Abstract
본 발명은 콜라겐 생성 촉진 펩타이드에 관한 기술로서, 구체적으로는 390%의 콜라겐 생합성 촉진 효과를 보이는 신규한 콜라겐 생성 촉진 펩타이드인 바이오티노일-GG-KTTKS 및 상기 펩타이드를 포함하는 화장품 조성물에 관한 것이다.
Description
본 발명은 콜라겐 생성 촉진 펩타이드 및 상기 펩타이드를 포함하는 화장품 조성물에 관한 기술로서, 구체적으로는 종래의 콜라겐 생합성 촉진 효과가 있다고 알려진 펩타이드에 비해 더욱 우수한 콜라겐 생합성 촉진 효과를 나타내는 신규한 펩타이드에 관한 기술이다.
일반적으로 콜라겐은 동물 또는 인간의 체내에서 세포 사이를 연결하는 섬유상 경단백질로, 글리신, 프롤린, 하이드록시프롤린(hydroxyproline) 3개의 아미노산 잔기(殘基)가 서로 꼬여있는 헬릭스 구조로 이루어져 있으며, 신체의 결합 조직 및 연골 조직에 많이 분포한다. 이러한 콜라겐은 세포 간 연결, 세포 기능의 활성화, 세포 증식 작용, 지혈, 면역력 등의 기능을 가지며, 신체를 유지 및 형성하기 위해 반드시 필요한 단백질이다. 현재 콜라겐은 각종 식품, 화장품 및 의약품 등 다양한 분야에서 원료 내지 첨가제로 사용되고 있으며, 그 적용되는 산업 분야가 점점 증가하고 있는 실정이다.
본 발명자들은 콜라겐 생성 촉진 펩타이드로 알려진 KTTKS, GHK, AHK 펩타이드에 다양한 생체 유래 저분자 활성 물질을 결합시킨 후 콜라겐 생성의 촉진 여부를 분석한 결과 판토텐산(pantothenic acid)이 결합된 KTTKS 펩타이드의 콜라겐 생성 촉진 효과가 가장 뛰어난 것으로 확인되었다. 그러나 판토텐산이 결합된 KTTKS도 콜라겐 생성 촉진 활성이 270%를 넘지 못하였다.
한편, 콜라겐 생성 효과가 있는 펩타이드에 관한 선행특허로서 특허등록 제10-1342485호 "트리펩타이드 및 이를 함유하는 노화방지, 주름개선, 미백 및 항염효능의 화장료 조성물"에서는 R1-X-Leu-Phe(상기에서, R1은 수소 또는 탄소수 1~18의 아실기 중에서 선택되거나 또는 적어도 하나 이상의 이중결합을 포함하는 알케닐기이며, Leu는 D-Leu 또는 L-Leu, Phe는 D-Phe 또는 L-Phe이 될 수 있다. 또한, X로 표시되는 부분은 아미노산, 특히 20개의 자연계 아미노산과 비자연계 아미노산인 페닐글리신(Phenylglycine) 중에서 선택될 수 있으며, C-말단은 카르복실산 또는 아마이드 형태가 될 수 있다.)의 트리펩타이드로서, 기존 물질보다 콜라겐분해효소(MMP-1)를 억제하고 콜라겐 합성능이 좋은 새로운 주름개선 트리펩타이드에 관한 기술을 공개하고 있다.
또한, 특허등록 제10-1093685호에서는 팔미토일-글리시닐-프롤리닐-히드록시프롤린(Palmitoyl-Glycinyl-Prolinyl-Hydroxyproline; Palm-Gly-Pro-Hyp)으로 표시되는 신규한 팔미토일트리펩타이드 화합물을 이용한 콜라겐 생성 개선 효과를 갖는 화장품 조성물에 관한 기술을 공개하고 있다.
본 발명은 종래의 콜라겐 생성 촉진 효과가 있다고 알려진 펩타이드에 비하여 콜라겐 생성 촉진 효과가 우수한 신규한 펩타이드를 제공하기 위하여 제안되었다.
본 발명은 종래 알려진 콜라겐 생성 촉진 펩타이드보다 우수한 콜라겐 생성 촉진 효과를 갖는 신규한 펩타이드를 제공하는 것을 목적으로 한다.
특히, 본 발명은 콜라겐 생성 효능이 판토텐산 결합 KTTKS보다 우수한 신규한 펩타이드를 제공하는 것을 목적으로 한다.
특히, 본 발명은 콜라겐 생성 효능이 우수함과 더불어, 세포독성, 보존성 등의 화장품 등으로 제품화시 필요한 물성이 우수한 신규한 펩타이드를 제공하는 것을 목적으로 한다.
본 발명은 콜라겐 생성 촉진 펩타이드로서 하기 화학식 1의 바이오티노일-GG-KTTKS 펩타이드를 제공한다.

또한, 본 발명은 상기 펩타이드를 포함하는 화장품 조성물을 제공한다.
특히, 상기 화장품 조성물 중 상기 펩타이드는 0.1 ~ 99.9 중량% 포함하는 것이 바람직하다.
특히, 상기 화장품 조성물은 피부노화방지 및 주름개선효과용 화장품 조성물로 제조될 수 있다.
본 발명에서는 바이오틴(biotine)과 KTTKS(여기서 K는 라이신, T는 트레오닌, S는 세린)을 중간에서 연결하는 링커(linker) 펩타이드로서 GG(여기서, G는 글리신)를 사용하여 신규한 펩타이드인 "바이오티노일-GG-KTTKS"를 합성하였으며, 상기 합성된 펩타이드의 콜라겐 생합성 촉진능 실험 결과 390%를 보여, 콜라겐 생합성 촉진 성능이 우수하다고 알려진 판토텐산 결합 KTTKS 펩타이드의 270%보다 높은 활성을 나타냈다.
또한, 본 발명의 신규한 펩타이드인 바이오티노일-GG-KTTKS는 세포독성 실험, 용해도, 세포독성, 보관 안정성, 피부자극 등에서 매우 우수한 결과를 나타냈다.

상기 화학식 1의 바이오티노일-GG-KTTKS는 아미노산 사이의 펩타이드 결합에 의해 첨부 도 1a 및 1b와 같은 과정을 거쳐 합성되나, 첨부 도 1a 및 1b는 일 실시예로서의 합성 과정을 나타낸 것일 뿐 상기 과정 이외에 다양한 루트를 통해 바이오티노일-GG-KTTKS의 합성이 가능하다. 본 발명의 바이오티노일-GG-KTTKS는 바이오틴과 KTTKS 사이의 연결을 위해 GG 펩타이드를 링커로서 사용하였다.
본 발명의 바이오티노일-GG-KTTKS는 피부용 화장품 조성물 또는 화장품 제형으로 제조되어, 피부노화방지, 주름개선 효과를 얻을 수 있는데, 상기 펩타이드는 화장품 조성물 전체에서 0.1 내지 99.9 중량%로 사용되는 것이 바람직하며, 나머지 성분은 부형제, 영양 성분 등 공지의 화장품 성분을 더 추가할 수 있다.
이하에서는 실험을 통해 본 발명의 펩타이드의 콜라겐 생성 촉진 효과 및 기타 물성에 대하여 자세히 설명하기로 한다.
[실험예 1] 세포 독성 확인
본 발명의 바이오티노일-GG-KTTKS의 콜라겐 생성 촉진 활성 검증에 앞서, 정상 섬유아세포를 이용한 세포독성을 확인하였다. 이를 확인하기 위해 정상 섬유아세포를 96 웰플레이트에 배양하면서 본 발명의 펩타이드뿐만 아니라 비교예의 다양한 펩타이드 유도체로 48시간 동안 처리한 후 MTT 어세이를 이용하여 세포 생존율을 검증하였다.
그 결과 첨부 2와 같이, 본 발명의 바이오틴이 결합된 펩타이드 유도체는 100uM 농도까지는 어떠한 세포 독성도 보여주지 않았으며, 200uM 처리 시 10% 정도의 세포생존율 저하가 나타내어, 본 발명의 펩타이드의 피부 도포시 세포 독성으로 인한 특별한 문제는 발생하지 않을 것임을 알 수 있었다.

2] 콜라겐 생합성
촉진능의
분석
본 발명의 펩타이드 유도체의 콜라겐 생합성 촉진능을 분석하기 위해, p12 섬유아세포에 본 발명의 펩타이드 및 비교예의 펩타이드를 처리하고 48 시간 후에 세포 배양액을 수거하여, "anti-collagen antibody/biotinylated anti-collagen antibody"를 이용한 "sandwich ELISA assay"를 이용해 배양액 내 콜라겐양을 정량적으로 분석하였다.
도 3과 같이, 본 발명의 펩타이드 및 비교예의 펩타이드들 모두 농도의존적인 콜라겐 생합성 촉진 효과를 나타내었으며, 그 중에서도 본 발명의 GG 링커가 삽입된 바이오티노일-GG-KTTKS 펩타이드가 200μM에서 390%의 콜라겐 생합성 촉진 효과를 보여주었으며, 비교예의 펩타이드들에 비하여 매우 높은 생합성 촉진 효과를 보여주었다. 특히, 판토텐산-KTTKs에 비하여 매우 높은 콜라겐 생합성 촉진 효과를 보여주어, 종래 기술에 비하여 본 발명의 펩타이드의 우수성을 확인할 수 있었다.

실험예
3] 용해도(Solubility) 확인
본 발명의 바이오티노일-GG-KTTKS 펩타이드의 용해도를 확인하기 위해 "turbidometric solubility assay"를 수행하였다. 이를 위해, 바이오티노일-GG-KTTKS 펩타이드 및 KTTKS 펩타이드를 증류수(DW)에 20mg/ml, 10mg/ml, 5mg/ml, 2.5mg/ml, 1.25mg/ml, 0.625mg/ml로 녹이고 2시간 동안 37℃에서 인큐베이션한 후 620nm에서 흡광도(absorbance)를 측정하였으며, 대조(control) 물질로 피렌(pyrene)을 사용하였다.
도 4와 같이, KTTKS 펩타이드는 20mg/ml까지 물에 잘 녹는 것으로 나타났으며, 바이오티노일-GG-KTTKS 펩타이드는 10mg/ml까지 물에 잘 녹았고 20mg/ml에서는 일부 침전이 발생하여 UV 흡광도가 증가하는 것으로 나타났다. 비록 KTTKS 펩타이드에 비해 용해도는 나빴으나 이 정도의 용해도면 제품화시 문제가 없다는 것을 확인할 수 있었다.

4] 보관
안정성
본 실험예에서는 바이오티노일-GG-KTTKS 펩타이드의 보관 안정성을 측정하였다. 이를 위해, 바이오티노일-GG-KTTKS 펩타이드를 증류수에 0.1mg/ml로 용해한 후 각각 상온(25℃) 및 고온(40℃)에서 보관하면서, 잔존 바이오티노일-GG-KTTKS의 함량을 HPLC로 분석하였다.
도 5와 같이, 바이오티노일-GG-KTTKS 펩타이드는 12주 동안 상온 및 고온에서 각각 96.8% 및 99.3%가 잔존하는 것으로 나타나, 안정성이 매우 뛰어남을 확인할 수 있었다.

실험예
5] 보관 안정성과 보관 활성과의 상관관계 확인
본 실험에서는 콜라겐 생합성 효과가 가장 뛰어난 펩타이드 유도체인 바이오티노일-GG-KTTKS 펩타이드의 보관 안정성과 보관 활성과의 상관관계를 확인하기 위해, 상기 펩타이드를 녹이고 보관하는 시점에 따른 콜라겐 생합성의 변화를 측정하였다. 이를 위해, 비교예로서 판토텐산-KTTKS와 본 발명의 바이오티노일-GG-KTTKS를 각각 증류수에 1mM로 녹이고 40℃에서 2주일, 1주일 보관 및 새로 녹인 펩타이드 용액을 각기 준비하여 96 웰플레이트에 배양중인 p9 섬유아세포에 각각의 농도로 48시간 처리한 후 배양액을 수거하여, "sandwich ELISA"를 이용하여 콜라겐 생합성량을 정량하였다.
그 결과 첨부 도 6과 같이, 판토텐산-KTTKS 펩타이드는 보관 시간이 증가할수록 콜라겐 생합성 효과가 저하되는데 반해, 본 발명의 바이오티노일-GG-KTTKS 펩타이드는 활성의 저하가 나타나지 않았다. 이를 통해 본 발명의 바이오티노일-GG-KTTKS 펩타이드는 보관 안정성뿐만 아니라 보관 활성도가 우수함을 확인할 수 있었다.

실험예
6] 피부 자극 확인
본 실험예에서는 화학물질의 피부자극여부를 분석하기 위해 "in-silico skin sensitization" 분석법인 DPRA(Direct Peptide Reactivity Assay)를 수행하였다. 이는 OECD/OCDE 가이드라인에 따른 시험법이며, 피부에 접촉되는 화학물질의 민감물질이 생체 물질과의 반응에 의해 야기되는 현상으로, 특히 생체 물질에 포함되어 있는 라이신의 -NH2기나 시스테인의 -SH기와의 반응을 통해 나타나게 됨을 이용한 실험법이다.
DPRA 테스트에서는 반응성 있는 NH2 공급원으로 Ac-RFAACAA-COOH 펩타이드를 사용하며, -SH 공급원으로 Ac-RFAAKAA-COOH 펩타이드를 사용한다. 본 실험 진행을 위해 Ac-RFAACAA-COOH 펩타이드 혹은 Ac-RFAAKAA-COOH 펩타이드와 본 발명의 바이오티노일-GG-KTTKS 펩타이드를 섞고 24시간 반응하여, 초기 펩타이드 용액과 반응 후 펩타이드 용액 내의 펩타이드 함량의 변화를 HPLC로 분석하였다.
분석 결과, 본 발명의 펩타이드는 지표 펩타이드인 Ac-RFAAKAA-COOH(도 7 참조) 및 Ac-RFAACAA-COOH(도 8 참조) 펩타이드와의 반응성이 전혀 없는 것으로 나타나, 본 발명의 펩타이드는 피부자극이 없음을 확인하였다.
