Abstract
본 발명은 가교결합된 피브리노겐을 포함하고, 피브린을 함유하지 않는, 볼 형태의 매트릭스, 및 (a) 피브리노겐 및 혈소판 인자를 포함하는 초기 조성물을 제공하는 단계, (b) 상기 초기 조성물을 50℃ 내지 80℃의 온도로 가열된 오일로 주입하여 에멀전을 형성하는 단계, (c) 볼 형태의 매트릭스가 얻어질 때까지 상기 에멀전을 50℃ 내지 80℃의 온도에서 혼합하는 단계, 및 (d) 얻어진 매트릭스를 분리하는 단계를 포함하는 이러한 매트릭스를 제조하는 방법에 관한 것이다. 매트릭스는 세포 운반체로서 사용된다.
Description
본 발명은 볼 형태의 매트릭스, 이러한 매트릭스를 제조하는 방법 및 세포 운반체로서의 이의 용도에 관한 것이다.
본 발명은 세포 배양, 및 특히 세포 치료의 맥락에서 사용된 세포의 증식, 생물활성 분자를 제조하기 위한 생물생성, 또는 세포 모델로서의 조사 및 개발의 분야에 적용된다. 이러한 세포는 예를 들어 1차 세포, 다능성 줄기 세포, 줄기 세포주 또는 불활화 세포주이다.
특히, 성체 줄기 세포의 사용에 기초한, 세포 치료는 선택되고/되거나, 증폭되고/되거나, 생체외 변형되는 건강하고 기능성인 세포를 투여함으로써 건강하지 않은 조직의 세포의 대체 및 재생으로 이루어진다.
상이한 줄기 세포 소스가 임상 사용을 위한 충분한 분량의 세포를 얻게 하지 않으므로, 세포 증식 단계가 필요하다.
현재, 부착성 줄기 세포는 2차원 시스템, 특히 플라스틱 배양 용기에서 배양된다.
더 많은 분량의 세포를 얻기 위해, 예를 들어 아주 많은 배양 용기를 사용함으로써, 배양 용기의 크기의 증가에 의해 또는 멀티레벨 배양 용기, 예컨대 Celltack(상표명)(Corning(미국))를 사용함으로써 더 넓은 배양 표면을 사용하는 것이 필요하다. 이런 경우에, 그러나, 배양 용기의 취급은 더 수고스럽다.
3차원 배양 시스템, 특히 현탁액 중의 마이크로운반체를 사용하는 것이 또한 시험된다.

문헌 WO 2011/017050은, 세포의 부착을 수월하게 하는 RGD 서열을 포함하는 폴리펩타이드가 그래프팅된, 중합체 코팅(예를 들어, 팽윤 메타크릴레이트)을 가지는 세포 배양을 위한 동물 기원의 단백질을 가지지 않는 마이크로운반체를 제안한다.
천연 성분에 기반한 세포 마이크로운반체가 또한 존재한다.
예를 들어, 국제 특허 출원 WO 99/15637로부터 생긴, 문헌 EP 1,015,570은 세포 운반체(cell carrier)로서의 피브린(피브리노겐) 마이크로비드를 제안한다. 이 마이크로비드는 피브리노겐, 트롬빈 및 XIII 인자로부터의 마이크로비드의 전파 동안 얻어진 피브린(피브리노겐)의 높은 정도의 가교결합을 가진다. 그러나, 가교결합제로서 사용된 트롬빈은 외인성이고, 이것은 제조 어려움 및 생물학적 위험을 증가시킨다.
Eibes G. 등에 의해 문헌(Maximizing the ex vivo expansion of human mesenchymal stem cells using a microcarrier-based stirred culture system. Journal of biotechnology, 2010, vol. 146, no. 4, pp. 194-197)에서, 젤라틴 마이크로다공성 비드(Cultispher S)가 간엽성 줄기 세포를 부착시키고 이것이 증식되게 할 수 있다는 것이 밝혀졌다.
마지막으로, Oliveira M. B. 등에 의한 문헌(Injectable PEGylated fibrinogen cell-laden microparticles made with a continuous solvent- and oil-free preparation method. (c) 볼 형태의 매트릭스가 얻어질 때까지 상기 에멀전을 50℃ 내지 80℃의 온도에서 혼합하는 단계, 및

제3 양태에 따르면, 본 발명은 세포 운반체로서의 제1 양태에 따른 매트릭스의 용도에 관한 것이다.

제1 양태에 따르면, 본 발명은 가교결합된 피브리노겐을 포함하는 볼 형태의 매트릭스에 관한 것이다. 매트릭스는 특히 피브린을 함유하지 않는다.
"매트릭스"는 세포가 부착하고/하거나 증식할 수 있는 고체 또는 반고체 3차원 구조를 의미한다.
본 발명의 매트릭스는 볼의 형태이고, 즉 이것은 구형 또는 회전타원체이다. 이것은 규칙적 또는 불규칙적 형상을 가질 수 있다.
매트릭스는 피브린을 함유하지 않고, 가교결합된 피브리노겐을 포함하고, 이것은 세포 배양 동안 분해되지 않은 기계적으로 안정한 매트릭스를 얻을 수 있게 한다.

특히, 피브리노겐은 혈소판 인자에 의해 가교결합된다.
혈소판 인자는 혈소판에 의해 포함되거나 분비된 1종 또는 수종의 물질을 의미한다. 특히, 혈소판 인자는 혈소판 용해물(platelet lysate)에 함유된 물질이다.
혈소판 용해물은 혈소판의 용해의 생성물, 즉 혈소판 내에 보통 함유된 분자(성장 인자, 사이토카인)의 방출을 발생시키는 세포막의 붕괴 후 얻어진 생성물을 의미한다.
혈소판의 용해는 예를 들어 1회 또는 수회의 냉동/해동 사이클, 초음파 또는 용매/세제 처리에 의해 얻어진다.
피브리노겐의 가교결합은 혈소판 인자의 존재에 의해 가능하게 된다. 초기 혈소판 용해물 조성물 대신에 혈소판 없이 초기 혈장 조성물로부터 수행된 하기 기재된 제조 방법은 매트릭스 형성을 발생시키지 않았다. 외인성 트롬빈 없이, 따라서 피브린 형성 없이 피브리노겐의 가교결합은 1종 또는 수종의 혈소판 용해물의 존재로 인한다.
특히, 화학 물질, 예컨대 글루타르알데하이드 또는 카보다이이미드 유도체 또는 외인성 물질, 예컨대 비인간 트롬빈, 게니핀 또는 당을 첨가할 필요 없이 가교결합이 발생한다.
매트릭스는 따라서 인간 기원의 단백질을 오직 함유하는 이점을 가진다. 매트릭스는 따라서 생체적합성이고, 즉 세포, 조직 및 장기와 관련하여 비독성이다.
일 실시형태에 따르면, 매트릭스는 20㎛ 내지 2㎜, 특히 50㎛ 내지 1000㎛을 포함하는 직경을 가진다. 예를 들어, 매트릭스는 200㎛ 내지 800㎛을 포함하는 직경을 가진다.

상기 방법의 출발 생성물은 피브리노겐 및 혈소판 인자를 포함하는 초기 조성물이다. 특히, 초기 조성물은 피브리노겐 소스로서 혈장을 포함하고 혈소판 인자 소스로서 혈소판 용해물을 포함한다. 더욱이, 초기 조성물은 외인성 트롬빈을 포함하지 않고, 이것은 제조를 단순화하고, 생물학적 위험을 감소시킨다.

계면활성제는 에멀전의 형성을 수월하게 하고, 이것을 안정화시키고, 형성된 액적의 분산을 촉진하도록 작용한다.

상기 방법은 다음에 에멀전을 형성하도록 50℃ 내지 80℃를 포함하는 온도로 가열된 오일로의 초기 조성물의 주입을 포함한다. 작은 액적이 이후 형성되고 뜨거운 오일 중에 분산될 것이다.
특정한 일 실시형태에 따르면, 오일은 동물 또는 식물 오일, 특히 식물 오일, 예컨대 해바라기, 올리브 또는 캐놀라 오일이다.
열의 영향 하에, 피브리노겐이 물 및 유기 용매 중에 불용성이 되도록, 피브리노겐 단백질의 접근 불가 결합 자리가 노출되고 가교결합을 허용할 것으로 추정된다.
초기 조성물이 혈장에서 현탁액 중의 혈소판의 용해에 의해 얻어진 혈소판 용해물인 경우, 피브리노겐의 가교결합은 본질적으로, 즉 외부 물질의 사용 없이 제조된다.
이와 같이 해서 얻어진 에멀전은 다음에 혼합되어 매트릭스를 형성한다.
에멀전의 혼합의 방식은 매트릭스에 대한 영향을 미친다. 특히, 에멀전의 혼합은 분당 100회 내지 1500회 사이클, 특히 분당 700회 사이클 초과를 포함하는 속도로 교반에 의해 수행된다. 교반 속도가 더 클수록, 매트릭스가 더 작아질 것이다.
혼합은 2시간 내지 10시간, 특히 3시간 내지 5시간을 포함하는 시간 동안 수행된다.
계면활성제의 분량 및 유형을 변경함으로써 및/또는 교반 속도를 변경함으로써, 매트릭스의 크기를 제어할 수 있다.
형성된 매트릭스를 단리하는 단계는 여과, 이어서 미량의 오일을 제거하기 위한 유기 용매를 사용한 매트릭스의 연속적 세척에 의해 수행된다.
상기 방법은 상이한 크기의 매트릭스를 형성한다. 대안적으로, 상기 방법은, 매트릭스의 단리 후, 보정된 매트릭스를 얻기 위해, 크기의 감소가 동반된, 체를 통한 매트릭스의 여과의 단계를 포함한다.
본 발명에 따른 매트릭스를 얻는 방법은 50℃ 초과의 기여 열을 특히 요하는 비천연 방법이다.
혈소판 용해물로부터 이 방법에 의해 얻어진 매트릭스는 다공성이고, 본질적으로 가교결합된 피브리노겐으로 이루어지고, 피브린을 함유하지 않는다.
제3 양태에 따르면, 본 발명은 세포 운반체로서의 제1 양태에 따른 매트릭스의 용도에 관한 것이다.
세포 배양을 위해, 매트릭스를 배양 용기에 함유된 배양 배지에서 현탁액 중에 위치시킨다. 배양되어야 하는 세포를 다음에 접종시키고, 즉 매트릭스와 접촉하게 위치시킨다.
예를 들어, 매트릭스는 세포 증식, 관심 있는 단백질의 생성 및 세포 재생에 사용된다.
매트릭스는 생체적합성이어서, 이것은 생물활성 물질과 생체내 또는 이 물질 없이 사용될 수 있다.
본 발명에 따른 매트릭스의 특정한 일 이점은 이의 매우 높은 세포 부착 파워이다. 사실, 세포의 75% 초과가 접종 단계 동안 매트릭스에 부착한다.
본 발명에 따른 매트릭스를 위한 다른 분야는 예를 들어 조직 재생, 반흔, 의료 제품의 제어 방출 또는 줄기 세포 선택이다.
실시예
볼 제조
200㎖의 해바라기 오일을 65℃에 도달할 때까지 자기 교반 하에 500㎖ 용기에서 가열하였다. 50㎖의 혈소판 용해물을 이후 따뜻한 오일에 첨가하였다. 이와 같이 해서 얻어진 용액을 볼이 나타날 때까지 65℃에서 교반 하에 유지시켰다. 제제의 가열을 다음에 중지시키고, 이것이 주변 온도로 복귀할 때까지 교반을 유지시켰다. 제제를 다음에 보정된 크기를 가지는 체에서 여과시키고, 회수된 볼을 다음에 유기 용매(자일렌, 헥산 및 에탄올)를 사용하여 3회 세척하고, 식염수 용액 중에 위치시켰다.
이의 특정한 구조로 인해, 볼은 이의 분말화 형태로 또한 유지될 수 있다. 볼은 다음에 장기간 저장을 위해 주변 온도에서 또는 4℃에서 저장될 수 있다.
이와 같이 해서 얻어진 볼은 구형이고, 30㎛ 내지 2000㎛의 직경을 가진다. 스크리닝에 의해, 원하는 크기를 가지는 볼의 집단을 얻을 수 있다.
주사 전자 현미경검사법 분석은 세포 부착을 허용하는 조밀한 구조가 보일 수 있게 하였다(도 1 및 도 2).
세포에 의한 볼의 접종
완전한 배양 배지(10㎖)의 적은 용적 중에 간엽성 줄기 세포의 공지된 수와 접촉하도록 용액 중의 볼을 위치시켰다. 세포는 볼에 부착한다(도 3).
37℃에서 항온처리기 내에서 접촉의 4시간 후, (볼이 없는) 배양 배지를 수집하고 원심분리하였다. 상청액 중에 발견된 볼에 부착되지 않은 세포의 수를 계수하여서, 75%의 세포 부착률을 결정하게 할 수 있게 하였다.
세포 회수를 위한 볼의 분해
세포에 의해 충전된 볼을 충분한 세포 수를 얻기 위해 배양 중에 유지하였다. 세포 배양 기간의 종료 시, 볼/세포 복합체를 전체 배양 배지의 원심분리에 의해 회수하였다. 볼/세포 복합체로 이루어진, 회수된 펠렛을 PBS 중에 2회 세척하였다. 세척 조작 후, 세포 사멸을 발생시키지 않으면서, 볼의 전체 분해를 허용하는 효소(TrypLE(상표명))를 펠렛 위에 첨가하고, 37℃에서 20분 동안 위치시켰다. 볼의 전체 분해 후, 용액을 원심분리하고 회수된 세포를 다음에 사용할 수 있다.
세포 증식률
본 발명에 따른 볼에서의 세포 증식 시험을 폴리스타이렌 볼과 비교하여 수행하였다.
쥣과 세포주 및 인간 1차 세포를 볼의 유형에 둘 다에 동일한 밀도로 접종하였다. 부착된 세포의 분량을 정확히 결정하도록 부착되지 않은 세포의 분량을 평가하였다.
볼 위의 세포를 3일마다 새로운 배지의 첨가에 의해 10일 동안 배양 중에 유지시켰다. 배양의 종료 시, 볼/세포 복합체를 회수하고, 생성된 세포의 전체 분량을 평가하였다.
시험은, 본 발명에 다른 볼 위에서, 쥣과 세포주에 대해 23배(폴리스타이렌 볼에 대한 30배 대비) 및 인간 1차 세포주에 대해 14배(폴리스타이렌 볼에 대한 12배 대비)의 세포 증폭을 나타내도록 본 발명자들에게 허용하였다.
볼의 규명
볼을 제조한 후, 이의 조성을 정확히 연구하도록 이것을 분해하였다. 면역학적 검정은 본 발명에 따른 볼이 주로 피브리노겐으로 이루어진다(평균적으로 100㎍/㎖로 존재)는 것을 결정할 수 있게 하였다. 검정은, 본 발명에 따른 볼 내에, 0.3pg/㎖의 농도의 IGF-1의 존재가 보일 수 있게 하였다.
피브린의 존재는 볼 내에 검출되지 않았다.
Claims (17)
- 가교결합된 피브리노겐을 포함하는 볼 형태의 매트릭스로서,
피브린을 함유하지 않는 것을 특징으로 하는, 매트릭스. - 제1항에 있어서, 다공성인 것을 특징으로 하는, 매트릭스.
- 제2항에 있어서, 1㎛ 미만의 직경을 가지는 기공을 가지는 것을 특징으로 하는, 매트릭스.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 상기 피브리노겐은 혈소판 인자를 통해 가교결합된 것을 특징으로 하는, 매트릭스.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 20㎛ 내지 2㎜, 특히 50㎛ 내지 1000㎛를 포함하는 직경을 가지는 것을 특징으로 하는, 매트릭스.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 적어도 하나의 생물활성 물질, 예컨대 세포, 단백질, 예를 들어 성장 인자, 의료 제품, 호르몬, 사이토카인 또는 이들의 조합을 추가로 포함하는 것을 특징으로 하는, 매트릭스.
- 제6항에 있어서, 상기 단백질은 성장 인자, 예컨대 IGF-1인 것을 특징으로 하는, 매트릭스.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 상기 매트릭스의 표면에 세포가 부착된, 매트릭스.
- 제1항 내지 제8항 중 어느 한 항에 따른 매트릭스를 제조하는 방법으로서,
(a) 피브리노겐 및 혈소판 인자를 포함하는 초기 조성물을 제공하는 단계,
(b) 상기 초기 조성물을 50℃ 내지 80℃의 온도로 가열된 오일로 주입하여 에멀전을 형성하는 단계,
(c) 볼 형태의 매트릭스가 얻어질 때까지 상기 에멀전을 50℃ 내지 80℃의 온도에서 혼합하는 단계, 및
(d) 얻어진 매트릭스를 분리하는 단계를 포함하는, 매트릭스를 제조하는 방법. - 제9항에 있어서, 상기 초기 조성물은 혈장에서 현탁액 중의 혈소판의 용해에 의해 얻어진 혈소판 용해물(platelet lysate)인 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제10항에 있어서, 상기 혈소판 용해물은 1종 또는 수종의 혈소판 농축물로부터 얻어지는 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제9항 내지 제11항 중 어느 한 항에 있어서, 상기 초기 조성물은 계면활성제를 추가로 포함하는 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제12항에 있어서, 상기 조성물은 0.5% 내지 20%, 특히 1% 내지 5%의 계면활성제를 포함하는 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제9항 내지 제13항 중 어느 한 항에 있어서, 상기 에멀전의 혼합은 분당 100회 내지 1500회 사이클을 포함하는 속도로 교반에 의해 수행되는 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제9항 내지 제14항 중 어느 한 항에 있어서, 상기 오일은 식물 오일, 특히 올리브 오일인 것을 특징으로 하는, 매트릭스를 제조하는 방법.
- 제9항 내지 제15항 중 어느 한 항에 있어서, 상기 에멀전의 혼합은 2시간 내지 10시간을 포함하는 시간 동안 수행되는 것을 특징으로 하는, 매트릭스를 제조하는 방법.
