도 2는 본 발명에 따른 항체 4D9와 F10과의 ELISA를 통한 항원 결합 친화력을 비교한 도표를 보여준다. 여기에서 본 발명에 따른 항체 4D9는 F10 보다 더 높은 친화력으로 H1, H2, H5 그리고 H9에 결합한다는 것을 보여준다(nAb 농도. ~ 100ng/ml). 본 실험을 위해 ELISA plate에 HA를 100 ng/ml의 농도로 코팅하였다(4 , overnight). 다음날 0.05% TBST로 3회 세척한 후 PBS에 희석한 20 % BSA를 200 ul/well로 처리하여 상온에서 1시간 반응하였다. TBST로 3회 세척한 후 100 ng/ml농도의 정제한 4D9과 F10을 100 ul/well 처리하고 상온에서 1시간 반응하였다. 다시 TBST로 3회 세척한 후 detection antibody인 goat anti-human IgG Fc-HRP를 TBST에 1:1000으로 희석하여 100 ul/well로 처리하고 상온에서 1시간 반응하였다. TBST로 3회 세척한 후 TMB로 develop하고 1M H2SO4 50 ul로 반응을 중지시켰다. ELISA reader를 이용하여 450 nm에서 흡광도를 측정하였다. F10과 비교하여 4D9이 HA1, HA2 그리고 HA5 에서 더 높은 binding affinity를 보였다. 본 실험을 위해 4D9이 cell에 bound되어 있는 HA에는 어떻게 binding하는지 알아보기 위해 pJK2b-HA를 293T cell에 transfection 하였다. 먼저 293T cell을 6 well plate에 접종하고 2일간 10% FBS가 포함된 DMEM을 이용하여 37℃, 5% CO2 incubator에서 배양하였다. cell이 plate의 80%이상 차면 pJK2b-HA 4과 Lipofectamin 2000(invitrogen사, cat#11668) 10를 각각 Opti-MEM I (invitrogen 사, cat#31985) 250에 희석하고 거품이 생기지 않도록 발현벡터와 Lipofectamine 2000을 섞어 혼합물을 상온에서 15분간 정치하였다. 5시간동안 배양한 후 DMEM + 10% FBS 배지로 교체하여 3일간 배양하였다. 배양 후 cell을 0.25% trypsin (invitrogen사, Cat#15050,)을 이용하여 떼어내고 세포수를 측정하여 5 x 106 cells 가 되도록 PBS에 현탁하였다. 200ul의 cell 현탁액을 각각의 tube로 옮기고 4D9과 F10을 10 ug/ml 농도로 처리하였다. rotator를 이용하여 섞어주면서 상온에서 1시간 반응하였다. 3000 rpm에서 5분간 원심 분리한 후 PBS에 1:500으로 희석한 fluorochrome-labled goat anti-human IgM 용액에 현탁하였다. 음성대조군으로 Remicade도 함께 실험하였다. rotator를 이용하여 섞어주면서 상온에서 1시간 반응한 후 3000 rpm에서 5분간 원심 분리하여 PBS에 현탁하고 GuavaPC96을 이용하여 분석하였다. 도 4은 항체 F10과 본 발명에 따른 항체 4D9 사이 경쟁적 ELISA를 보여준다. 본 실험에서는 4D9와 F10이 서로 같은 epitope에 binding하는지 확인하기 위하여 compitition ELISA를 수행하였다. 우선 pKB-FabD-4D9, pKB-FabD-F10 plasmid를 가진 ER2738에 M13 helper phage를 감염하여 4D9과 F10을 display 하는 phage를 만들었다. ELISA plate에 HA1과 HA5를 200 ng/well로 코팅하여 4 , overnight 반응하였다. 다음 날 코팅용액을 버린 후 PBS에 희석한 4% skim milk로 37℃ 에서 2시간동안 blocking하였다. TBST로 3회 세척한 후 미리 준비한 phage 용액에 정제한 4D9과 F10을 5 ug/ml 농도로 혼합하였다. 용액을 100 ul/well로 넣은 후 37℃ 에서 1시간 반응하였다. 다시 TBST로 3회 세척한 후 anti-M13-HRP를 TBST에 1:1000으로 희석하여 100 ul/well로 넣은 후 37℃ 에서 1시간 반응하였다. TBST로 3회 세척한 후 TMB로 develop하고 1M H2SO4 50 ul로 반응을 중지시켰다. LEISA reader를 이용하여 450 nm에서 흡광도를 측정하였다. 이 결과는 두 개의 항체 4D9과 F10이 동일한 항원 결정 부위를 공유한다는 것을 나타낸다. 도 5은 본 발명에 따른 항체 4D9가 H5-매개 세포 융합을 억제한다는 것을 보여준다. 여기서는 4D9이 HA에 의한 cell fusion을 얼마나 inhibition 하는지 알아보기 위해 cell fusion inhibition assay를 수행하였다. 먼저 HeLa cell을 6 well plate에 well 당 3x105으로 접종하고 2일간 10% FBS가 포함된 DMEM을 이용하여 37℃ , 5% CO2 incubator에서 배양하였다. cell이 plate의 95%이상 차면 cell을 PBS를 이용하여 2회 wash하고 pJK2b-HA5와 Lipofectamin 2000(invitrogen 사, cat#11668) 10를 각각 Opti-MEM I (invitrogen 사, cat#31985) 250에 희석하고 거품이 생기지 않도록 발현벡터와 Lipofectamine 2000을 섞어 혼합물을 상온에서 15분간 정치하였다. 5시간동안 배양한 후 DMEM + 10% FBS 배지로 교체하여 2일간 배양하였다. 배양 후 4D9이 함유된 serum-free DMEM 배지로 교체하여 1시간 반응하였다. PBS로 세척한 후 0.1M citric acid가 함유된 pH4.8의 산성배지를 15분간 처리하였고 다시 PBS로 세척한 후 DMEM+10%FBS 배지로 교체하여 3시간 배양하였다. 배양 후 현미경으로 fusion 양상을 관찰하였다. HeLa cell에 HA5 transfection 후 산성배지를 처리하니 plate 전체에서 cell fusion이 관찰되었으며 4D9을 처리한 경우에서는 50-60%의 inhibition을 보였다.
