Abstract
Description
The present invention relates to a process for producing ornithine-containing kimchi which can increase the content of ornithine by inducing an increase in the number of lactic acid bacteria in the process of manufacturing kimchi, and an oropharyngeal kimchi produced by the process.
Kimchi is a traditional fermented food which is fermented at low temperature in order to preserve the preservability and aging of the product after mixing red pepper powder, garlic, ginger, persimmon and radish into the main ingredient, pickled cabbage. Until the 1990s, kimchi was mainly produced at home, but began to be commercialized in the 1990s due to rapid economic growth, an increase in national income, and changes in the industrial structure and living environment. In recent years, domestic demand and exports have been steadily increasing along with the improvement of awareness of kimchi.
Kimchi is rich in various fermented products and ingredients such as dietary components, vitamins, carotene, lactic acid, lactic acid bacteria, acetylcholine, dextran and acetate, It is known as a functional food having a function of preventing the generation of carcinogens and mutagenic substances. Particularly, the abundant lactic acid bacteria have a probiotic effect of inhibiting the growth of harmful microorganisms in the intestines and improving intestinal microflora.
Kimchi has major strains such as Lactobacillus genera, Leuconostoc genus, Pediococcus genus, and Weissella genus, and these strains play an important role in kimchi fermentation . In particular, strains such as Leuconostoc citreum and Weissella koreensis are known to be dominant species for determining the taste of Korean kimchi.
Of the strains contained in kimchi, the strain of Weissella is known to produce L-ornithine (hereinafter referred to as L-ornithine). Ornithine is widely used in the United States as a food material that secretes growth hormone, increases muscle synthesis, promotes basic metabolism and prevents obesity. Recently, it has been found that it has new functionalities such as a skin cosmetic effect such as improvement of wrinkles.
내지 50 mg/100g F.W. In kimchi, ornithine is produced by a strain of Weissella, but the content of ornithine produced in the case of conventional kimchi is 30 mg / 100 g F.W. To 50 mg / 100 g F.W.
Accordingly, various techniques for increasing the content of ornithine in kimchi have been disclosed. In particular, a technique has been disclosed in which the content of ornithine can be increased by fermenting kimchi using a strain of genus Weissella, which is an ornithine-producing strain, than that of normal kimchi. In this regard, Korean Patent Registration No. 10-96258 discloses a technique for increasing the content of ornithine in kimchi by adding a strain of Weissella, a strain having ornithine-producing ability, in the process of manufacturing kimchi.
However, the technologies known in the above-mentioned patents require that strains of Weissella, which are artificially ornithine-producing strains, should be added in the process of manufacturing kimchi, so that it is difficult to purchase strain materials, and in order to produce a large amount of kimchi, Therefore, it is disadvantageous in that the production of kimchi is troublesome. Accordingly, the present inventors have completed the present invention after studying a method of containing ornithine at a high concentration in kimchi without artificially adding ornithine-producing strain.
Hereinafter, the present invention will be described in detail.
The ornithine-containing kimchi according to the present invention is prepared by adding a seasoning to a main ingredient to prepare a kimchi, and aging the kimchi prepared in the step. According to the present invention, the kimchi prepared in the step of aging the kimchi is cooked at 14 to 16 ° C for 42 to 50 hours and then stored.
In order to produce the ornithine-containing kimchi according to the present invention, the present invention first involves adding a seasoning to the main ingredient to prepare a kimchi.
In the present invention, the term " kimchi " refers generally to those generally known in the art which are prepared by adding seasonings to the main ingredients of ordinary kimchi.
The main ingredients of the kimchi may vary depending on the kind of kimchi to be produced. For example, the main ingredient of kimchi may be Chinese cabbage, radish, mustard, or heat. In addition, the above-mentioned sauce may include materials such as wave, garlic, ginger, liquid milk, sugar, salt, red pepper powder and the like, and these materials may also vary depending on the kind of kimchi to be manufactured and main ingredients. The kimchi prepared by adding seasonings to the main ingredients may be, for example, Chinese cabbage kimchi, mustard kimchi, non-kimchi, kakdugi, etc. However, the present invention does not necessarily limit the kinds of kimchi. That is, the step of producing the kimchi according to the present invention may be one which is produced by a method generally known in the art.
According to the present invention, a kimchi prepared by a method generally known in the art is prepared by aging. According to the present invention, the kimchi prepared in the step of aging the kimchi is cooked at 14 to 16 ° C for 42 to 50 hours and then stored.
That is, when the prepared kimchi is cooked at 14 to 16 ° C for 42 to 50 hours and then stored, the kimchi is preserved without adding the strain of Weissella, which is an ornithine high productivity strain, It is possible to increase the number of strains belonging to the genus Weissella, which is a tin-high productivity strain, so that a kimchi containing ornithine can be produced at a high concentration. Preferably, in the step of aging the kimchi, it is preferred that the prepared kimchi is cooked at 14 to 16 ° C for 44 to 48 hours and then stored. More preferably, the kimchi is cooked at 15 ° C for 48 hours and then stored.
At this time, when the temperature is less than 14 or exceeds 16 캜, the content of ornithine decreases. In addition, when the temperature is less than 42 hours, the content of ornithine is not sufficiently increased. If the cooking time is more than 50 hours, the ornithine content is not increased any more. Therefore, it is preferable that the prepared kimchi is stored at 14 ~ 16 ° C for 44 ~ 50 hours before the kimchi is aged.
When the kimchi is cooked under the specific conditions as in the present invention at the stage of ripening the kimchi, the content of ornithine in the kimchi is higher than or equal to 120 mg / 100 g FW on a 40-day storage standard, When stored next, the content of ornithine in kimchi is high as 169 mg / 100g FW or higher on a 40-day storage standard.
내지 110 mg/100g F.W.) The content of ornitine in Kimchi is higher than that of Kimchi (80 mg / 100 g FW to 110 g / 100 g FW) when the Weissella sp. Strain, which is a strain having ornithine troubling performance disclosed in Korean Patent No. 10-96258, mg / 100 g FW). Therefore, in the case of applying the method according to the present invention, it is possible to manufacture kimchi containing ornithine at a high concentration without addition of a strain of genus Weissella, which is ornithine-high productivity strain, separately in the step of producing kimchi.
The kimchi prepared in the step of preparing kimchi under the above-mentioned conditions is cooked and then stored. The storage may be carried out under the temperature conditions generally used in the art. Preferably 5 DEG C or less. More preferably, the storage is carried out at -2 to 5 ° C.
When storage is carried out under temperature conditions, the content of ornithine increases rapidly after 20 days, and reaches the highest value in the vicinity of 40 days.
In order to effectively increase the number of Weissella spp. Strains, ornithine-high productivity strains in the process of storing kimchi, it is preferable that the seasonings contain arginine. When the seasoning contains arginine, the ornithine high productivity strain increases the population by using arginine for metabolism, thereby rapidly increasing the content of ornithine.
It is preferable that the arginine is contained in an amount of 0.001 to 2% by weight based on the total weight of the kimchi prepared in the step of preparing the kimchi.
The addition of arginine may comprise adding an arginine containing food. For example, the arginine-containing food may be a spice extract. The above-mentioned retort extract may be manufactured by a known technique. For example, the extract may be prepared by adding 20 to 1000 parts by weight of water to 100 parts by weight of the paste, boiling the mixture for 1 to 3 hours, and heating.
The ornithine-rich kimchi according to the present invention produced by the above-mentioned method contains ornithine in the kimchi in an amount of 120 mg / 100 g FW or more on a 40-day storage basis without adding ornithine high productivity strain, If you store it at 15 ℃ for 48 hours and then store it, you can obtain a product with high content of ornithine in kimchi of 169 mg / 100g FW or more with 40 days storage standard.

1. Kimchi lactic acid bacteria analysis
1) Measurement of change in the number of lactic acid bacteria
DE MAN, ROGOSA and SHARPE(MRS, Difco, USA)에 0.002% bromocresol purple (BCP, Sigma, USA)를 첨가하여 사용하였다. BCP가 첨가된 MRS agar 고체 배지에 희석된 시료 100 ㎕를 도말한 후 25℃ 배양기에 48시간 배양하여 생성된 단일 노란색 colony를 계수하여 시료 ㎖당 colony forming unit(CFU)로 나타내었다(Park & Oh 2006; Yu 등 2009; Yu 등 2011).Each of the cabbage kimchi samples was diluted to a range of 10 -4 to 10 -6 by the number of peptones and 0.002% bromocresol purple (BCP, Sigma, USA) was added to DE MAN, ROGOSA and SHARPE (MRS, Difco, USA) Were added. 100 μl of the diluted sample was plated on the MRS agar solid medium supplemented with BCP and cultured in a 25 ° C. incubator for 48 hours. The resulting single yellow colony was counted and expressed as colony forming unit (CFU) per sample ml 2006; Yu et al. 2009; Yu et al. 2011).
2) Measurement of change in lactic acid bacteria
추출된 gDNA 농도를 측정한 뒤 Primer(27F:AGAGTTTGATCMTGGCTCAG, 1392R: ACGGGCGGTGTGTRC), Dr. Taq DNA Polymerase(Doctor Potein, Korea) 등을 이용하여 PCR 실시하였으며, 얻어진 PCR 산물을 (주)마크로젠에 의뢰하여 16S rDNA sequence 결과를 얻었으며, 이를 토대로 유산균 속 분포를 분석하였다(Kim & Chun 2005; Kim 등 2012).Changes in the lactic acid bacteria of cabbage kimchi were obtained by randomly sampling 10 colonies in each sample group on the plate used for the measurement of the number of lactic acid bacteria, and inoculating them into the MRS liquid medium, followed by suspension culture in a 25 ° C. incubator. 1 ml of the suspension-cultured sample was collected, and the cells were collected by centrifugation and then extracted with a genomic DNA (gDNA) extraction kit (Bioneer, Korea). After measuring the extracted gDNA concentration, Primer (27F: AGAGTTTGATCMTGGCTCAG, 1392R: ACGGGCGGTGTGTRC), Dr. PCR was carried out using Taq DNA Polymerase (Doctor Potein, Korea). The obtained PCR product was subjected to 16S rDNA sequence analysis by Macrogen Co., and the distribution of lactic acid bacteria was analyzed based on this result (Kim & Chun 2005; Kim et al.
As shown in Table 2 and FIG. 1 and FIG. 2, the lactic acid bacterium content was very high in the S4 and S5 stored after being cooked within the preferred range of the present invention. Especially, The stored S5 has a very high lactic acid bacterium content. In addition, as shown in FIG. 3, the content of Weissella lactic acid bacteria producing ornithine is more than 40% in the case of S5 stored after being cooked at 15 ° C for 48 hours.
2. Analysis of ornithine content
오르니틴 함량은 HPLC(ACCQ.TagTM Amino Acid Analysis System, Waters, USA)를 이용하여 측정하였다. 오르니틴의 형광 유도체화를 위해 AccQ.Fluor Reagent를 사용하였으며, 이들 유도체의 분리를 위해 3.9×150 ㎜ column(For Hydrolysate Amino Acid Analysis, Waters, USA)을 사용하였다. 오르니틴 함량은 표준 L-Ornithine(MERCK, Germany)의 분석결과와 비교하여 산출하였다(Park and Oh 2006; Yu & Oh 2010).The ornithine content was measured by HPLC (ACCQ.TagTM Amino Acid Analysis System, Waters, USA). AccuFluor Reagent was used for fluorescent derivatization of ornithine, and 3.9 × 150 mm column (For Hydrolysate Amino Acid Analysis, Waters, USA) was used for separation of these derivatives. In order to elute the derivative from the column, AccQ.Tag Eluent A (Waters, USA) and 60% acetonitrile were flowed at a flow rate of 1 ml per minute. The ornithine content was calculated by comparison with the standard L-Ornithine (MERCK, Germany) analysis (Park and Oh 2006; Yu & Oh 2010).
As shown in Table 3 and FIG. 3, it can be seen that the content of ornithine is very high on the 40-day storage standard in the case of S4 and S5 which are stored after being cooked within the preferred range of the present invention. In particular, when stored at 15 ° C for 48 hours, the ornithine content was 169.9 mg / 100 g FW, which is very high considering that the recommended intake of ornithine is 250 mg.
