After homogenizing 10 g of the prepared sample, 50 mL of distilled water, and 0.2 mL of BHT at 10,000 rpm for 2 minutes, 47.5 mL of distilled water and 2.5 mL of 4 N HCl (Sigma Chemical, Co., St. Louis, MO, USA) were added to a TBA water machine. After using a distiller, 50 mL of the distillate was collected. 5 mL of the collected distillate and TBA (Sigma Chemical, Co., St. Louis, MO, USA) reagent are mixed in a test tube and reacted at 100°C for 35 minutes. After the reaction was completed, the absorbance was measured at 538 nm after cooling and calculated by the following formula.

5-2. 저장기간(30일, 4℃) 동안 소고기 안심의 지방산패를 확인하기 위해 지방의 2차 산화물인 말론다이알데하이드(malondialdehyde)를 정량적으로 측정하는 TBARS 실험을 실시하였다(표 2 참조). 식육은 일반적으로 0.46 MDA mg/kg부터 이취를 느낄 수 있다고 보고되었다. 저장실험 0일차에서 0.25 MDA mg/kg로 실험에 이용된 원료육이 신선한 상태임을 확인했다. 대조구에서 2일차부터 TBARS값이 상승했고 5일차부터 급격하게 상승해 30일차에서 13.95 MDA mg/kg로 완전히 산패되었다. 반면, 모든 저장기간에서 에센셜 오일을 적용한 흡수 패드로 포장된 처리구는 대조구와 비교했을 때 현저히 낮은 지방산패도를 보였다. 특히, 타임과 클로브 에센셜 오일을 이용한 포장에서는 저장기간 10일차까지 0.45 MDA mg/kg 이하의 낮은 TBARS값을 유지하였다. 5-2. 6-2. 메트미오글로빈은 근세포내 적색단백질 미오글로빈이 산화되어 갈변화 된 상태이며 식육의 색도에 직접적으로 관여한다. 미오글로빈의 산화도를 측정하기 위해 메트미오글리빈 함량을 측정했다(표 3 참조). 실험 결과 0일차에 15.84%였던 메트미오글로빈 함량은 모든 실험군에서 저장기간에 따라 상승하였고 5일차부터 에센셜 오일 처리구의 메트미오글로빈 함량이 대조구보다 유의적으로 낮았다. 10일차에 대조구는 메트미오글로빈 함량이 30%에 가까워진 반면, 에센셜 오일을 적용 포장은 18-22% 정도로 저장기간 2일차와 유사한 값을 보였다. 6-2. 에센셜 오일을 적용한 흡수 패드의 항균효과 관찰을 위하여 저장기간에 식육의 일반 세균수를 측정했다(표 4 참조). 식품의약품안전처에 따르면 식육 중 일반 세균수 기준으로 쇠고기는 식육판매장에서 6.7 log10 CFU/g 이하로 권장한다고 고시되어 있다. 0일 차에서는 대조구 및 처리구(오레가노, 타임, 클로브, 혼합)에서 총 호기성 미생물은 검출되지 않았다. 대조구는 2일 차, 에센셜 오일 처리구는 5일 차부터 총 미생물이 검출되었으며 처리구 사이에서 통계적으로 차이를 보이지 않았다. 저장기간 증가에 따라 전체 실험군에서 총 미생물 수가 통계적으로 유의미하게 증가했다. 최종 30일차에서 대조구는 6.26 log10 CFU/g로 가장 높은 값을 보였다. 처리구에서는 타임, 클로브, 혼합 에센셜 오일을 처리한 흡수 패드에서 가장 우수한 항균효과를 보였다. 2일차까지 대조구와 처리구(오레가노, 타임, 클로브, 혼합) 사이에 색깔을 포함한 외관의 차이를 보이지 않았지만 5일차의 대조구에서 부분적인 갈변 현상이 발생했다(도 3 참조). 10일차에 대조구는 전체적으로 갈변이 진행되어 상품성이 없어진 반면에 에센셜 오일을 적용한 흡수 패드로 포장된 처리구는 색깔의 선명도가 소폭 감소하는 것 이외에 육안으로 변색을 관찰할 수 없었다. 15일차에는 모든 처리구에서 색깔이 다소 어둡게 변하였지만 대조구에 비해 갈변 현상이 현저히 느리게 진행되었다. 30일차에서는 전체적으로 갈변이 진행되었지만 혼합 에센셜 오일을 적용한 흡수 패드로 식육을 포장한 처리구에서 갈변 현상이 가장 크게 지연되는 것으로 관찰됐다.
