Abstract
본 발명은 본 발명은 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효 성분으로 함유하는 암 예방 또는 치료용 약학적 조성물 및 건강기능식품에 관한 것이다. 본 발명의 상기 피라졸 아마이드 화합물은 스탯3 단백질의 활성을 억제함으로써, 암세포의 성장을 억제하는 효과가 있으므로, 암 예방 또는 치료용 약학적 조성물에 유용하게 사용할 수 있다.
Description
본 발명은 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효 성분으로 함유하는 암 예방 또는 치료용 약학적 조성물에 관한 것이다.
문명이 발달되어 갈수록 암의 발생률이 증가되고 있어 항암제 개발은 생명과학관련 기술개발연구의 가장 핵심을 차지하고 있고, 암은 우리나라 뿐 아니라 미국, 일본 등에서도 사망원인 1-2 위를 다투고 있으며 특히 대형병원 사망률의 과반수이상을 차지하고 있다(생명과학동향, 생명공학 연구소, 3: 55, 1995; 鶴尾降, 西條長壙, 암 화학적 요법, 서광서림, 351, 1991). 이러한 암을 정복하기 위한 노력의 일환으로 여러 가지 암의 조기 발견을 위한 진단법에 많은 발전이 있었으며, 암의 치료법에 있어서도 수술, 방사선 요법, 화학적 요법, 생물학적 방법 등이 발달해 왔다.
그 중 화학적 요법의 경우 다양한 종류의 암세포를 이용한 시도가 많았지만, 현재까지 진정한 의미의 치료제로서의 항암제는 없는 상태이고 단지 보조치료제 내지는 단기간의 생명연장을 돕는 정도의 약제밖에는 개발되지 못한 상태이다. 또한, 상기 치료법들은 대개 조기 암 환자나 특정 암에만 처치가 국한되어 있어서 암으로 인한 사망은 계속 증가하고 있는 추세이다.
최근에는 암 치료의 가능성을 높이기 위하여 암의 발생과 전이, 암세포의 생리, 암의 진단과 치료에 대한 연구 및 천연 추출물 검색을 통하여 일반적인 항암제 뿐만 아니라, 혈관신생억제제(angiogenesis inhibitor), 암 전이 억제제 새로운 암 치료제를 개발하고 있는 추세이다. 그 중 특정 분자 목표점에 작용하는 선택적 항암제는 보다 안전하고 효과적인 치료법을 제공할 뿐 아니라 맞춤 의약 및 병행치료법(combination therapy)에 적용될 수 있어 더욱 주목받고 있다.
신호전달 및 전사조절 단백질로서, 84~113 kDa의 분자량을 갖는 스탯(STAT)단백질[스탯1(STAT1), 스탯2(STAT2), 스탯3(STAT3), 스탯4(STAT4), 스탯5(STAT5) 및 스탯6(STAT6)]은 각각 활성화된 수용체의 세포질 내에 1 또는 2 이상의 포스포 티로신 서열을 인식할 수 있는 SH2 도메인을 포함한다.
상기 SH2(Src Homology-2) 도메인은 수용체 인식 및 DNA 결합을 조절하는 인산화 의존성 스위치로서의 역할을 수행한다. 그 결과 스탯(STAT) 단백질이 세포표면 수용체의 활성을 유전자 조절에 직접적으로 연결시킬 수 있게 한다(Darnell, J. E., Proc. Natl. Acad. Sci. (USA), 94:11767-11769 (1997)).
동물세포에서 잠재성 세포질 스탯(STAT) 분자의 활성화는 사이토카의 표면 수용체 및 이들과 비공유적으로 연결되어 있고 야크(Jak) 키나아제, 또는 고유의 티로신 키아나제 활성을 갖는 성장 인자 수용체에 의해 이루어진다.
상기 세포 표면에 동질의 리간드의 결합은 상기 수용체의 세포질 영역 내에 티로신의 인산화를 유발함으로써, 스탯(STAT) SH2 도메인의 결합자리를 만들게 된다. 상기 수용체에 스탯(STAT)의 결합은 야크 또는 수용체 키나아제에 의한 티로신 상의 인산화를 유발한다. 인산화된 스탯(STAT) 단백질은 SH2 도메인에 의해 매개되는 이량체(dimer)를 형성한 후, 핵 내부로 이동하고, 핵 내부에서 이들은 DNA와 결합하고 특정 유전자의 전사개시를 유도한다.이런 스탯(STAT) 단백질의 신호체계는 탈인산화 작용 및 단백질 분해에 의해서 정지될 수 있다.
연구자들은 다양한 암종에서 스탯(signal transducers and activators of transcription) 단백질의 활성화된 형태를 발견하였는데, 특히 스탯3(STAT3)는 백혈병과 같은 혈액암 뿐만 아니라, 유방암, 두부경부암, 흑색종, 난소암, 폐암, 췌장암, 전립선암과 같은 고형암에서 활성화 되어있어서 중요한 항암 목표점을 제시하고 있다[Hua Yu and Richard Jove, Nature Review Cancer (2004), 8, 945].
따라서, 스탯 단백질의 억제는 세포사멸, 신생혈관 억제, 면역회피 차단의 복합적 항암 기작을 통해서 종양을 제어함으로써 보다 효과적이고 실질적인 항암제 개발의 원천기술로 중요하며, 기존의 단선적 작용을 하는 항암제에 비해 높은 치료효과를 기대할 수 있다.
이에, 본 발명자들은 스탯 단백질을 억제할 수 있는 화합물을 발굴하기 위해 노력한 결과, 본 발명의 화합물들이 스탯 단백질의 활성을 억제하는 것을 확인하였고, 이를 이용해 다양한 인체 암 세포주의 성장을 억제시키는 것을 확인하여 본 발명을 완성하였다.
본 발명의 목적은 피라졸 아마이드 화합물을 유효 성분으로 함유하는 암 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여,
본 발명은 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암 예방 조성물을 제공한다.
또한, 본 발명은 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명의 피라졸 아마이드 화합물은 스탯 단백질(signal transducers and activators of transcription protein, STAT protein)의 인산화를 억제하여 항암 효과를 나타내므로, 상기 피라졸 아마이드 화합물은 암 예방 또는 치료용 조성물 로 유용하게 사용될 수 있다.

이하, 본 발명에 사용된 용어를 설명한다.
본 발명에 사용된 용어 "항암"은 암세포의 증식을 억제하거나 사멸하는 작용을 의미하는 것으로 암의 예방 및 치료 모두를 의미한다.
본 발명에 사용된 용어 "예방"은 조성물의 투여로 암형성을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에 사용된 용어 "치료"는 조성물의 투여로 질환의 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명에 사용된 용어 "투여"는 임의의 적절한 방법으로 환자에게 소정의 물질을 제공하는 것을 의미한다.
본 발명에 사용된 용어 "환자"는 본 발명의 조성물을 투여하여 증상이 호전될 수 있는 질환을 가진 인간과 원숭이, 개, 염소, 돼지 또는 쥐 등의 동물을 의미한다.
본 발명에 사용된 용어 "약학적으로 유효한 양"이란, 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미한다.
본 발명에 사용된 용어 "건강기능식품"이란, 일상 식사에서 결핍되기 쉬운 영양소나 인체에 유용한 기능을 가진 원료나 성분(기능성원료)을 사용하여 제조한 것으로, 인체의 정상적인 기능을 유지하거나 생리기능 활성화를 통하여 건강을 유지하고 개선하는 식품으로 식품의약안전청장이 정한것을 의미하나, 이에 한정되지 않으며 통상적인 의미의 건강식품을 모두 포함하는 의미로 사용한다.
본 발명에 사용된 용어 "이중 루시퍼라제(dual luciferase)"란, 두종류의 루시퍼라제를 사용하는 방법을 의미하며, 구체적으로 일반적인 Luciferase assay에서는 세포의 수와 상태에 따라 발광량이 달라지게 되므로, 보다 정확하게 측정하기 위하여 이런 영향을 보정하기 위한 내부 표준물이 필요한데, 목적 유전자의 측정을 위한 반딧불 루시퍼라제(firefly Luciferase gene)과 컨트롤(control)로서 전사활성이 일정한 유전자의 프로모터(promoter)에 Renilla Luciferase gene을 연결한 벡터가 이용되고 있다. 이 벡터를 이용할 경우에는 반딧불 루시퍼라제에서의 발광량을 측정한 후에 Renilla Luciferase의 발광량을 측정한다. 다음에 Renilla Luciferase의 발광량을 1로 하였을 때의 반딧불 루시퍼라제의 상대 발광량을 구한 후, 이것을 보정치로 하고 있다.
이하, 본 발명을 상세히 설명한다.
본 발명은 하기 화학식 1로 표시되는 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 암 예방 또는 치료용 약학적 조성물을 제공한다.
상기 피라졸 아마이드 화합물은 C22H19FN4O의 분자식을 갖는 화합물로서, 이의 이화학적 성질을 살펴보면, 알코올, DMSO 등의 극성 용매에는 잘 녹고, 헥산 등의 비극성 용매에는 녹지 않으며, 125℃의 융점 및 374.42의 분자량 값을 갖는 백색 고체 분말이다.
본 발명의 조성물은 약제학적으로 허용 가능한 첨가제를 더 포함할 수 있으며, 이때 약제학적으로 허용 가능한 첨가제로는 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 사용될 수 있다. 본 발명에 따른 약제학적으로 허용 가능한 첨가제는 상기 조성물에 대해 0.1 ~ 90 중량부 포함되는 것이 바람직하나 이에 한정되는 것은 아니다.
즉, 본 발명의 조성물은 실제 임상 투여 시에 경구 및 비경구의 여러 가지 제형으로 투여될 수 있는데, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 스탯3 단백질의 발현 또는 활성 억제제에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(Calcium carbonate), 수크로스(Sucrose), 락토오스(Lactose) 또는 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용될 수 있다.
경구를 위한 액상 제제로는 현탁제, 내용액제, 유제 및 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함될 수 있다. 비수성용제, 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여할 수 있으며, 비경구 투여시 피부 외용 또는 복강내주사, 직장내주사, 피하주사, 정맥주사, 근육내 주사 또는 흉부내 주사 주입방식을 선택하는 것이 바람직하다. 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다.
투약 단위는, 예를 들면 개별 투약량의 1, 2, 3 또는 4배를 함유하거나 또는 1/2, 1/3 또는 1/4배를 함유할 수 있다. 개별 투약량은 구체적으로 유효 약물이 1회에 투여되는 양을 함유하며, 이는 통상 1일 투여량의 전부, 1/2, 1/3 또는 1/4배에 해당한다. 본 발명의 조성물의 유효용량은 0.0001 ~ 10 g/㎏일 수 있고, 구체적으로 0.001 ~ 1 g/kg일 수 있으며, 하루 1 ~ 6회 투여될 수 있다. 그러나, 투여 경로, 질병의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 조성물은 신경보호를 위하여 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
또한, 본 발명의 상기 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물은 스탯(signal transducers andactivators of transcription) 단백질의 활성을 억제하는 것을 특징으로 하는 약학적 조성물을 제공하나, 이에 한정되지 않는다.
또한, 본 발명의 상기 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물은 cyclin D, survivin, TBX21, CD40, CD80, CDKN1A, caspase, IL-12, IL-17, IL-23, BCL-XL, BCL-2, MCL-1, CCND1, CCND2, VEGF, INFγ, GATA3 및 FOXP3로 이루어진 군으로부터 선택되는 하나 이상의 유전자의 발현을 억제하는 것을 특징으로 하는 약학적 조성물을 제공하나, 이에 한정되지 않는다.
또한, 상기 암은 대장암, 치종암, 혈액암, 후두암, 식도암, 구강암, 기저세포암, 담도암, 갑상선암, 직장암, 위암, 전립선암, 유방암, 신장암, 간암, 뇌종양, 폐암, 자궁암, 결장암, 방광암 및 췌장암으로 이루어진 군으로부터 선택되는 어느 하나인 약학적 조성물을 제공하나, 상기 기재된 외에 당업자에 알려진 모든 공지의 암을 포함한다.
또한, 본 발명의 상기 화학식 1로 표시되는 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염을 유효성분으로 하는 암 예방 치료 또는 치료용 약학적 조성물은 암 예방 또는 개선용 건강 기능식품을 제공하며, 이에 한정되지 않는다.
상기 암은 대장암, 치종암, 혈액암, 후두암, 식도암, 구강암, 기저세포암, 담도암, 갑상선암, 직장암, 위암, 전립선암, 유방암, 신장암, 간암, 뇌종양, 폐암, 자궁암, 결장암, 방광암 및 췌장암으로 이루어진 군으로부터 선택되는 어느 하나인 약학적 조성물을 제공하나, 상기 기재된 외에 당업자에 알려진 모든 공지의 암을 포함한다.
본 발명의 구체적인 실시예에 있어서, 본 발명 피라졸 아마이드 화합물에 의한 스탯3(STAT3) 단백질의 억제 활성 효과를 확인하기 위하여, 직장암 세포주(HCT116)에 본 발명의 피라졸 아마이드 화합물을 투여하고 이중 루시퍼라제법(dual luciferase assay)을 통해 스탯3(STAT3)의 활성을 측정하였다.
그 결과 스탯3(STAT3)의 활성에 비례하여 발현이 증가하는 반딧불 루시퍼라제를 감염시킨 인간 대장암 세포주(HCT116)에서 본 발명의 화학식 1 화합물이 스탯3(STAT3)의 활성을 저해함을 확인하였고(도 1),
또한, 본 발명의 구체적인 실시예에 있어서, 본 발명자들은 피라졸 아마이드 화합물이 스탯3 단백질의 활성을 억제하는 기작을 확인하기 위하여, 피라졸 아마이드 화합물을 처리한 전립선암 세포주(DU-145)에 대해 웨스턴 블랏(western blot)을 통해 인산화된 스탯3 단백질의 양을 측정한 결과, 인산화된 스탯3 단백질이 감소함을 확인함으로써, 피라졸 아마이드 화합물에 의한 스탯3의 활성 저해는 스탯3의 인산화 감소에 의한 것임을 알 수 있었다(도 2).
또한, 본 발명의 구체적인 실시예에 있어서, 본 발명자들은 스탯3 단백질의 활성 저하에 의한 유전자 발현변화를 확인하기 위하여, 암 관련 유전자로 알려진 사이클린 D(cyclin D), 사이클린 A(cyclin A), BCL-2, survivin의 단백질의 발현수준 변화를 웨스턴 블랏에 의해 측정한 결과, 상기 단백질들의 발현이 감소함을 확인할 수 있었고(도 3),
또한, 본 발명의 구체적인 실시예에 있어서, 피라졸 아마이드 화합물이 암세포의 성장을 억제하는지 확인하기 위하여, 유방암 세포주(MDA-MB-231, MDA-MB-468), 전립선암 세포주(DU-145), 대장암 세포주(HCT116), 췌장암 세포주(AsPC-3), 결장암 세포주(SW620)에 대하여 각각 피라졸 아마이드 화합물을 처리하고 엘라이자 방법(ELISA)에 의해 흡광도를 측정한 결과, 15-30uM의 농도범위에서 50%의 성장억제 효과를 확인함으로써, 피라졸 아마이드 화학물에 의한 암세포의 성장 억제 효과를 확인하였다(도 4).
또한, 본 발명의 구체적인 실시예에 있어서, 본 발명자들은 피라졸 아마이드 화합물이 in vivo에서 항암 효과가 있는지 확인하기 위하여, 인체유래 전립선암 세포(DU-145)를 암세포에 이식하여 종양세포가 형성된 마우스에 대하여, 피라졸 아마이드 화합물을 투여하고 상기 마우스에서 종양을 분리한 후 종양 조직의 크기와 무게를 측정한 결과, 피라졸 아마이드 화합물의 투여에 의해 종양 조직의 크기 및 무게가 감소함을 확인함으로써, 피라졸 아마이드 화합물이 in vivo에서도 암세포 성장 억제 효과가 있음을 확인할 수 있었다(도 4).
이하, 본 발명을 하기 실시예에 의해 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다.
통계처리
실험결과는 평균±표준편차로 표기하였고, Student's t-test로 분석하여 p값이 0.05 미만일 때 통계적으로 유의하다고 판단하였다.
<실시예 1> 피라졸 아마이드 화합물의 스탯3(STAT3) 활성 억제 확인
본 발명의 피라졸 아마이드 화합물에 의한 스탯3(STAT3) 활성 억제 효과를 알아보기 위해 이중 루시퍼라제법(dual luciferase assay)을 사용하여 하기의 실험을 수행하였다. 상기 피라졸 아마이드 화합물(CH-806)은 한국화합은행으로부터 분양받았다.
직장암 세포주(HCT116)에 스탯3(STAT3)의 활성에 비례하여 반딧불 루시퍼라제(Firefly luciferase)의 발현이 증가하는 플라스미드와 스탯3(STAT3)의 활성과 무관하게 레닐라 루시퍼라제(Renilla luciferase)를 발현하는 플라스미드를 동시에 감염시켰다. 플라스미드들이 감염된 직장암세포들은 0.05% 트립신-EDTA(trypsin-EDTA)를 이용하여 떼어내어 96-웰 검정 플레이트의 각각의 웰에 10,000개의 세포를 접종하였다. 10% FBS가 포함된 배지에서 5% 이산화탄소를 포함하는 37 ℃ 배양기에 3시간 배양하고, 대조군(1% DMSO)과 피라졸 아마이드 화합물(DMSO에 녹인 화합물)을 농도별로 첨가하였다(1/100배 희석).
스탯3(STAT3) 활성은 12시간 또는 24시간 경과 후에 반딧불 루시퍼라제(Firefly luciferase)와 레닐라 루시퍼라제(Renilla luciferase)에 특이적인 별도의 기질(Beetle luciferin과 coelenterazine)을 순차적으로 첨가한 후 측정하였다. 기질의 분해에 의한 발광 강도는 월락사의 루미노메터를 이용하여 측정하였다(Luminometer, Wallac 1420).
그 결과, 투여된 피라졸 아마이드 화합물의 농도 증가와 비례하여 스탯3의 발현이 감소하였으며, 특히 피라졸 아마이드 화합물의 농도가 10μM일 때, 스탯3의 활성이 50% 감소함을 확인한 결과, 피라졸 아마이드 화합물은 스탯3 단백질의 활성 억제 효과가 있음을 확인하였다(도 1).
<실시예 2> 피라졸 아마이드 화합물에 의한 항암효과의 기전 확인
<실시예 2-1> 피라졸 아마이드 화합물에 의한 스탯3 단백질의 활성 억제의 기전 확인
본 발명자들은, 피라졸 아마이드 화합물에 의한 스탯3 단백질의 활성 감소가 스탯3의 인산화 감소에 의한 것인지 확인하기 위하여 웨스턴 블랏을 통해 인산화된 스탯3의 발현변화를 측정하였다.
구체적으로, 전립선암 세포주(DU-145)를 세개의 60mm 플레이트에 각각 80,000개씩 분주하여 5% C02 , 37℃, 10% FBS 배지환경에서 배양하였다. 24시간 후, 각각의 플레이트에 0.25% DMSO(대조군)과 20ug/ml 및 50ug/ml 농도의 활성 분획물을을 처리하고 다시 24시간 배양하였다. 그 후, 각각의 플레이트의 배지를 버리고 PBS를 이용해 워싱(washing)한 후, Ripa lysis buffer를 200ul씩 넣고 스크래퍼(scraper)를 이용하여 플레이트에 붙어있는 세포를 포집하였다. 포집된 세포들을 1.5ml 튜브로 옮긴 후, 4℃ centrifuge에서 13,000rpm으로 15-30분간 원심분리하여, 상층액을 새로운 1.5ml 튜브로 옮기고 각각의 용해된 단백질의 양을 측정하였다.
상기 튜브에 80ul의 3차 증류수와 각각의 용해물(lysate) 10ul을 넣고 bradford assay 시약 200ul을 넣은후 흔들어(vortex) 96웰 플레이트(96 well plate)에 넣었다.
이후, ELISA 판독기를 이용하여 595nm에서 흡광도를 측정하여 각각의 용해물의 단백질을 정량하고, 같은 양의 단백질이 들어가게 계산하여 용해물과 용해버퍼(lysis buffer), 5x 염료(dye)를 이용하여 로딩샘플을 만들었다. 상기 로딩샘플은 80℃에서 10분간 가열하여 비활성화시켰다. 그 후, 인산화된 스탯3 단백질에 대한 아크릴아마이드 SDS 젤을 만들고, 상기의 로딩샘플을 로딩한 후, 0.25A에서 2시간 동안 트랜스퍼하여 젤에 있는 단백질을 멤브레인으로 옮겼다.
상기 옮겨진 단백질은 탈지유(skim milk)로 1시간 동안 블로킹(blocking)한 후, 인산화된 스탯3 단백질(p-stat3)의 항체(1차 항체, Cell Signaling Technology)를 결합시킨 후, 1차 항체에 대한 2차 항체(Cell Signaling Technology)와 1시간 반응시킨 후, 멤브레인에 substrate를 분사한 후, LAS 이미지 분석기를 이용해 화학발광(chemoluminescence)를 측정하여 p-stat3 발현양을 측정하였다.
그 결과, 스탯3의 발현양은 변화가 없었으나 인산화된 스탯3의 발현양은 감소하였고, 피라졸 아마이드 화합물의 처리에 의한 스탯 3 단백질의 활성 저하는 스탯3의 인산화의 감소에 의한 것임을 확인할 수 있었다(도 2).
<실시예 2-2> 스탯3 인산화 감소에 의한 유전자 발현 변화 확인
본 발명자들은, 피라졸 아마이드 화합물의 처리에 의한 스탯3의 타겟 유전자의 발현 변화를 확인하기 위하여, 상기 <실시예 2-1>과 같은 방법으로 사이클린 A(cyclin A), 사이클린 D(cyclin D), BCL-2 및 survivin 단백질의 발현양을 측정(β-actin은 control)하였다.
그 결과, 피라졸 아마이드 화합물의 처리에 의해 스탯3의 타겟 유전자인 상기 사이클린 A(cyclin A), 사이클린 D(cyclin D), BCL-2 및 survivin의 발현양이 감소함을 알 수 있었다(도 3).
<실시예 3> 암세포주의 성장 억제
본 발명자들은, 피라졸 아마이드 화합물이 인간 암세포주에 대한 성장 억제 효과를 확인하기 위해 WST-1 assayf를 통해 유방암 세포주(MDA-MB-231, MDA-MB-468), 전립선암 세포주(DU-145), 대장암 세포주(HCT116), 췌장암 세포주(AsPC-3), 결장암 세포주(SW620)에 대하여 피라졸 아마이드 화합물을 처리한 후, 시간경과에 따른 암세포의 성장율을 측정하였다.
구체적으로, 상기 암세포주들을 10% 소태아혈청(FBS)이 포함된 배지에서 37 ℃, 5% 이산화탄소를 유지하며 배양하였고, 0.05% 트립신-EDTA(trypsin-EDTA)를 이용하여 세포를 수득하였다. 96-웰 플레이트의 각 웰에 헤마토사이토미터(hematocytometer)로 계산한 5,000개(MDA-MB-231, MDA-MB-468), 7,000개(HCT116, SW620)의 세포를 각각 분주하였다.
그 후 5% 이산화탄소를 포함한 37 ℃ 배양기에서 10% FBS가 포함된 배지에서 배양하였고, 24시간 후에 대조군(0.1 % DMSO)과 합성한 피라졸 아마이드 화합물을 농도별로 포함(DMSO에 녹인 화합물을 배지로 희석)한 배지로 교환하였다. 24시간 동안 처리한 후에 각 웰에 WST-1(Roche사 제품) 발색시약을 10 μl씩을 첨가하고 2시간 동안 배양한 후, ELISA 판독기(ELISA Reader, Bio-Rad사 제품)를 이용하여 450 ㎚에서 흡광도를 측정하였다.
그 결과, 각 암세포들은 피라졸 아마이드 화합물의 처리에 의하여 시간 경과에 따라 대조군에 비하여 감소한 세포 성장률을 나타냄을 확인하였다. 구체적으로, 각각의 암세포는 15-30 μM의 농도범위에서 50% 성장 억제 효과가 관찰하였고, 특히 성장 억제가 가장 효과적으로 나타난 세포는 HCT116으로써 GI50이 15 μM이었고, 그 외에 MDA-MB-468과 SW620은 각각 40과 70 μM의 GI50을 나타내었으며, MDA-MB-231은 100 μM 이내의 농도에 의하여 50% 이상의 성장 저해를 나타내지는 않았다.
<실시예 4> 동물모델에서 피라졸 아마이드의 항암효과 확인
본 발명자들은, 피라졸 아마이드 화합물이 in vivo에서 항암 효과를 나타내는지 확인하기 위하여 하기의 시험을 수행하였다.
액체 질소에서 냉동보관한 인체유래 전립선암 세포주(DU-145)를 상온에서 해동한 후 CO2 인큐베이터(Forma, USA)에서 37℃, 5% CO2 환경에서 배양하였다.
상기 배양된 암세포는 무혈청 배지(serum free media)를 이용하여 3x107 cells/mouse의 농도로 준비하였다. 그 후 상기 암세포 배양액을 각각의 마우스(BALB/C 계통의 특정병원체 부재 암컷 누드 마우스, 6주령, Nara Biotech Co.)에 0.3ml(9x106 cells/mouse)씩 우측 견갑부와 흉벽사이의 액와부위 피하에 주입하였다.
상기 암세포 이식 후 측정가능한 종양이 형성되었을 때부터 25일째까지 개체별로 총 8회 버어니어 캘리퍼스(vernier caliper)를 이용하여 3 방향을 측정한 후, 암세포의 길이, 폭 및 높이를 측정하여 길이x폭x높이/2dml 계산식으로 표현하였다.
그 후, 상기 마우스에 10%의 디메틸아세트아마이드, 10%의 Tween80 및 20%의 하이드록실프로필-베타-사이클로덱스트린(hydroxypropyl-β-cyclodextrin) 용액을 혼합한 80%의 용매를 이용하여 1 mg/ml 및 3 mg/ml의 피라졸 아마이드 화합물 용액(CG-806)을 제조하여, 10mg/kg 용량으로 마우스에 20g 당 0.2ml 씩 16회 반복하여 복강투여하였다.
상기 피라졸 아마이드 화합물 용액을 투여하고 25일이 경과한 후, CO2로 마우스를 치사시킨 후 종양을 분리하여 종양 조직의 크기와 무게를 측정하였다.
그 결과, 대조군 마우스와 비교하여 CG-806 10mg/kg 투여한 실험군에서 약 37.5%의 통계적으로 유의한 종양성장 억제가 확인되었고, 종양을 절제하여 그 무게를 측정한 결과 대조군과 비교하여 37.6%의 종양무게 감소를 확인하였는바, 피라졸 아마이드 화합물이 in vivo에서 항암 효과가 있음을 알 수 있었다(도 4).
Claims (5)
- 제 1항에 있어서, 상기 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염은 스탯(signal transducers andactivators of transcription, STAT) 단백질의 활성을 억제하는 것을 특징으로 하는 약학적 조성물.
- 제 1항에 있어서, 상기 피라졸 아마이드 화합물 또는 이의 약학적으로 허용가능한 염은 cyclin A, cyclin D, survivin, TBX21, CD40, CD80, CDKN1A, caspase, IL-12, IL-17, IL-23, BCL-XL, BCL-2, MCL-1, CCND1, CCND2, VEGF, INFγ, GATA3 및 FOXP3로 이루어진 군으로부터 선택되는 하나 이상의 발현을 억제하는 것을 특징으로 하는 약학적 조성물.
- 제 1항에 있어서, 상기 암은 대장암, 치종암, 혈액암, 후두암, 식도암, 구강암, 기저세포암, 담도암, 갑상선암, 직장암, 위암, 전립선암, 유방암, 신장암, 간암, 뇌종양, 폐암, 자궁암, 결장암, 방광암 및 췌장암으로 이루어진 군으로부터 선택되는 어느 하나인 약학적 조성물.
- 제 2항에 있어서, 상기 STAT 단백질은 STAT1, STAT2, STAT3, STAT4 , STAT5 및 STAT6 단백질로 이루어지는 군으로부터 선택되는 어느 하나인 것을 특징으로 하는 약학적 조성물.
