Abstract
본 발명은 miR-486-5p 또는 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는 간섬유증 예방 또는 치료용 약학적 조성물에 관한 것으로, 상세하게 miR-486-5p는 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(HSC)를 활성화를 억제하는 바, 간섬유증의 진행을 억제시킴으로써 간섬유증의 예방이나 치료에 있어서 유용하게 이용될 수 있다.
Description
본 발명은 miR-486-5p 또는 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는 간섬유증 예방 또는 치료용 약학적 조성물에 관한 것으로, 상세하게 miR-486-5p는 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(HSC)를 활성화를 억제하는 바, 간섬유증의 진행을 억제시킴으로써 간섬유증의 예방이나 치료에 있어서 유용하게 이용될 수 있다.
만성 간질환은 바이러스성 간염, 비만 또는 알코올 남용과 같은 지속적인 장기 간 손상에 대한 반응으로 발생한다. 간 질환의 원인은 다양할 수 있지만 대부분의 만성 간 질환의 일반적인 병리학적 특징은 간섬유증(liver fibrosis) 이다. 간세포에 의해 재생이 되지 못한 부분을 세포외기질(ECM)로 채우게 되는 간섬유화은 만성 그리고/또는 심한 간 손상에 의해 과도한 세포외기질(ECM)의 축적을 가져오고, 이는 간의 정상적인 기능을 교란시킴으로써 결국 간경변 또는 간암의 말기 단계로 이어진다. 이런 간섬유화에 주요 원인인 간성상세포(Hepatic stellate cells, HSC)는 섬유생성세포 유형으로, 간 섬유 생성은 간성상세포(HSC) 활성화에 의해 시작된다. 정상 간에서 휴지 상태(Q-)인 간성상세포(HSC)는 비증식 표현형을 유지하며 비타민 A/레티노산(Vitamin A/retinoic acid)을 저장한다. 간 손상 시 간성상세포(HSC)는 활성화되어 손상된 간에서 증식하고, 콜라겐과 같은 세포외기질(ECM)을 생성하는 근섬유 모세포 간성상세포로 전환하여 분화된다. 따라서 간성상세포(HSC) 활성화를 조절하는 것은 항섬유증 치료를 위한 잠재적인 치료 전략이 될 수 있다.
한편, 마이크로RNA(microRNA)는 유전자의 전사 후 과정에서 RNA 간섭을 통해 수백 개의 유전자의 발현을 제어함으로써, 광범위한 생체경로, 예컨대 세포 증식, 분화 및 세포자살을 조절하는 작은 비단백질-암호 RNA이다(Calin et al., Nat.Rev. Cancer, 6: 857-866 (2006)). 마이크로RNA는 여러 가지 질병의 임상적 예후를 예측하거나 치료하는데 유용하나, 아직까지 간 섬유증 예방 또는 치료에 효과적인 마이크로RNA는 거의 보고된 바 없다.
이러한 배경 하에, 본 발명자들은 간 섬유증 예방 또는 치료에 효과적인 마이크로RNA를 발굴하고자 하였다.
본 발명의 발명자들은 편도선 유래 중간엽 줄기세포(T-MSC)의 엑소좀 유래인 miR-486-5p에 대한 간섬유증에 대한 치료효과를 평가하였으며, miR-486-5p가 간 섬유증 동물 모델에서 상당히 우수한 간섬유증 치료 효과가 있음을 확인하여 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 miR-486-5p, 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는 간섬유증 예방 또는 치료용 약학적 조성물. 또는 간 간섬유증 개선용 건강기능식품 조성물를 제공하는 것이다.
따라서 상기 목적을 달성하기 위하여, 본 발명은 miR-486-5p, 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는, 간섬유증 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 miR-486-5p, 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는, 간섬유증 개선용 건강기능식품 조성물을 제공한다.
본 발명에 따른 간섬유증 예방 또는 치료용 조성물은 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(Hepatic stellate cells, HSC)를 활성화를 억제하고, 간에서 콜라겐 침착(collagen deposition)을 감소시키는 바, 간섬유증의 예방이나 치료를 위해 유용하게 이용될 수 있다.
도 1은 SMO mRNA의 3 'UTR에서 miR-486-5p의 야생형 또는 돌연변이 결합 부위를 갖는 psiCHECK-2 벡터이다. SMO mRNA의 3'UTR 상의 miR-486-5p 결합 부위의 야생형(WT) 또는 돌연변이(MUT) 뉴클레오티드가 표시되었다. 돌연변이된 뉴클레오티드에는 밑줄을 그었다.
도 2는 miR-486-5p가 SMO를 직접 표적으로 함을 확인한 결과이다. 본 발명자들은 miR-486-5p가 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(Hepatic stellate cells, HSC)를 활성화를 억제하는 바, 간섬유증의 예방이나 치료를 위해 유용하게 이용할 수 있다는 것을 밝혀냄으로써 본 발명을 완성하였다.

상기 간섬유증은 바이러스성 간섬유증(HBV 감염) 및 비-바이러스성 간섬유증을 모두 포함하는 것이 바람직하고, 구체적으로 비-바이러스성 간섬유증인 것이 보다 바람직하나, 이에 한정되지 않는다. 더욱 구체적으로, 상기 비-바이러스성 간섬유증은 간섬유화, 알코올성 간경변증, 비알코올성 간경변증, 자가면역성 간경변증, 원발성 담즙성 담관염 관련 간경변증, 윌슨병 관련 간경변증, 혈색소증 관련 간경변증, 문맥성 간경변증, 괴사후성 간경변증, 영양결핍과 관련된 간경변증 및 심장성 간경변증으로 구성된 군으로부터 선택되는 어느 1종 이상의 질병인 것이 보다 바람직하나, 이에 한정되지 않는다.
본 발명에서 용어 "간섬유화(liver fibrosis)"는 조직의 손상으로 인해 염증반응이 계속 진행됨에 따라 활성 간성상세포 (hepatic stellate cell)에서 다량 분비된 교원질(collagen)과 세포외기질(extracellularmatrix, ECM)이 결합함으로써 나타나는 상처치유의 과정을 지칭한다. 간섬유화가 지속적으로 진행되면 간 조직 내 교원질이 다량 침착되고, 재생결절이 교원질로 둘러싸여 비정상적인 구조를 지니게 되는 간경화로 발달할 수 있다.
본 발명의 일 구현예로, 상기 miR-486-5p는 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(Hepatic stellate cells, HSC)를 활성화를 억제함으로써 간섬유증을 억제할 수 있으며, 본 발명의 실시예에서 이를 구체적으로 확인하고 있다. 또한, 간에서 콜라겐 침착(collagen deposition)을 감소시킬 수 있다.
본 발명의 다른 구현예로, 상기 조성물은 혈중 아스파르트산염 아미노기 전달 효소(Aspartate aminotransferase; AST), 혈중 알라닌 아미노기전달효소(Alanine aminotransferase; ALT), LW/BW(Ratio of liver weight to body weight) 수준을 감소시킬 수 있다.
본 발명의 조성물은 간섬유증의 예방 또는 치료 효과를 갖는 공지의 유효성분을 1종 이상 더 포함할 수 있다.
본 발명의 약학적 조성물은 약학적 분야에서 통상의 방법에 따라 개체의 신체 내 투여에 적합한 단위투여형의 제제로 제형화시켜 투여할 수 있다. 이러한 목적에 적합한 제형으로는 비경구투여 제제로서 주사용 앰플과 같은 주사제, 주입 백과 같은 주입제, 및 에어로졸 제제와 같은 분무제 등이 바람직하다. 상기 주사용 앰플은 사용 직전에 주사액과 혼합 조제할 수 있으며, 주사액으로는 생리 식염수, 포도당, 링거액 등을 사용할 수 있다. 또한, 주입 백은 염화폴리비닐 또는 폴리에틸렌 재질의 것을 사용할 수 있다.
본 발명의 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체를 더 포함할 수 있다. 예를들어, 주사제의 경우에는 보존제, 무통화제, 가용화제 또는 안정화제 등을, 국소 투여용 제제의 경우에는 기제(base), 부형제, 윤활제 또는 보존제 등을 추가로 포함할 수 있다.
본 발명의 조성물은 개체에게 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를들면, 경구, 직장, 복강내, 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관 내 주사에 의해 투여될 수 있으며, 바람직하게는 복강내 투여가 가능하나 이에 한정되지는 않는다.
본 발명의 조성물은 간섬유증의 치료 또는 예방을 위하여 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명에서 용어 “예방”은 본 발명에 따른 간섬유증의 예방 또는 치료용 조성물을 개체에 투여하여 간섬유증의 발병을 억제하거나 지연시키는 모든 행위를 의미할 수 있다.
본 발명에서 용어, “치료”는 본 발명에 따른 조성물을 간섬유증 발병 의심 개체에 투여하여 간섬유증의 증세가 호전되도록 하거나 이롭게 되도록 하는 모든 행위를 의미할 수 있다. 본 발명에서 용어, "개체"는 간섬유증이 발병되었거나 발병할 가능성이 있는 인간을 포함한 모든 동물을 의미할 수 있다.
또한, 본 발명의 조성물은 간섬유증 예방 또는 개선을 목적으로 식품 조성물, 바람직하게는 건강기능식품에 첨가될 수 있다.
본 발명에서, ‘건강기능식품’이란 질병의 예방 및 개선, 생체방어, 면역, 병후의 회복, 노화 억제 등 생체조절기능을 가지는 식품을 말하는 것으로, 장기적으로 복용하였을 때 생체에 무해하여야 한다.
본 발명의 식품 조성물에 포함되는 유효성분의 혼합양은 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있으며, 이에 제한되지 않는다.
상기 언급한 바와 같이, 본 발명에 따른 간섬유증 예방 또는 치료용 조성물은 헤지호그(Hedgehog, Hh) 경로에서 수용체인 SMO의 발현을 억제하여 간성상세포(Hepatic stellate cells, HSC)를 활성화를 억제하고, 간에서 콜라겐 침착(collagen deposition)을 감소시키는 바, 간섬유증의 예방이나 치료를 위해 유용하게 이용될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
<실험예>
하기의 실험예들은 본 발명에 따른 각각의 실시예에 공통적으로 적용되는 실험예를 제공하기 위한 것이다.
1. 세포 실험
LX2, 인간 HSC 라인(한국과학기술원 제공), 인간 1차 HSC(pHSC, 미국 노스캐롤라이나주 Zen-Bio Inc. 구입), HepG2, 인간 간세포 암종 셀 라인을 Dulbecco's Modified Eagle Medium(DMEM; Gibco)에서 10% FBS(Gibco) 및 1% P/S(Gibco)를 첨가한 Dulbecco(부산대학교 제공) 라인을 37℃에서 5% CO2를 포함하는 습한 대기에서 배양되었다.
HSC 활성화에 대한 miR-486-5p의 효과를 조사하기 위해, 70% 융합(confluence)의 pHSC를 FBS가 없는 배지에서 밤새 혈청 고갈시키고 24시간 동안 2% FBS 및 P/S가 없는 배지에서 배양했다. 그 후, 제조업체의 지시에 따라 Lipofectamine RNAiMAX transfection 시약(Invitrogen, Thermo Fisher Scientific)을 사용하여 음성 대조군으로 25 nM의 miR-486-5p 유사체(Bioneer, 대전, 대한민국) 또는 스크램블 miRNA(Bioneer, 대전, 대한민국)로 세포를 형질감염시켰다.
6시간 후, 2% FBS 및 1% P/S를 포함하는 새로운 배지로 교체한 다음 이러한 형질감염된 세포를 24시간 또는 48시간 동안 배양하였다.
2. 동물 모델 실험
수컷 C57BL/6 마우스는 Hyochang(Dae-gu, Korea)에서 구입했으며, 12 시간 명암 주기로 수용되었으며 일반 음식과 물을 자유롭게 이용할 수 있었습니다.
급성 간 손상 마우스에서 miR-486-5p의 직접적인 효과를 조사하기 위해 7주령 수컷 C57BL/6 마우스는 1주일 동안 2회 복강 내로 1.0mg/kg 체중의 CCl4(n = 10)를 투여 받았다. 8일째에 in vivo-jetPEI(Polyplus Transfection, Illkirch, France)를 사용하여 miR-486-5p 유사체(n = 5) 또는 스크램블 miRNA(n = 5) 마우스 당 8nmol을 복강 내로 주입하였다. 대조군으로 옥수수유 주입 마우스(n = 8)에 miR-486-5p 모방체(n = 4) 또는 스크램블 miRNA(n = 4)를 처리하였다. 희생 시 마우스의 체중을 측정하고이소플루란 흡입 마취 하에 멸균 심장 천자에 의해 혈액 샘플을 수집하였다.
그 다음 마우스를 경추 탈구에 의해 마취 하에 인도적으로 안락사 시키고 간을 동물로부터 즉시 해부하였다. 모든 동물 관리 및 수술 절차는 부산 대학교 기관 동물 관리 및 사용위원회의 승인을 받았으며 실험실 동물의 관리 및 사용에 대한 국립보건원(National Institutes of Health, NIH) 지침의 조항에 따라 수행하였다.
3. RNA 분석
전체 RNAs(Total RNAs)를 TRIzol 시약(Ambion, Thermo Fisher Scientific)을 사용하여 세포, 간 조직 또는 엑소좀에서 추출하였다.
RNA의 순도와 농도는 나노드랍(nanodrop)을 사용하여 결정하였다. 주형 cDNA는 제조사의 지시에 따라 SuperScript Ⅱ First-strand Synthesis System(Invitrogen) 또는 HB_I RT Reaction kit(HeimBioteck, Seongnam, Korea)를 사용하여 전체 RNA(Total RNAs)로부터 합성하였다.
제조업체의 사양(Mastercycler realplex Real-time PCR, Eppendorf, Hamburg, Germany)에 따라 Power SYBR Green Master Mix(Applied Biosystems, Thermo Fisher Scientific) 또는 HB_I Real-Time PCR Master Mix (HeimBioteck)를 사용하여 QRT-PCR(Quantitative real-time PCR)를 수행하였다. 모든 반응은 세 번 수행하였으며, 데이터는 ΔΔCt 방법에 따라 분석하였다. 발현 수준의 정규화를 위해 mRNA와 miRNA에 각각 40S 리보솜 단백질 S9 mRNA와 U6B Small nuclear RNA(RNU6B)를 사용하였다. mRNA 발현을 평가하는데 사용되는 모든 프라이머의 서열은 통상적으로 사용되는 것을 사용하였다. 아울러, miR-486-5p 의 경우 상업적으로 사전 설계된 프라이머를 HeimBioteck에서 구입했다. 모든 유전자 확인을 위해 PCR 산물을 직접 시퀀싱하였다(Macrogen, Seoul, Korea).
4. 웨스턴 블롯 분석(Western blot analysis)
프로테아제 억제제(Complete Mini 11836153001; Roche, Indianapolis, IN, USA)가 보충된 트리톤 용해 완충액(TLB)을 이용하여 전체 단백질(Total protein)을 세포, 간 조직 또는 엑소좀로부터 추출하였다. 간 조직에서 Gli2 검출을 위해 핵 분획화를 수행하였다. 단백질 농도는 Pierce BCA 단백질 분석 키트(Thermo Fisher Scientific)로 측정하였다.
동일한 양의 단백질을 8% 또는 10% SDS-PAGE로 분리하고 폴리비닐리덴디플루오라이드막(PVDF)(Millipore, Darmstadt, Germany)으로 옮겼다. 이 연구에 사용된 1차 항체는 다음과 같다: CD63(희석1: 200; sc-5275, Santacruz Biotech.), CD9(희석1 : 500; ab92726, Abcam), CD81(희석1 : 200; sc-16602, Santacruz Biotech.), calreticulin(희석1 : 1000; ab2907, Abcam), TGF-β(희석1 : 1000; 3711S, Cell Signaling Technology, MA, USA), α-SMA(희석1 : 1000; A5228, Sigma-Aldrich ), VIMENTIN(희석1 : 1000; sc-5565, Santacruz Biotech.), COL1α1(희석1 : 1000; NBP1-30054, Novus Biologicals, CO, USA), GFAP(희석1 : 1000; Z0334, Dako, Carpinteria, CA, USA), SMO(희석1 : 1000; ab72130, Abcam), GLI2(희석1 : 1000; GWB-CE7858, Genway Biotech., CA, USA), LAMIN B1(희석1 : 1000; ab16048, Abcam) 및 GAPDH(희석1 : 1000; MCA4739, AbD Serotec, Oxford, UK). Horseradish peroxidase (HRP)-conjugated anti-rabbit(ADI-SAB-300-J, ENZO Life science, NY, USA) 및 anti-mouse lgG (ADI-SAB-100-J, ENZO Life science)를 2차 항체로 사용하였다. 아울러, 제조업체의 사양(Ez-Capture Ⅱ, ATTO Corporation)에 따라 EzWestLumi ECL 솔루션(ATTO Corporation, Tokyo, Japan)을 사용하여 단백질 밴드를 검출하였다. 면역블롯(immunoblots)의 누적 밀도 분석(cumulative densitometric analysis)은 CS Analyzer 4 소프트웨어(ATTO Corporation)를 이용하여 측정하였다.
5. 간 조직학(Liver histology) 및 면역화학염색(immunohistochemistry)
간 표본을 10% 중성 완충 포르말린(Sigma-Aldrich)에 고정시키고 파라핀에 임베딩한 후 4μm 두께 섹션으로 절단하였다. 절편을 섬유화 과정에 대한 형태 및 시리우스(sirius) 레드 염색을 검사하기 위해 표준 H&E 염색을 사용하여 표준 프로토콜에 따라 탈랍, 수화 및 염색하였다. H&E 염색의 경우 섹션을 헤마톡실린(Gill's Hematoxylin V, Muto Pure Chemicals, Tokyo, Japan)에서 15분 동안 염색한 다음 에오신(1% Eosin Y Solution, Muto Pure Chemicals)으로 5분 동안 염색하였다. 시리우스 레드 염색을 위해, 섹션을 포화 피크르산 및 0.1% Fast Green을 함유하는 0.1% 시리우스 레드 F3B(Sigma-Aldrich)에서 30분 동안 배양하였다.
면역조직화학의 경우, 섹션을 3% 과산화수소에서 10분 동안 배양하여 내인성 과산화효소를 차단하였다. 10mM 구연산나트륨 완충액(pH 6.0)에서 가열하여 항원을 회수하였다. 절편을 TBS로 세척한 후 Dako 단백질 블락 (Dako)으로 30분 동안 처리하고, 1차 항체, anti-α-SMA 항체(ab5694; Abcam) 및 anti-GLI2(GWB-CE7858; Genway Biotech)와 함께 4℃에서 밤새 배양하였다. 또한 염색 특이성을 입증하기 위해 추가적인 절편을 비면역혈청에서 밤새 4℃에서 배양하였다. HRP-conjugated anti-rabbit(K4003; Dako)를 2차 항체로 사용하였다. 검출절차에 따라 3,3'-다이아미노벤지딘(Dako)을 사용하였다. 슬라이드를 Olympus CX41 광학 현미경(Olympus Optical Co. Ltd, Tokyo, Japan)로 관찰하였고, 조직 절편의 염색된 영역의 형태학적 분석은 cellSens 소프트웨어(Olympus Optical Co. Ltd)를 사용하여 수행하였다.
6. AST/ALT 측정
혈청 아스파르테이트 아미노트랜스퍼라제(AST/GOT, 글루타메이트-옥 살로아세테이트트랜스아미나제) 및 알라닌 아미노트랜스퍼라제(ALT/GPT, 글루타메이트 피루베이트 트랜스아미나제)는 GOT 및 GPT 시약(한국 서울 아산 제약)을 사용하여 제조업체의 지침에 따라 측정하였다.
7. 벡터 구축물의 클로닝 및 루시퍼라제 리포터 분석
벡터 구축물의 클로닝을 위해, 인간 miR-486-5p의 표적 유전자는 온라인 데이터베이스(http://mirwalk.umm.uni-heidelberg.de)를 사용한 생물 정보학 분석을 통해 예측하였다. 인간 miR-486-5p에 대한 결합 부위를 포함하는 게놈 DNA에서 인간 SMO의 3'-UTR을 PCR로 증폭하였다.
벡터 구축에 사용되는 프라이머 서열은 다음과 같습니다: 정방향 프라이머(Forward primer): TTT TCT CGA GGG GGC CAT GTC CTC TCT TAA(서열번호 2), 역방향 프라이머(Reverse primer) : TGC GGC CGC TTG GAG GCT ATG GAA GGT GG(서열번호 3). PCR. 산물은 AccuPrep PCR Purification Kit(Bioneer)를 사용하여 정제하고 제한효소 Xho1 및 Not1로 절단한 다음 psiCHECK2 벡터(Promega, WI, USA)에 클로닝하였다.
SMO의 3'-UTR이 포함된 벡터 구축물을 대장균으로 형질전환한 다음 AccuPrep Plasmid Mini Extraction Kit(Bioneer)를 사용하여 형질전환된 암피실린 내성 대장균에서 플라스미드 DNA를 추출하였다. 시퀀싱 분석을 통해 SMO의 3'-UTR의 miR-486-5p 결합 부위를 확인하였다. miR-486-5p 결합 부위가 없는 돌연변이 벡터는 Enzynomics(대한민국, 대전)에서 제조하였다.
루시퍼라제 리포터 분석의 경우, HepG2 또는 LX2 세포를 형질감염 하루 전에 P/S 없이 배양 배지에서 24 웰 배양 플레이트에 접종하였다. Lipofectamine 2000(Invitrogen)을 사용하여 세포를 psiCHECK-2 벡터 구축물과 25nM의 miR-486-5p 모방체[5'-UCCUGUACUGAGCUGCCCCGAG-3 '(서열번호 4); Bioneer] 또는 음성대조군으로서 스크램블 miRNA(SMC-2003; Bioneer)의 혼합물로 형질감염 시켰다. 형질감염 후 48시간 후 세포를 수확하고 제조업체의 프로토콜에 따라 Dual Luciferase Reporter Assay System (Promega)으로 수행하였다. 모든 반딧불이 루시퍼라제 활성 데이터를 레닐라(Renilla) 루시퍼라제 활성으로 정규화하고 최소 세 번의 반복 실험에서 얻은 값의 평균 ± s.e.m 로 표시하였다.
8. 통계 분석
데이터 결과를 평균± s.e.m로 표시하였고, 통계적 차이를 짝을 이루지 않은 two-sample Student’s t-test로 분석하였다. P-values <0.05 는 통계적으로 유의한 것으로 간주하였다.
실시예 1. miR-486-5p의 SMO 발현 억제를 통한 간성상세포 활성 억제
miRWalk를 사용한 생물 정보학 분석을 통해, miR-486-5p의 추정 대상으로 Hh 경로에서 수용체인 smoothened(SMO)를 예측하였다(도 2A). 루시퍼라제 리포터 분석을 통해 HepG2 또는 LX2 세포 모두에서 miR-486-5p가 SMO mRNA의 3’-untranslated region(3’-UTR)에 직접 결합함을 확인할 수 있었다(도 2B).
아울러, miR-486-5p가 HSC 활성화에 영향을 미치는지 여부를 조사하기 위해, 인간 활성화 pHSC를 miR-486-5p 모방체(mimic) 또는 스크램블(Scr)-miR로 형질감염시켰다. miR-486-5p는 활성화된 pHSC에서 거의 발현되지 않았지만, miR-486-5p로 형질감염된 pHSC에서는 그 발현이 크게 증가하였다. 순차적으로, Hh 전사인자인 miR-486-5p을 타겟으로 하는 SMO 및 glioblastoma 2(GLI2) 발현 수준은 스크램블(Scr)-miR로 형질감염된 세포와 대비하여 유의하게 감소함을 확인할 수 있었다(도 3A).
또한 Scr-miR에 비해 miR-486-5p로 형질감염된 pHSC은 profibrotic 마커 TGF-β, α-SMA, VIMENTIN 및 CTGF가 하향 조절되었다. 웨스턴 블롯 분석은 miR-486-5p로 형질감염된 pHSC에서 SMO, GLI2 및 전섬유증(profibrotic) 마커의 감소를 확인할 수 있었다(도 3B 및 도 3C). 이러한 결과는 miR-486-5p가 SMO를 직접 표적화함으로써 HSC 활성화를 억제함을 시사한다.
실시예 2. MIR-486-5p의 간섬유화 완화 확인
CCl4로 인한 급성 간 손상이 있는 마우스에서 miR-486-5p의 직접 작용 여부를 조사하였다. 스크램블 RNA 처리된 마우스(CCl4+Scr 그룹)와 비교하여 CCl4를 주입하고 miR-486-5p 처리된 마우스(CCl4+miR-486-5p 그룹)에서 AST/ALT 양이 감소하고 간세포의 기능 마커 중 하나인 G6pc수준이 증가함으로 통해 간 손상이 개선됨을 확인할 수 있었다(도 4A, 4B 내지 도 4C).
miR-486-5p는 유의하게 상향 조절되었고, Smo, Gli2 및 섬유성 마커 인 α-Sma, Vimentin, CTGF 및 Col1α1은 CCl4+Scr 그룹과 대비하여 CCl4+ miR-486-5p 그룹에서 현저하게 하향 조절되었음을 확인할 수 있었다(도 4D).
시리우스 레드 염색 및 α-Sma에 대한 IHC 결과는 CCl4+Scr 그룹에 비해 CCl4+miR-486-5p 그룹에서 간 섬유증 및 HSC 활성화가 감소되었음을 명확하게 보여주었다(도 4E). 이러한 결과는 간섬유증에서 miR-486-5p가 Smo를 표적으로 억제하여 간 섬유증 완화에 기여함을 시사한다.
종합하여 보건데, 이러한 결과는 miR-486-5p가 헤지호그(Hedgehog, Hh) 경로를 억제하고 HSC의 활성화를 억제하여 간 섬유증 완화에 기여함을 시사한다.
본 발명에 따른 miR-486-5p는 Smo를 표적으로 하고 Hh 신호를 억제하여 HSC를 비활성화 함을 확인 하였는바, miR-486-5p는 간 섬유증에 대한 새로운 치료법이 될 수 있음을 확인하였다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

- miR-486-5p, 이의 활성화제 또는 이의 발현촉진제를 유효성분으로 포함하는, 간섬유증 개선용 건강기능식품 조성물.
