RocheCellavista High-Content Imaging System)을 이용해서분석하였다(도 1 참조).
고속-처리 증식 선별 검정으로부터, ABT702 2염산염(아데노신 키나제 길항제), 페노밤(Fenobam)(mGlu5 수용체길항제) 및 SX 011(p38 MAPK 길항제)이 200nM 농도에서 최고 증식 수준의 hDF를 유도한 것으로 판명되었다(도 2참조).
실시예 2
FGF7 생산을 위한 고속-처리 선별 검정 FGF7 생산 검정을 위하여, hDF를 DMEM(깁코사) 1X N2/B27 보충물(인비트로젠사(Invitrogen)) 중 19,000 세포/웰에서 96 웰 투명 조직-배양 처리 플레이트(팔콘사(Falcon))에서 평판배양하였다. 그 다음날에 세포를 2.5μM최종 농도에서의 소분자 라이브러리(토시스 1120 생물학적 활성 화합물)로 처리하였다. 처리 3일 후, 상청액을수집하여 FGF7 인간 ELISA 키트(압캠사(Abcam)) 및 바이오텍 시너지 2 다중-검출 마이크로플레이트 리더(BioTekSynergy 2 Multi-Detection Microplate Reader)를 이용해서 FGF7 함량에 대해서 분석하였다(도 1 참조) FGF7 생산 검정을 위한 고속-처리 선별 검정을 위하여, 25μM 농도에서의 프로스타글란딘 E2가 hDF 중 최고 생산 수준의 FGF7을 유도한 것으로 판명되었다(도 3 참조).
실시예 3 TGFβ1 생산을 위한 고속-처리 선별 검정 TGFβ1 생산 검정을 위하여, hDF를 DMEM(깁코사) 1X N2/B27 보충물(인비트로젠사) 중 19,000 세포/웰에서 96웰 투명 조직-배양 처리 플레이트(팔콘사)에 평판배양하였다. 그 다음날에 세포를 2.5μM 최종 농도의 소분자라이브러리(토시스 1120 생물학적 활성 화합물)로 처리하였다. 처리3일 후, 상청액을 수집하여 TGFβ1 인간ELISA 키트(압캠사) 및 바이오텍 시너지 2 다중-검출 마이크로플레이트 리더를 이용해서 TGFβ1 함량에 대해서분석하였다(도 1 참조).
TGFβ1 생산 검정을 위한 고속-처리 선별 검정으로부터, 24nM 농도에서의 프록시판 옥살산염(히스타민 H3 수용체 신호전달 효능제), 24nM 농도에서의 m-3M3FBS(포스포리파제 C 신호전달), 1.6μM 농도에서의 KB-R7943 메실산염 및 98nM 농도에서의 LY294002 염산염이 hDF 중 최고 생산 수준의 TGFβ1을 유도한 것으로 판명되었다(도 4참조). 실시예 4 EGF 생산을 위한 고속-처리 선별 검정 EGF 생산 검정을 위하여, hDF를 DMEM(깁코사) 1X N2/B27 보충물(인비트로젠사) 중 19,000 세포/웰에서 96 웰투명 조직-배양 처리 플레이트(팔콘사)에서 평판배양하였다. 그 다음날에 세포를 2.5μM 최종 농도에서의 소분자 라이브러리(토시스 1120 생물학적 활성 화합물)로 처리하였다. 처리 3일 후, 상청액을 수집하여, EGF 인간ELISA 키트(압캠사) 및 바이오텍 시너지 2 다중-검출 마이크로플레이트 리더를 이용해서 EGF 함량에 대해서 분석하였다(도 1 참조).
EGF 생산 검정을 위한 고속-처리 선별 검정 위하여, 25 및 12.5 μM 농도에서의 CD 437 및 CD 1530(레티노이드 RAR 신호전달 효능제), 12.5μM 농도에서의 CY 208-243(도파민 D1 수용체 효능제) 및 12.5 μM 농도에서의PSB 06126(NTPDase 3 길항제)이 hDF 중 최고 생산 수준의 EGF를 유도한 것으로 판명되었다(도 5 참조). 실시예 5 콜라겐 제1형 생산을 위한 고속-처리 선별 검정 콜라겐 제1형 산 검정을 위하여, hDF를 DMEM(깁코사) 1X N2 보충물(인비트로젠사) 중 5,000 세포/웰에서 96웰 투명 조직-배양 처리 플레이트(팔콘사)에 평판배양하였다. 그 다음날에 세포를 2.5μM 최종 농도에서의 소분자 라이브러리(토시스 1120 생물학적 활성 화합물)로 처리하였다. 처리 3일 후, 상청액을 수집하여 프로콜라겐제1C형-펩타이드 EIA 키트(타카라사(Takara)) 및 바이오텍 시너지 2 다중-검출 마이크로플레이트 리더를 이용해서 콜라겐 제1형 함량에 대해서 분석하였다(도 1 참조).
콜라겐 제1형 검정을 위한 고속-처리 선별 검정으로부터, 390nM 농도에서의 다이피리다몰(아데노신 수송저해제), 390nM 농도에서의 메틸리카코니틴 시트르산염(α7 뉴런 니코틴 수용체 길항제), 6.25μM 농도에서의실로스타졸(PDE3A 길항제), 780nM 농도에서의 클레마스틴 푸마르산염(H4 수용체 길항제) 및 97.5nM 농도에서의WIN 64338(브라디키닌 B2 수용체 길항제)이 hDF 중 최고 생산 수준의 EGF를 유도한 것으로 판명되었다(도 1 참조).
본 발명은 상기 실시예를 참조하여 설명되었지만, 본 발명의 정신과 범위 내에 변형 및 변경이 포함되는 것이이해될 것이다. 따라서, 본 발명은 단지 하기 특허청구범위에 의해서만 제한된다
